抗黄曲霉毒素B1单链抗体的筛选和鉴定 总被引:1,自引:0,他引:1
目的】从Tomlinson(I)噬菌体抗体库中筛选人源化抗黄曲霉毒素B1单链抗体蛋白(scFv)并进行鉴定。【方法】分别采用甘氨酸洗脱、胰蛋白酶洗脱、游离AFB1竞争洗脱和AFB1竞争洗脱加胰蛋白酶处理4种方法对噬菌体抗体进行特异性洗脱。将筛选到的噬菌体阳性克隆转化到大肠杆菌 (Escherichia.coli ) HB2151,IPTG诱导表达scFv。ELISA检测和基因序列测定。【结果】比较四种洗脱方法,发现用AFB1竞争加胰蛋白酶洗脱筛选到阳性克隆的的概率最高,把此方法得到的阳性噬菌体克隆转化大肠杆菌HB2151表达,竞争性ELISA检测得到2个能特异性结合游离的AFB1阳性克隆。间接性ELISA测定相对亲和力分别为0.4 μg/mL和0.7 μg/mL 。测序证实scFv属于人类免疫球蛋白可变区。【结论】利用噬菌体展示技术获得高特异性抗黄曲霉毒素B1的人源化单链抗体,本实验方法可以为其它抗半抗原重组抗体的筛选提供一定的借鉴意义。 相似文献

Cottonseeds having fluorescent fibers were harvested from fields in Arizona and examined utilizing light microscopy and transmission electron microscopy. The occurrence of fluorescent fibers indicated that seeds had been infected by Aspergillus flavus during development. Presence of A. flavus was verified by plating portions of seeds with fluorescent fibers. Hyphae, conidial heads, and conidia were identified readily in differentially-stained cotyledon tissue processed for light microscopy. Utilization of transmission electron microscopy permitted observations on lignified seed coats and cotyledons of mature cottonseeds. Hyphae were located throughout the cotyledon and in the nonlignified layers of the seed coat. The identification of hyphae in cross sections of vessel elements within the seed coat provided ultrastructural evidence supporting the hypothesis that A. flavus may enter seeds via the vascular tissue. Controls for the microscopy studies included observations on cottonseeds with no visual signs of infection and on laboratory-grown cultures of A. flavus. These observations demonstrated that the hyphae localized within fluorescent seeds had features characteristic of A. flavus and that fungal-like structures do not occur within uninfected seeds. 相似文献

In Argentina, there is rather little information about the natural occurrence of mycotoxins in feedstuffs. The aim of this
work was to determine the fungal flora and natural incidence of aflatoxin B1 (AFB1), zearalenone (ZEA) and deoxynivalenol
(DON) in poultry feeds from 5 factories of Río Cuarto, Córdoba. Three hundred samples were taken from May 1995 to May 1996.
Fungal counts of poultry feeds ranged 104 to 106 CFU g-1. The lowest counts were obtained on the first months from the sampling
(May to September 1995) with mean values significantly different from those found at the last of the sampling (October 1995
to April 1996). The most prevalent species isolated of poultry feed samples belonged to the genera Penicillium that was present
in 98% of the samples, Fusarium (87%) and Aspergillus (52%). Fusarium species isolated were: F moniliforme in 73% of the samples,
F subglutinans (35%), F graminearum (20%) and within Aspergillus species: A. parasiticus (33%) and A. flavus (8%) were identified.
In poultry feeds aflatoxin B1 (AFB1) was the most significant mycotoxin with levels ranging from 17 to 197 ng/g. For deoxynivalenol
(DON) the levels ranged from 240 to 410 ng/g. Only three out of 300 samples were contaminated with zearalenone (ZEA) in concentrations
of 30, 120 and 280 ng/g. These are preliminary data on this subject in our region.

