Reactive oxygen species (ROS) production and scavenging in plants under drought stress have been studied intensively in recent years. Here we report a global analysis of gene expression for the major ROS generating and scavenging proteins in alfalfa root and shoot under gradual drought stress followed by one-day recovery. Data from two alfalfa varieties, one drought tolerant and one drought sensitive, were compared and no qualitative differences in ROS gene regulation between the two were found. Conserved, tissue-specific patterns of gene expression in response to drought were observed for several ROS-scavenging gene families, including ascorbate peroxidase, monodehydroascorbate reductase, and peroxiredoxin. In addition, differential gene expression within families was observed. Genes for the ROS-generating enzyme, NADPH oxidase were generally induced under drought, while those for glycolate oxidase were repressed. Among the ROS-scavenging protein genes, Ferritin, Cu/Zn superoxide dismutase (SOD), and the majority of the glutathione peroxidase family members were induced under drought in both roots and shoots of both alfalfa varieties. In contrast, Fe-SOD, CC-type glutaredoxins, and thoiredoxins were downregulated.  相似文献

Aims: To investigate the growth of salmonellae on sprouting alfalfa seeds as affected by the inoculum size, microbial load and Pseudomonas fluorescens 2–79. Methods and Results: Alfalfa seeds pre‐inoculated with ≤101–103 CFU g?1 of salmonellae and with or without Ps. fluorescens 2–79 were sprouted in glass jars and the population of salmonellae were determined daily for up to 6 days. The population of salmonellae on germinating seeds reached the maximum 2–3 days after sprouting when total bacterial count reached the maximum (109 CFU g?1). The population of salmonellae on sprouting seeds not treated with Ps. fluorescens 2–79 showed a net increase of 3–4 log units. However, the population of salmonellae on alfalfa seeds treated with Ps. fluorescens 2–79 showed a net increase of only 1–2 log units. Disinfection of seeds with calcium hypochlorite enhanced the growth of salmonellae. Conclusions: Treatment of seeds with Ps. fluorescens 2–79 reduced the growth of salmonellae by 2–3 log units. Significance and Impact of the Study: The potential of Ps. fluorescens 2–79 as a biological agent for use in control of salmonellae on sprouting seeds was demonstrated and warrants further investigation.  相似文献

不同播期对紫花苜蓿生长性状及越冬性的影响研究   总被引:11,自引:1,他引:10  
采用随机试验设计方法,在宁南旱作农业区进行了不同播期对紫花苜蓿出苗率、生长发育、越冬率和产草量影响的研究。结果表明,在宁南地区人工种植紫花苜蓿的出苗率与播期温度存在极显著正相关关系,而不同播期土壤耕层含水量变化率较小,且基本能满足苜蓿萌发与生长的需要;播期越早,根颈越粗,入土越深,但4月份前播种,出苗率低,群体小且易形成“小老苗”;7月份以后播种。由于生长期短,植株根颈细嫩,越冬率很低或完全冻死。综合不同播期出苗率、产草量和越冬率等因素,认为在宁南地区紫花苜蓿的适宜播种期应在4月30日~6月30日。  相似文献

通过对不同地理种群和致害性的褐飞虱体内共生菌,在高温和抗性水稻品种作用下的种群动态研究表明,在正常湿度下褐飞虱体内共生菌的数量随若虫的生长发育而增加,5龄若虫期是共生菌增长的关键期;同一种群的褐飞虱秋季共生菌数量明显高于夏季。广西田间种群的短翅雌虫体内的共生菌数量显高于浙江省的杭州和龙游种群,经高温处理后,3个生物开共生菌数量均显减少,但其子代共生菌的数量在正常温度下马上恢复,与未经高温处理的褐飞虱共生菌数量基本相同,取食抗性品种的生物型1和田间种群的褐飞虱体内的共生菌数量均明显减少,取食抗性品种的第2代的共生菌又均比第1代少。这说明高温对共生菌的影响是暂的,而抗性品种的对共生菌的作用与褐飞虱种群对品种的适应性有关。  相似文献

