The biserial agglutinated foraminifer Textularia hauerii d’Orbigny frequently occurs at shallow depths in reefal habitats off Bazaruto Island, Mozambique. X-ray diffraction, scanning electron- and light microscopy analyses show the enhanced concentration of the titanium-bearing mineral ilmenite (FeTiO3) in the agglutinated tests of this species. The mineral constitutes an insignificant component in the surrounding sediment (< 1%), but accounts for more than 26% of the foraminiferal test content. The accumulation of ilmenite in T. hauerii and its absence in co-occurring agglutinated Foraminifera suggest the preferential selection for ilmenite. The tendency for ilmenite particles to be arranged along sutures and on the apertural face (1) provides further evidence that certain Foraminifera can specifically select grains, and (2) implies that there exists a selective mechanism to control the timing and placement during test and chamber construction. The preferential uptake represents the first record of ilmenite selection in Foraminifera. Potential driving forces controlling the selective agglutination of Ti minerals, the preferential adsorption to Ti surfaces, and interactions between cellular compounds of the foraminiferal reticulopods and the mineral surfaces are explored.  相似文献

AIM: To find out if testing of up to 10 Staphylococcus aureus isolates from each sample from raw milk and raw milk products for staphylococcal enterotoxin (SE) might increase the chances of identifying potential sources of food intoxication. METHODS AND RESULTS: Altogether 386 S. aureus isolates were tested for the presence of SE by reversed passive latex agglutination (SET-RPLA), and SE genes (se) by a multiplex polymerase chain reaction (PCR). In 18 of 34 (53%) S. aureus positive samples a mixture of SE and/or se positive and negative isolates were identified. Multiplex PCR increased the number of potential SE producing strains, i.e. isolates that harboured se, with 51% among the product and 48% among the raw bovine milk isolates. Examination by pulsed-field gel electrophoresis mostly confirmed clonal similarity among isolates sharing SE/se profile, but did not further differentiate between them. CONCLUSIONS: Isolates of S. aureus collected from one sample may show great diversity in SE production and different plating media seem to suppress or favour different strains of S. aureus. SIGNIFICANCE AND IMPACT OF THE STUDY: Several isolates of S. aureus from each sample should be tested for enterotoxin production in cases with typical SE intoxication symptoms with methods that are able to reveal new SE/se.  相似文献

An important consideration in the manufacture of products derived from animal or human sources is the virus reduction capacity of the manufacturing process as estimated using validated bench-scale models of relevant manufacturing steps. In these studies, manufacturing process intermediates are spiked with virus and processed using the bench-scale model and the resulting viral titres of input and output samples are typically determined using cell-based infectivity assays. In these assays, the Spearman-Kärber (SK) method is commonly used to estimate titres when there is one or more positive observation (i.e., the presence of any viral cytopathic effect). The SK method is most accurate when the proportion of positive observations ranges from <0.1 to >0.9 across dilutions but can be biased otherwise. Maximum likelihood (ML) based on a single-hit Poisson model is an alternative widely used estimation method. We compared SK with ML and found the methods to have similar properties except for situations in which the concentration of virus is low but measurable. In this case, the SK method produces upwardly biased estimates of titres. Based on our results, we recommend the use of either ML or SK at most virus concentrations; however, at low virus concentrations ML is preferred.  相似文献

A technique which enables cells to be observed in suspension for times of the order of minutes (employing acoustic radiation foreces in a 1 MHz ultrasonic standing wave field) is described. Video recordings of the mutual adhesion of human erythrocytes in suspension have been analysed. Concave-ended cell doublets and linear rouleaux developed in 0.5–1.5% w/v Dextran T500 by a gradual (2.5–17 s) increase in the area of cell contact over the cell cross-section. The concave-ended rouleaux form was not seen in polylysine or in polyethylene glycol. In 5–7% dextran and in 20g/ml polylysine mutual adhesion was a two stage process. Cells first form a strong local contact which persists (without apparently growing in area) for a number of seconds following which the cell surfaces move suddenly to form a spherical doublet. The average initial contact time and engulfment time for cells in 7% Dextran T500 are 18 and 2.7 s, respectively. The corresponding values for cells in 20 g/ml, 14 kDa, polylysine are 2.7 and 0.3 s. There was no initial contact delay during spherical doublet formation in mg/ml polylysine. Electron microscopy showed that the intercellular seam for spherical doublets formed with all three agglutinating molecules was bent in a wavy (4 m) profile. The thickness of the intercellular space varied in a spatially periodic way (0.8 m) for cells in polylysine. Examples of periodic intercellular spaces were seen by light microscopy in polyethylene glycol induced clumps. The role of interfacial instability in the adhesion processes is discussed.  相似文献

