目的:观察有氧运动对2型糖尿病大鼠骨骼肌细胞外信号调节激酶(ERK1/2)活性的影响,探讨有氧运动对2型糖尿病的预防和调控机制。方法:将75只SD大鼠随机分为正常对照组(CON)、糖尿病对照1组(DC1)、糖尿病运动1组(DE1)、糖尿病对照2组(DC2)、糖尿病运动2组(DE2)5组(n=15)。正常对照组用普通饲料喂养,糖尿病组用高脂高糖配方饲料喂养。经过8周高脂高糖喂养后,糖尿病2组大鼠腹腔内注射链脲佐菌素(STZ),诱发2型糖尿病;糖尿病运动1组游泳的最后1周初和糖尿病对照1组同时注射STZ,注射剂量为35 mg/kg,3 d后尾部取血测血糖≥ 16.7 mmol/L为造模成功。运动干预8周后,测定大鼠血清胰岛素、骨骼肌中ERK1/2蛋白表达等指标。结果:①与正常对照组比较,糖尿病各对照组血液中总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)、游离脂肪酸(FFA)显著升高(P<0.05,P<0.01),空腹血糖(FBG)、胰岛素(FIN)含量和胰岛素抵抗指数(HOMA-IR)显著升高(P<0.01),ERK1/2磷酸化的蛋白表达显著下降(P<0.05),糖尿病对照2组ERK1/2蛋白含量显著下降(P<0.05);②8周游泳运动后,与糖尿病对照组比较,糖尿病运动组血液中TC、TG、FFA、LDL-C显著下降(P<0.05),FBG、FIN、HOMA-IR显著下降(P<0.05,P<0.01),ERK1/2磷酸化蛋白表达显著升高(P<0.05)。结论:长时间有氧运动,增加了骨骼肌ERK1/2磷酸化水平,改善了2型糖尿病大鼠胰岛素抵抗的状况,降低血糖。这可能是改善糖代谢紊乱,提高胰岛素敏感性的机制之一。  相似文献

Lab‐scale experiments were conducted to investigate the effects of ferrous iron on nutrient removal performance and variations in the microbial community inside aerobic granular sludge for 408 days. Two reactors were simultaneously operated, one without added ferrous iron (SBR1), and one with 10 mg Fe2+ L?1 of added ferrous iron (SBR2). A total of 1 mg Fe2+ L?1 of added ferrous iron was applied to SBR1 starting from the 191st day to observe the resulting variations in the nutrient removal performance and the microbial community. The results show that ammonia‐oxidizing bacteria (AOB) could not oxidize ammonia due to a lack of iron compounds, but they could survive in the aerobic granular sludge. Limited ferrous iron addition encouraged nitrification. Enhanced biological phosphorus removal (EBPR) from both reactors could not be maintained regardless of the amount of ferrous iron that was applied. EBPR was established in both reactors when the concentration of mixed liquor suspended solid (MLSS) and the percentage of Accumulibacteria increased. A total of 10 mg Fe2+ L?1 of added ferrous iron had a relatively adverse effect on the growth of AOB species compared to 1 mg Fe2+ L?1 of added ferrous iron, but it encouraged the growth of Nitrospira sp. and Accumulibacteria, which requires further study. It could be said that the compact and stable structure of aerobic granular sludge preserved AOB and NOB from Fe‐deficient conditions, and wash‐out during the disintegration period. © 2017 American Institute of Chemical Engineers Biotechnol. 目的:探讨有氧运动对老年大鼠胃肠动力以及血清中胃促生长素(ghrelin)和瘦素(leptin)的影响。方法:雄性SD大鼠45只,分为青年对照组、老年对照组以及老年有氧运动组(n=15)。老年有氧运动组采用递增负荷的方式进行8周跑台运动。实验过程中连续观察并记录各组大鼠的摄食量、体重以及行为学特征;实验结束测定各组大鼠胃排空速率以及小肠推进比,血清Ghre-lin和Leptin。结果:与青年对照组相比,老年对照组大鼠摄食量、胃排空速率、小肠推进比、血清Ghrelin水平明显降低,体重、体脂和Leptin水平显著升高;与老年对照组相比,老年有氧运动组大鼠摄食量、胃排空速率、小肠推进比、血清Ghrelin水平明显升高,体重、体脂和Leptin水平明显降低。结论:有氧运动能促进老年大鼠食欲、提高胃肠动力、改善身体成分,其保护机制可能与血清中Ghrelin含量升高和Leptin含量降低有关。  相似文献