研究了Hg2 + 对苜蓿叶片的毒害效应。结果表明 :随着Hg2 + 浓度的增加和处理时间的延长 ,叶绿素含量下降 ,电导度上升 ;低Hg2 + 浓度及短时间处理 ,超氧阴离子自由基 (O·2 )和丙二醛 (MDA)含量增加 ,SOD、POD和CAT等保护酶活性升高 ,表明膜系统受到了伤害 ;高Hg2 +浓度及长时间处理 ,O·2 和MDA含量下降 ,SOD、POD和CAT等保护酶活性降低 ,表明细胞结构和功能受到了不可逆的伤害。  相似文献

Genomic DNA extracted from leaves of sixteen alfalfa genotypes, six-week-old callus cultures derived from them and from a suspension culture was used in Southern blots and slot blots probed with alfalfa E180 satellite DNA. The restriction patterns revealed by the E180 probe did not differ among alfalfa genotypes and no differences between the leaf and callus restriction patterns were seen. However, the number of copies of the E180 satellite was lower in the callus samples than in the corresponding leaf samples and significant differences in copy number among callus samples were detected. Genomic stress induced by tissue culture may have caused the reduction in copy number. This phenomenon may be important in the generation of somaclonal variation in alfalfa.  相似文献

Genetic distances (GDs) based on molecular markers are important parameters for identifying essentially derived varieties (EDVs). In this context information about the variability of molecular markers within maize inbred lines is essential. Our objectives were to (1) determine the variation in the size of simple sequence repeat (SSR) fragments among different accessions of maize inbreds and doubled haploid (DH) lines, (2) attribute the observed variation to genetic and marker system-specific sources, and (3) investigate the effect of SSR fragment size differences within maize lines on the GD between maize lines and their consequences for the identification of essentially derived varieties. Two to five accessions from nine inbred lines and five DH lines were taken from different sources or drawn as independent samples from the same seed lot. Each accession was genotyped with 100 SSR markers that evenly covered the whole maize genome. In total, 437 SSR fragments were identified, with a mean of 4.4 alleles per locus. The average polymorphic information content (PIC) was 0.58. GD estimates between two accessions of the same genotype ranged from 0.00 to 0.12 with an average of 0.029 for inbred lines and 0.001 for DH lines. An average of 11.1 SSRs was polymorphic between accessions of the same inbred line due to non-amplification (8.1 SSRs), heterogeneity (4.0 SSRs) or unknown alleles (2.6 SSRs). In contrast to lab errors, heterogeneity contributed considerably to the observed variation for GD. In order to decrease the probability to be suited for infringing an EDV threshold by chance, we recommend to increase the level of homogeneity of inbred lines before applying for plant variety protection.  相似文献

试验结果表明小麦品种 (系 )的抗性对麦蚜种群数量影响很大 ,百株蚜量随着小麦品种抗性增强而下降。而同一小麦品种对不同种蚜虫的抗性存在质的差异 ,铭贤 1 69品种 ,蚜高峰期百株蚜量麦长管蚜 63 0头 ,禾谷缢管蚜只有 1 1 5头 ,两者相差 5 5倍。另一方面 ,小麦品种抗性对麦田天敌的种群数量影响不大 ,而对天敌的发生期有些影响。因此 ,小麦品种抗性、天敌对麦蚜的自然控制能力 ,可把小麦中后期的蚜虫虫口密度控制在经济损失允许水平之下。  相似文献

The greenbugs, Schizaphis graminum (Rondani) were collected from the barley fields in Isfahan region of Iran. The aphid colonies were maintained on each of six barley cultivars including Karoon, Kavir, Zarjoo, Nosrat, Afzal and Rihane. All the experiments were done on the mentioned barley varieties at 26 ± 1°C, 60 ± 5% relative humidity (RH) and at a photoperiod of 16:8 (L:D) h. The shortest and longest developmental times were obtained on Nosrat 6.35 ± 0.11 and Rihane 6.75 ± 0.07 days, respectively. The survivorship of immature stages varied from 71.95% on Nosrat to 82.14% on Zarjoo. The total number of offsprings were 71.05 and 63.22 nymphs per female on Kavir and Karoon. The highest and lowest r m values were observed on Kavir (0.336 ± 0.005) and Rihane (0.299 ± 0.008), respectively. The statistical analysis of jackknife did not show a significant influence of the tested barley varieties for the mean generation time and a similar procedure of difference for λ and r m was estimated.  相似文献
