Insulin resistance is a key feature of Type 2 diabetes and an important therapeutic target to address glycemic control to prevent diabetic complications. Lifestyle advice is the first step in the ADA/EASD consensus guidelines followed by metformin therapy. Aerobic exercise (AE) can increase insulin sensitivity by several molecular pathways including upregulation of insulin transporters in the cellular membrane of insulin-dependent cells. In addition, AE improves insulin sensitivity by amelioration of the pathophysiologic pathways involved in insulin resistance such as the reduction of adipokines, inflammatory and oxidative stress responses, and improvement of insulin signal transduction via different molecular pathways. 目的:探讨有氧运动对高脂诱导小鼠非酒精性脂肪肝(NAFLD)的影响及其肝脏冠层成纤维细胞生长因子信号调节器2 (CNPY2)-PKR样内质网激酶(PERK)机制。方法:8周龄雄性C57BL/6J小鼠随机分为对照组(C)、对照+运动组(CE),NAFLD模型组(M)和NAFLD模型+运动组(ME),每组10只。C组和CE组小鼠给予普通饲料,M组和ME组小鼠给予高脂饲料(脂肪供能占比为60%),连续喂养18周至实验结束,取小鼠血清和肝脏。CE组和ME组从第10周起进行有氧跑台训练(12 m/min,每次60 min,每周5 d)。检测小鼠血清总胆固醇(TC)、总甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-c)、低密度脂蛋白胆固醇(LDL-c)、丙氨酸氨基转移酶(ALT)和天冬氨酸转氨酶(AST)水平;观察小鼠肝组织病理学形态;检测肝组织CNPY2、PERK、p-eIF2a、CHOP、CNPY2 mRNA、PERK mRNA表达和CNPY2、PERK的阳性表达。结果:与C组比较,M组小鼠的血清LDL-c、TC、TG、ALT和AST水平显著升高(P<0.05),HDL-c水平显著降低(...  相似文献

Twenty-one bacterial associations isolated from the soda lakes of the southern Transbaikal region were found to be able to actively grow at pH 9–10 on methanol as the source of carbon and energy. Two alkalitolerant facultatively methylotrophic strains, Bur 3 and Bur 5, were obtained in pure cultures. Both strains represent gram-negative, nonmotile, bean-shaped, encapsulated cells that reproduce by binary fission. The strains are able to grow at temperatures ranging from 6 to 42°C, with an optimum growth temperature of 25–29°C (strain Bur 3) and 35–37°C (strain Bur 5) and at pH between 6.5 and 9.5, with an optimum pH value of 8.0–8.5. At pH 9.0, strain Bur 3 exhibits an increased content of phosphatidylglycerol and a decreased content of phosphatidylethanolamine. Strains Bur 3 and Bur 5 are similar in the G+C content of their DNAs (66.2 and 65.5 mol %, respectively) and in the type of the dominant ubiquinone (Q 10). Unlike Bur 5, strain Bur 3 is able to grow autotrophically in an atmosphere of CO2+ O2+ H2. The strains oxidize, by the respective dehydrogenases, methanol to CO2, which is assimilated by the ribulose bisphosphate pathway. Ammonium ions are assimilated in the glutamate cycle and by the reductive amination of -ketoglutarate. The strains are highly homologous to each other (92%) and are much less homologous (at a level of 28–35%) to representatives of the genus Ancylobacter, A. aquaticusATCC 25396Tand A. vacuolatumDSM 1277. The steady-state behavior of a glucose-limited, aerobic, continuous cultivation of Saccharomyces cerevisiae CEN.PK113-7D was investigated around the critical dilution rate. Oxido-reductive steady states were obtained at dilution rates up to 0.09 h(-1) lower than the critical dilution rate by operating the bioreactor as a productostat, where the dilution rate was controlled on the basis of an ethanol measurement. Thus, the experimental investigations revealed that multiple steady states exist in a region of dilution rates below the critical dilution rate. The existence of multiple steady states was attributed to two distinct physiological effects occurring when growth changed from oxidative to oxido-reductive: (i) a decrease in the efficiency of ATP production and utilization (at ethanol concentrations below 3 g/L) and (ii) repression of the oxidative metabolism (at higher ethanol concentrations). The first effect was best observed at low ethanol concentrations, where multiple steady states were observed even when no repression of the oxidative metabolism was evident, i.e., the oxidative capacity was constant. However, at higher ethanol concentrations repression of the oxidative metabolism was observed (the oxidative capacity decreased), and this resulted in a broader range of dilution rates where multiple steady states could be found.  相似文献

本研究以高胆固醇饮食小鼠为实验对象,观察有氧运动对脂质水平异常的动物个体血脂及脂蛋白代谢的影响。结果显示,经11周有氧耐力训练后,高脂膳食+运动组小鼠血浆TG、TC及LDL-C水平均显著低于高脂膳食组(P<0.05),而HDL-C/TC和HDL-C/LDL-C比值均显著高于高脂膳食组(P<0.05)。表明长期有氧耐力训练能显著改善高胆固醇饮食小鼠血脂及脂蛋白代谢状况。  相似文献

