Detached ears of three winter wheat ( Triticum aestivum L.) varieties were cultured in solution for 12 days with sucrose levels varying from 36.5 to 292 m M. The dry weight and starch content of grains increased asymptotically with the sucrose level in the solution. At 4 days of culture, glucose phosphate isomerase (EC 5.3.1.9) activity grain−1 was lower with 36.5 m M than with higher sucrose levels in the medium; at 8 days, adenosinc diphosphoglucose pyrophosphorylase (EC 2.7.7.27) and (soluble plus bound) starch synthase (EC 2.4.1.21) activities grain−1 were higher with 146 and 292 m M sucrose than with 36.5 and 73 m M sucrose. The multiple regression of starch content over these enzyme activities showed that starch synthase was relatively more important as an independent variable. The dry weight and starch content of grains were higher in the variety Maris Huntsman than in Splendeur and Hobbit. The water content of grains was lower in Splendeur than in the other two varieties. At 4 days the glucose phosphate isomerase, adenosine diphosphoglucose pyrophosphorylase and starch synthase activities grain−1 were smaller in Splendeur than in Hobbit and Maris Huntsman and al 8 days they were higher in Maris Huntsman than in Hobbit and Splendeur. The varietal differences in starch content of grains were related to the activities of glucose phosphate isomerase and especially of starch synthase.  相似文献

This study was conducted with a view to understand the effect of plant characteristics on the incidence of pests on most popular jute varieties, viz. JRO-524, JRO-632, JRO-878, JRO-7835 of olitorius jute and JRC-212, JRC-321, JRC-4444, JRC-7447 of capsularis jute. Correlation studies of plant characteristics with pest incidence in jute (Corchorus spp.) revealed that the basal girth of plant exhibited a positive significant relationship with the incidence of Apion corchori Marshall but not with other pests such as jute semilooper (Anomis sabulifera Guen.), Bihar hairy caterpillar (Spilarctia obliqua Wlk.), grey weevil (Myllocerus discolor Bohemus) and yellow mite (Polyphagotarsonemus latus Banks). However, plant height showed positive correlation with the incidence of all the major pests of jute. Leaf characteristics (number of leaves/plant, leaf area, leaf thickness, moisture and chlorophyll content of leaves of all the varieties recorded at three different growth stages, i.e. at 50, 80 and 110 days after sowing), number of leaves/plant had positive and significant relationship with the incidence of all the pests except stem weevil. However, leaf area and moisture content of leaves showed significant positive relationship with the incidence of yellow mite. Leaf moisture also showed a positive role on the infestation of grey weevil. Leaf thickness and chlorophyll content of leaves and stem, fibre thickness and moisture content of stem did show any significant effect on pest's incidence. Upon comparing the plant characteristics between the two jute species, olitorius varieties had greater fibre thickness than that of capsularis varieties but fibre thickness had no significant impact on incidence of pests.  相似文献

Summary Commercialization and marketing of agricultural products derived from recombinant DNA technologies is now becoming a reality. Genetically engineered tomatoes are poised to appear in supermarket shelves in the United States sometime next year, with other crops, including cotton, maize, soybean, rapeseed, potatoes, cucurbids, raspberries and others, scheduled for release 1 to 2 yr later (Hamilton and Ellis, 1992). One of the major breakthroughs responsible for the rapid creation of commercializable products has been the development of gene transfer methods capable of introducing foreign DNA into elite germplasm. This review examines some of the principles and applications of such gene transfer technologies in relation to conventional alternatives that are limited by cell culture, host, and genotype. Advantages and disadvantages of various gene transfer methods will be discussed. Special emphasis will be given to particle bombardment, as methods based on this technology paved the way for the engineering of a number of important agronomic species. The major viruses infecting apple cultivars throughout the world including India are apple mosaic virus (ApMV), apple stem pitting virus (ASPV), apple stem grooving virus (ASGV), apple chlorotic leaf spot virus (ACLSV), and recently, a new virus, apple necrotic mosaic virus (ApNMV), was reported from mosaic-infected apple cultivars in India. The aim of this study was to detect the ApNMV virus along with the other three viruses (ApMV, ASPV and ASGV) simultaneously by multiplex RT-PCR. Four primer-pair-produced amplicons of 670, 550, 350 and 210 bp corresponding to ApNMV, ApMV, ASPV and ASGV, respectively, were found to be specific for these viruses when tested individually. The annealing temperature (55°C), primer concentration (0·8 µl) and other components of the master mix were standardized for the development of one-step m-RT-PCR assay. The m-RT-PCR protocol developed was further validated with 30 samples from seven symptomatic or asymptomatic apple cultivars, which revealed the presence of more than one virus in these cultivars. Most of the viruses were found to be present either alone or in mixed infection; however, ASPV was more common in tested cultivars. An easy, cost-effective and rapid multiplex RT-RCR protocol was developed to detect the four viruses, which infect apple plants either in individually or together in the field. This assay will help in the surveying and indexing of apple germplasm and the distribution of all four viruses in the apple growing regions of India.  相似文献

Ditylenchus dipsaci-infected and noninfected alfalfa plants in a naturally infested field were studied from July 1980 to September 1982. Forty-one percent of the plants died during the study. Ninety-seven percent of the plants that died were infected with D. dipsaci. Sixty-nine percent of the observed mortality occurred during winter. Forage yield of infected plants was significantly lower than yield of noninfected plants at each harvest. Stored carbohydrates in infected plants were significantly lower than in noninfected plants. In a controlled environment test, significantly greater mortality occurred in frozen severely infected plants than in frozen noninfected plants, while no mortality occurred in severely infected or noninfected plants that were not frozen. Both forage yield and stored carbohydrates were significantly lower in severely infected than noninfected, non-frozen plants. Mortality in greenhouse-grown plants that were transplanted to field plots was significantly greater in D. dipsaci-infected plants than in noninfected plants after one winter.  相似文献

In July, 2011, alfalfa plants were observed in Yangling, Shaanxi Province, China with typical witches’ broom symptoms. The presence of phytoplasma was confirmed by transmission electron microscopy and a nested PCR, which amplified a 1.2‐kb fragment using universal primer pairs P1/P6 followed by R16F2n/R2. Sequence, phylogeny and RFLP analyses showed that the alfalfa witches’ broom disease was associated with a phytoplasma of group 16SrV, subgroup V‐B. This is the first record of the 16SrV phytoplasma group infecting alfalfa plants.  相似文献

Maize (Zea mays cv W64A × W182E) was grown in a low nutrient sandy loam. Inoculation with legume soils (4.1% v/v) gave a 3 to 4 fold increase in shoot growth relative to the control after 5 to 8 weeks growth in greenhouse conditions. Plants were routinely irrigated with 1/10 Hoagland solution (with 10mM KNO3). With half strength Hoagland solution (10mM KNO3) there was no clear growth response. This growth response was observed with a variety of legume soils but not with any of the maize soils tested. The response to alfalfa soil was eliminated or much reduced by gamma irradiation (3.6 Mrad) or autoclaving of the inoculum. The bactericide streptomycin had no effect on the growth response whereas the fungicides benomyl and PCNB eliminated it. This suggests that fungi and not bacteria are involved in the growth promotion.  相似文献