The distribution and species diversity of aerobic organotrophic bacteria in the Dagang high-temperature oil field (China), which is exploited with water-flooding, have been studied. Twenty-two strains of the most characteristic thermophilic and mesophilic aerobic organotrophic bacteria have been isolated from the oil stratum. It has been found that, in a laboratory, the mesophilic and thermophilic isolates grow in the temperature, pH, and salinity ranges characteristic of the injection well near-bottom zones or of the oil stratum, respectively, and assimilate a wide range of hydrocarbons, fatty acids, lower alcohols, and crude oil, thus exhibiting adaptation to the environment. Using comparative phylogenetic 16S rRNA analysis, the taxonomic affiliation of the isolates has been established. The aerobic microbial community includes gram-positive bacteria with a high and low G+C content of DNA, and γ and β subclasses of Proteobacteria. The thermophilic bacteria belong to the genera Geobacillus and Thermoactinomyces, and the mesophilic strains belong to the genera Bacillus, Micrococcus, Cellulomonas, Pseudomonas, and Acinetobacter. A gram-positive bacterium, identified as Corynebacterium K37, was isolated from the waste effluent of a dairy farm. The bacterium thrived and expressed nitrate-reducing activity at nitrate concentrations of up to 2 M, and reduced nitrate concentration from 0.4 M to 11.4 mM and also from 0.4 M to 23.4 mM in aerobic and anaerobic fed-batch cultures, respectively. Cells of K37 were able to utilize a variety of carbon sources for nitrate reduction with little or no accumulation of nitrite. In aerobic cultures, the residual nitrite was minimal and it was completely reduced after prolonged incubation. Growth on acetate or pyruvate in anaerobic cultures resulted in lower nitrite reductase activities and concomitant higher residual nitrite concentrations than did growth on ethanol or glucose, suggesting that diminished electron availability was a factor in the accumulation of residual nitrite. The bacterium also survived in 2 M concentrations of NaCl, KCl, and CaCl2. Corynebacterium sp. K37 may be useful in bioremediation of high nitrate pollution in contaminated soils and water.  相似文献

Aims:  This paper investigates a selection-based acclimation strategy for improving the performance and stability of aerobic granules at a high chloroanilines loading.
Methods and Results:  The experiments were conducted in a sequencing airlift bioreactor (SABR) to develop aerobic granules fed with chloroanilines (ClA). The evolution of aerobic granulation was monitored using image analysis and scanning electron microscopy, and PCR–DGGE analysis of microbial community was performed. The sludge granulation was apparently developed by decreased settling time and gradual increased ClA loading to 0·8 kg m−3 day−1. A steady-state performance of the granular SABR was reached at last, as evidenced by biomass concentration of 6·3 g l−1 and constant ClA removal efficiency of 99·9%. The mature granules had a mean size of 1·55 mm, minimal settling velocity of 68·4 m h−1, specific ClA degradation rate of 0·181 g gVSS−1 day−1. Phylogenetic analysis of aerobic ClA-degrading granules confirmed the dominance of β - , γ -Proteobacteria and Flavobacteria.
Conclusions:  The chosen operating strategy involving step increase in ClA loading and enhancement of major selection pressures was successful in cultivating the aerobic ClA-degrading granules.
Significance and Impact of the Study:  This research could be helpful for improving the stability of aerobic granules via optimizing operating conditions and developing economic feasible full-scale granular bioreactor.  相似文献