Serial thermotherapy with hot water involving different treatment series was followed in two sugarcane cultivars namely Co 740 and CoC 671 for assessing its effect on sugarcane mosaic virus (SCMV) titre in the regenerated plants. Observations on virus titre and sett regeneration were recorded at different intervals after planting the treated setts. The direct antigen coating (DAC) ELISA method was followed with the polyclonal antisera raised against the widespread local SCMV strain. All the thermo treatments affected the sett regeneration in both the cultivars. Although none of the treatment series produced virus free plants, the SCMV titre was comparatively reduced in the regenerated plants at an early phase (4 weeks after planting) in severe hot water treatment series but drastic reduction in sett germination was observed. However, the SCMV titre picked up later (8 weeks after planting) in all the treatment series. The early reduction in SCMV titre could be utilized for obtaining more virus free plants in meristem culture.  相似文献

A lectin (LEL) was isolated from the fresh fruiting bodies of the shiitake mushroom Lentinula edodes by a combination of gel filtration chromatography on Sephadex G-150 and affinity chromatography on an N-acetyl-Dgalactosamine-Sepharose 4B column. Its molecular mass, as determined by gel filtration, was estimated to be 71, 000 Daltons and its structure is homotetrameric with subunit molecular weight of approximately 18,000 Daltons. LEL agglutinated non-specifically red blood cells from the human ABO system as well as rabbit erythrocytes and in haemagglutination inhibition assays, exhibited sugar-binding specificity toward N-acetyl-D-galactosamine. EDTA had no inhibitory effect on its haemagglutinating activity, which was stable up to 70°C and was not affected by changes in pH. The lectin had no covalently-linked carbohydrate and amino acid composition analysis revealed that it contained 124 amino acid residues and was rich in tyrosine, proline, phenylalanine, arginine, glutamic acid and cysteine. LEL did not cause mortality neither was it observed to alter the morphology of key organs when administered intraperitoneally at concentrations up to 10,000 mg kg-1 body weight of mice.  相似文献

SYNOPSIS The galactosephilic and mannosephilic hemagglutinins of Pseudomonas aeruginosa adsorbed onto Euglena gracilis, Chlamydomonas reinhardi , and Tetrahymena pyriformis . Furthermore, peroxidase binding to the 3 protozoan species was shown to be mediated by these lectins. Binding of Pseudomonas lectins to E. gracilis and C. reinhardi caused their specific agglutination, whereas no agglutination was observed with T. pyriformis , even after treatment by papain or by NaF. Added to the culture medium, the Pseudomonas hemagglutinins stimulated growth of E. gracilis and T. pyriformis due to their binding to these protozoa: this effect was partly inhibited by the specific sugar.  相似文献

A lectin was isolated and characterised from Salvia bogotensis seeds. Removal of the abundant pigments and polysaccharides, which are present in seeds, was an essential step in its purification. Several procedures were assayed and the best suited, including Pectinex treatment, DEAE-cellulose and affinity chromatography, led to a protein being obtained amounting to 18-20mg/100g seeds having high specific agglutination activity (SAA). The lectin specifically agglutinated human Tn erythrocytes and was inhibited by 37mM GalNAc, 0.019mM ovine submaxillary mucin (OSM) or 0.008mM asialo bovine submaxillary mucin (aBSM). Enzyme-linked lectinosorbent assay (ELLSA) revealed strong binding to aOSM and aBSM, corroborating Tn specificity, whereas no binding to fetuin or asialo fetuin was observed. The lectin's monomer MW (38,702Da), amino acid composition, pI, carbohydrate content, deglycosylated form MW, thermal stability and Ca(2+) and Mn(2+) requirements were determined. Evidence of the existence of two glycoforms was obtained. The lectin's specificity and high affinity for the Tn antigen, commonly found in tumour cells, makes this protein a useful tool for immunohistochemical and cellular studies.  相似文献

Many isolates of cocoa swollen shoot virus (CSSV) have been found in Ghana. Relationships between these isolates have been based on symptom expression and limited serological information. This paper reports on the serological relationships between 44 accessions of CSSV using the virobacterial agglutination test. The CSSV group is differentiated into eight groups using seven antibody 'types'. The largest group comprising those isolates closely related to CSSV 1A is sub-divided into four further groups. These groupings are compared with previous results. Differences are seen between accessions of the same isolate which may be due to contamination of the source plants. These serological results can be used for studying mixed isolate infections as well as for determining the most closely related CSSV mild isolate for appropriate cross-protection against severe isolates.  相似文献