Propionic acid is commonly found as a fermentation product in the gastrointestinal tracts of food animals and has also been used to limit the microbial contaminants in animal feeds. Because propionic acid is known to have antibacterial activity, the propionic acid encountered by foodborne pathogens during their life cycles may play an important role in inhibiting the survival of the pathogens. The survival patterns of Salmonella typhimurium poultry isolate were determined both in aerobic and anaerobic tryptic soy broth (TSB; pH 5.0 or 7.0) containing various concentrations of propionic acid (0-200 mM). The levels of recovered cells were consistently greater at pH 7.0 compared to those at pH 5.0. For the first 4 days, the levels were significantly decreased by incubation under anaerobic conditions as compared to aerobic condition at pH 7.0 (P<0.05). However, there were fluctuations of cell populations with different patterns depending on both concentrations and growth conditions. To characterize the nature of the capability which allowed the cell multiplication following decreases in cell population during incubation at pH 7.0, the cells isolated from the outgrowth cultures were tested for survival in aerobic or anaerobic TSB (pH 5.0 or pH 7.0) containing propionic acid (50 mM). The outgrowth isolates did not show significant differences in the level of recovered cells in the presence of propionic acid when compared to the wild type strain (P>0.05), suggesting that the cells in the outgrowth cultures did not harbour mutation(s) conferring increased resistance to propionic acid. In addition, the level of recovered cells of isogenic rpoS mutant strain of S. typhimurium was not significantly different from that of the wild type strain in the same assay conditions (P<0.05). The results of this study show that the bactericidal activity of propionic acid on S. typhimurium can be affected by environmental conditions such as acidic pH levels and anaerobiosis in food materials and gastrointestinal tracts. However, S. typhimurium is also able to multiply in the presence of sublethal concentrations of propionic acid at neutral pH during prolonged incubation under both aerobic and anaerobic conditions.  相似文献

[Purpose]

The purpose of this study is to examine the effects of six-weeks of endurance swim training and short-term intake of Rg1 on the expression of related proteins as well as improvement of aerobic exercise capacity in 8-week-old male SD rats.

[Methods]

The groups were divided into placebo (NP, n=6), Rg1 (NRG, n=6), exercise+placebo (EP, n=7), and exercise+Rg1 (ERG, n=7). On completion of the 6-week swimming exercise, Rg1-intake groups were treated with acute uptakes (3 times within 24hrs) of Rg1. After the treatment, all groups were subjected to a swim to exhaustion test, and then the mass of muscle tissue, mRNA expression level and activity of citrate synthase (CS) were analyzed on plantaris.

[Results]

There were no differences in the effect of 6-week swimming exercise and short-term intake of Rg1 on body weight and muscle mass between groups. Although the CS mRNA expression was elevated in the exercise group and combined treatment group, there was no significant difference in CS activity. Acute uptakes of Rg1 did not affect swimming time to exhaustion, but it was increased by 235% and 314% by the 6-weeks of exercise and combined treatment of exercise and Rg1, respectively, which suggests that the combined treatment increased the effect on the capacity of aerobic exercise.

[Conclusion]

Based on these results, it was confirmed that even a short-term treatment of Rg1 can give an additive effect for improvement of exercise function, and additional studies are needed for the mechanisms and modes of its working.  相似文献

【目的】从处理海洋养殖循环水的生物滤器生物膜中分离到1株具有好氧反硝化活性的细菌(菌株2-8),并进一步研究了该菌的分类地位及反硝化特性。【方法】采用16S rRNA基因序列分析对菌株进行初步鉴定,采用好氧培养技术,探讨了碳源种类、起始pH、NaCl浓度、C/N、温度和摇床转速对菌株2-8好氧反硝化活性的影响。【结果】该菌株的16S rRNA基因序列与Pseudomonas segetis FR1439T(AY770691)的相似性最高,达到99.9%,因此初步鉴定菌株2-8属于假单胞菌属(Pseudomonas sp.2-8)。碳源类型和C/N对其好氧反硝化作用的影响最为显著,以柠檬酸钠为唯一碳源,C/N为15时脱氮效率最高,低C/N导致亚硝酸盐的积累;其好氧反硝化的最适温度和pH分别为30℃和7.5;菌株2-8在摇床转速为160r/min下脱氮效果最好;NaCl浓度对其反硝化活性的影响不明显。【结论】在初始硝酸氮浓度为140mg/L,以柠檬酸钠为唯一碳源、C/N为15、pH为7.5、NaCl浓度为30g/L,30℃以及160r/min摇床培养的条件下,菌株2-8在48h内脱氮率可达92%且无亚硝酸盐积累。  相似文献

A comparison was made of the structures of the Fnr and ArcA modulons and regulons. The data on modulon composition were taken from published microarray assays, whereas regulons were characterized using comparative genomic approaches. The regulatory cascade involving Fnr and ArcA contributes greatly to the extension of the Fnr modulon over the Fnr regulon by adding operons of the ArcA modulon. The Fnr and ArcA regulons were shown to contain 26 and 16 operons, respectively. Ten operons had high-score and highly conserved sites for both Fnr and ArcA and were isolated as a so-called core of regulons.  相似文献