Bioassay-directed fractionation of an extract of a gibberella ear rot-resistant corn hybrid resulted in the isolation of a fraction that inhibited mycotoxin production, but not growth, of Fusarium culmorum. The fraction contained diferuloylputrescine and a new allelochemical 4-acetyl-benzoxazolin-2-one (4-ABOA). This report describes the isolation of the bioactive fraction and tests of the effect of 4-ABOA on 3-acetyl deoxynivalenol (3ADON) and aflatoxin production. In addition, data are reported on the effects of 4-ABOA and related compounds on a number of insect species and some preliminary studies on the mechanism of insect toxicity. The effect of 4-ABOA on F. culmorum and Apergillus flavus was similar. Production of toxin was reduced without a concomitant reduction of growth. A 50% inhibition of 3ADON production occurred at ca. 4 μM and 0.12 mM for alfatoxin. 4-ABOA was slightly toxic to Spodoptera frugiperda. 相似文献

[14C]Aflatoxin B1 (AFB1) was isolated from cultures of Aspergillus parasiticus grown on [1-14C]sodium acetate. Covalent binding of AFB1 to liver DNA of rat and mouse was determined 6–8 h after oral administration. The effectiveness of covalent binding, expressed as DNA binding per dose in the units of a ‘Covalent Binding Index’ (CBI), (μmol aflatoxin/mol DNA nucleotides)/(mmol aflatoxin/kg animal), was found to be 10 400 for rats and 240 for mice. These CBI partly explain the different susceptibility of the two species for the incidence of hepatic tumors.The corresponding values for pig liver DNA, 24 and 48 h after oral administration, were found to be as high as 19 100 and 13 300. DNA-binding has not so far been reported for this species although it could represent an appropriate animal model for studies where a human-like gastrointestinal tract physiology is desirable.Aflatoxin M1 (AFM1) is a metabolite found in the milk of cows that have been fed AFB1-contaminated diet. [14C]AFM1 was also found to be produced by cultures of A. parasiticus giving a yield of about 0.3% of the total aflatoxins. A test for covalent binding to rat liver DNA revealed a CBI of 2100 showing that AFM1 must also be regarded as a strong hepatocarcinogen. The metabolism of chemical carcinogens has been studied in cultured human bronchus, colon, duodenum, pancreatic duct, and esophagus. Metabolite patterns and carcinogen-DNA adducts are generally qualitatively similar among animal species, individuals within a species, and tissues within an individual. However, wide quantitative differences are observed between individuals in out-bred animal species, including humans. These interindividual differences in amounts of carcinogen-DNA adducts and in activities of enzymes that are important in the metabolism of chemical carcinogens are similar in magnitude (10-to 150-fold) to those observed in pharmacogenetic studies of drug metabolism. The role of these differences as risk factors in human cancer is being investigated. 相似文献

Physiological processes of mung seeds (Vigna radiata variety Pusa Baishakhi) and their germination were found to be affected by different concentrations of aflatoxin B1. Inhibition in seed germination, seedling growth, chlorophyll, protein and nucleic acid syntheses was found to be due to aflatoxin B1. 【目的】研究微嗜酸寡养单胞菌(Stenotrophomonas acidaminiphila)CW117中磷酸吡哆胺氧化酶基因pnpox(phosphopyridoxamine oxidase,pnpox)在维生素B6(VB6)合成中的贡献及对黄曲霉毒素B1(aflatoxin B1,AFB1)的降解活性。【方法】采用基因插入突变方式,对菌株CW117中磷酸吡哆胺氧化酶基因pnpox进行突变,得到突变菌株。通过高效液相色谱法(high performance liquid chromatography,HPLC)检测突变株对AFB1的降解活性,以及突变株中吡哆醇和吡哆醛的合成情况,确定基因pnpox在寡养单胞菌体内VB6合成中的贡献和黄曲霉毒素降解代谢作用。【结果】成功构建了磷酸吡哆胺氧化酶基因突变子pnpox::pK19mobΩ2HMB,突变子吡哆醛的合成量较野生型菌株显著减少,吡哆醇合成量与野生型菌株无显著性差异;同时,突变子与野生型株CW117对AFB1的降解活性未发现显著性差异。【结论】菌株CW117中磷酸吡哆胺氧化酶在吡哆醛合成的过程中起着重要作用,该基因突变会导致VB6的严重缺乏,影响寡养单胞菌正常生长,但该基因对CW117降解黄曲霉毒素无显著性贡献。 相似文献
