AbstractImmobilization of enzymes from different sources on various supports in designed systems increases enzymes’ stability by protecting the active site of it from undesired effect of reaction environment. Also, immobilization decreases the cost of separation and facilities the reuse of the enzymes. Therefore, the design of new immobilization enzyme preparations has been an inevitable area of modern biotechnology. Herein, Rhizomucor miehei lipase (RML) was immobilized on montmorillonite K-10 (MMT-RML) by adsorption and in polyvinyl alcohol (PVA-RML) by entrapment to obtain a more stable and active lipase preparation. The free and immobilized lipase preparations were characterized for p-nitrophenyl palmitate hydrolysis. The apparent Michaelis–Menten (Kmapp) constant was almost the same for the free RML and PVA-RML, whereas the corresponding value was 17.7-fold lower for MMT-RML. PVA-RML and MMT-RML have shown a 1.1 and 23.8 folds higher catalytic efficiency, respectively, than that of the free RML. The half-lives of PVA-RML and MMT-RML were found to be 7.4 and 3.4 times longer than the free RML at 35?°C, respectively. PVA-RML and MMT-RML maintained 65% and 87% of their initial activities after four reuses. These results showed that the catalytic performance of RML has improved significantly by immobilization. 相似文献

Improved efficiency of Markov chain Monte Carlo facilitates all aspects of statistical analysis with Bayesian hierarchical models. Identifying strategies to improve MCMC performance is becoming increasingly crucial as the complexity of models, and the run times to fit them, increases. We evaluate different strategies for improving MCMC efficiency using the open‐source software NIMBLE (R package nimble) using common ecological models of species occurrence and abundance as examples. We ask how MCMC efficiency depends on model formulation, model size, data, and sampling strategy. For multiseason and/or multispecies occupancy models and for N‐mixture models, we compare the efficiency of sampling discrete latent states vs. integrating over them, including more vs. fewer hierarchical model components, and univariate vs. block‐sampling methods. We include the common MCMC tool JAGS in comparisons. For simple models, there is little practical difference between computational approaches. As model complexity increases, there are strong interactions between model formulation and sampling strategy on MCMC efficiency. There is no one‐size‐fits‐all best strategy, but rather problem‐specific best strategies related to model structure and type. In all but the simplest cases, NIMBLE's default or customized performance achieves much higher efficiency than JAGS. In the two most complex examples, NIMBLE was 10–12 times more efficient than JAGS. We find NIMBLE is a valuable tool for many ecologists utilizing Bayesian inference, particularly for complex models where JAGS is prohibitively slow. Our results highlight the need for more guidelines and customizable approaches to fit hierarchical models to ensure practitioners can make the most of occupancy and other hierarchical models. By implementing model‐generic MCMC procedures in open‐source software, including the NIMBLE extensions for integrating over latent states (implemented in the R package nimbleEcology), we have made progress toward this aim. 相似文献

Evaluating how decomposition rates and litter nutrient release of different litter types respond to changes in water conditions is crucial for understanding global carbon and nutrient cycling. However, it is unclear how decreasing water affects litter mixture interactions for the maize–poplar system in arid regions. Here, the responses of the litter decomposition process and litter mixture interactions in the agroforestry system to changes in water conditions (control, light drought, and moderate drought) were tested. Moderate drought significantly decreased the decomposition rate for poplar leaf and mixed litters, and decomposition rate was significantly reduced for maize straw litter in light and moderate drought stress. The mass loss rates of maize straw and mixed litters were significantly higher than that of the poplar leaf litter under drought conditions, but there was no significant difference among the three litter types in the control. There was no interaction between mass loss of the mixed litter in the control and light drought conditions, and the litter mixture interaction showed nonadditive synergistic interactions under moderate drought. In terms of nutrient release, there was also no interaction between litter mixture with nitrogen and carbon, but there was antagonistic interaction with potassium release under the light drought condition. Our results demonstrate that drought conditions can lead to decreasing decomposition rate and strong changes in the litter mixture interactions from additive effects to nonadditive synergistic effects in moderate drought. 目的:探讨莱菔子乙醇提取物(AERS)对ApoE-/-小鼠血脂血糖及肝脂肪变性的影响及其机制。方法:随机将60只ApoE-/-小鼠分为对照组(CG)、生理盐水组(NG)、罗格列酮组(RG)、AERS治疗组,后者再分成AERS低、中、高剂量组(AERS-L/M/H),每组10只。除CG常规饲料外,其他动物均以高脂高糖高盐饲料饲养9周,CG和NG分别每天以等容量NS灌胃1次,RG每天灌胃罗格列酮(1.33 mg·kg-1,NS稀释成等容量溶液,每次0.2 ml·10 g-1),AERS组动物每天分别予AERS灌胃(100、300和900 mg·kg-1)9周后。肝病理学观察,检测FPG、FIns,计算胰岛素抵抗指数(IRI)和肝系数(LC)。检测血清ALT、AST、瘦素(LEP)、TNF-α及血脂水平(TC、FFA等)。Western blot检测肝脂质代谢相关蛋白表达(HMG-R、LDL-R、LEP-R)。结果:与CG相比,NG和RG肝脏外观(色泽、肿胀情况等)和病理学(脂肪变性、肝细胞坏死等)改变明显,而AERS-M/H与CG相似;与CG相比,血清FPG、Fins、IRI、ALT、AST、TNF-α、LC、TG、LDL-C、FFA、LEP等指标明显升高(P<0.05);与NG相比,AERS呈现剂量依赖性降低(P<0.05);与RG相比,AERS-H的FPG、Fins明显升高(P<0.05),而IRI明显降低(P<0.05);与NG和RG相比,AERS组的HMG-R和LEP-R蛋白表达呈剂量依赖性减少,而LDL-R蛋白表达呈剂量依赖性增加(P<0.05)。结论:AERS能阻止高脂高糖饲料诱导ApoE-/-小鼠血脂血糖升高及肝脂肪变性,其机制与减少FFA和LEP的生成,抑制TNF-α、HMG-R、LEP-R蛋白表达和促进LDL-R的蛋白表达有关。 相似文献

The monoterpene perillyl alcohol (POH), an intermediate in the plant terpenoid biosynthetic pathway, has well-established tumor chemopreventive and chemotherapeutic potential. We have previously shown that the primary hydroxyl group of POH is essential for its antitumor and anti-angiogenic activities. In the current study we present the enzymatic synthesis of two POH derivatives with different polar and hydrophobic characteristics, namely perillyl glucoside and perillyl glucoside fatty ester, through a two-step modification. Initial glucosylation of POH on its active hydroxyl group with D-(+)-glucose and subsequent esterification of the perillyl glucoside product with vinyl laurate were carried out using almond β-glucosidase and lipase B from Candida antarctica, respectively, in a low-water system. Optimization of enzymatic reactions was performed to achieve the highest possible conversion yields. The antitumor cell proliferation activity against mouse Lewis lung carcinoma cells was retained in both derivatives, although the perillyl glucoside ester showed greater inhibition than perillyl glucoside. Our results underline the feasibility of enzymatically producing novel bioactive analogs of phytochemicals displaying useful physicochemical properties. 相似文献

A major difficulty in addressing chemical mixtures through legislation or regulations revolves around our limited understanding of their potential impacts. This review provides an overview of recent research on pesticide mixture toxicity to aquatic biota and the methods employed to predict toxic effects. The most common approaches are to assume concentration-addition or independent action of chemicals in a mixture. There are a number of cases in the literature of interactions between pesticides. However, models accounting for possible interactions between mixture components are used infrequently. Although results are limited, studies investigating the effects of pesticide mixtures have not demonstrated significant synergism at environmentally relevant concentrations. Based on the results of our review, we conclude that the concentration-addition model is a generally conservative and practical first-tier model for the ecological assessment of pesticide mixtures in aquatic systems. 相似文献

In mammalian skin, melanocyte proliferation and melanogenesis can be stimulated by keratinocytes, fibroblasts and other regulatory factors. To determine whether hydroxybenzyl alcohols (HBAs) show more inhibitory in melanocytes cultured alone or in melanocytes co-cultured with keratinocytes, we developed a murine melanocyte–keratinocyte co-culture model to investigate the pigmentation regulators in company with other melanogenic inhibitors and stimulators. It was found that the effects of HBAs and melanogenic factors were more evident in melanocytes co-cultured with keratinocytes. Keratinocytes may play a synergistic role in melanocyte melanogenesis and influence the pigment production. The tests in the co-culture model also imply that the inhibitory effects of HBAs on melanogenesis are due to the direct inhibition of melanosomal tyrosinase activity. HBAs showed a low cytotoxicity. The eventual results proved that HBAs are promising and safe agents for skin whitening in melanocyte alone and in co-culture systems. The co-culture model provides a more physiologically realistic condition to study the interaction between melanocytes and keratinocytes, which enables a reliable screening system for depigmenting compounds. 相似文献