Understanding the properties of aerobic sludge granules as hydrogels   总被引:2,自引:0,他引:2  
Aerobic sludge granules are larger, denser microbial aggregates than activated sludge flocs with a smoother and more regular surface, which facilitates greater wastewater treatment intensity. Factors important in their growth are still poorly understood, which is an impediment to the construction and operation of full-scale aerobic sludge granule processes. Data in this article obtained with granules treating an abattoir wastewater provide evidence that aerobic sludge granules are hydrogels. The results also demonstrate a method for characterizing macromolecular associations. The rheological profile of these granules was found to be analogous with that of typical polymer gels. Water uptake or swelling reflects an equilibrium between granule elastic modulus and osmotic pressure, whereby uptake is increased by reducing solute concentration or the elastic modulus. A weakening of the extracellular polymeric substance (EPS) matrix as demonstrated with mechanical spectroscopy was induced by several environmental factors including temperature, pH and ionic strength. Uniform and elastic deformation was observed at low strain. Enzymatic degradation studies indicate that proteins and alpha-polysaccharides were the major granule structural materials. The aerobic sludge granules in the current study were therefore protein-polysaccharide composite physical hydrogels. While aerobic sludge granules treating an abattoir wastewater are used as a case study, many of the fundamental principles detailed here are relevant to other granulation processes. The paradigm established in this study can potentially be applied to better understand the formation of aerobic sludge granules and thus overcome a hurdle in the acceptance of aerobic sludge granulation as an alternative to more traditional wastewater treatment processes.  相似文献

研究了温度、水分和演替阶段及其交互作用对中亚热带丘陵红壤区森林土壤氮素矿化过程及其矿化速率的影响.结果表明:温度和演替阶段对土壤氨化速率影响显著,其中12 ℃<24℃<36 ℃,灌丛林和马尾松(Pinus massoniana)林低于常绿阔叶林(P<0.05);而水分的影响不显著.水分和演替阶段对土壤硝化速率有显著影响,土壤半饱和含水量高于自然含水量及饱和含水量,且马尾松林高于灌丛林(P<0.05);而温度的影响不显著.温度、水分和演替阶段对土壤氮净矿化速率的影响均显著,其中12 ℃<24 ℃<36 ℃,土壤半饱和含水量高于自然含水量和饱和含水量,灌丛林<马尾松林<常绿阔叶林(P<0.05).温度升高有利于提高土壤氨化速率和净矿化速率,温度过高则抑制土壤硝化速率;土壤含水量适中有利于土壤氮素矿化过程;顺行演替将提高土壤供氮能力,且抑制过强的硝化作用.  相似文献

Bioenergetics of the aerobic bacteriochlorophyll a-containing (BCl a) bacterium (ABC bacterium) Roseinatronobacter thiooxidans is a combination of photosynthesis, oxygen respiration, and oxidation of sulfur compounds under alkaliphilic conditions. The photosynthetic activity of Rna. thiooxidans cells was established by the photoinhibition of cell respiration and reversible photobleaching discoloration of the BCl a of reaction centers (RC), connected by the chain of electron transfer with cytochrome c 551 oxidation. The species under study, like many purple bacteria and some of the known ABC bacteria, possesses a light-harvesting pigment-protein (LHI) complex with the average number of 30 molecules of antenna BCl a per one photosynthetic RC. Under microaerobic growth conditions, the cells contained bc 1 complex and two terminal oxidases: cbb 3-cytochrome oxidase and the alternative cytochrome oxidase of the a 3 type. Besides, Rna. thiooxidans was shown to have several different soluble low- and high-potential cytochromes c, probably associated with the ability of utilizing sulfur compounds as additional electron donors.  相似文献

Rhizobium leguminosarum bv trifolii is a soil-inhabiting bacterium that has the capacity to be an effective nitrogen fixing microsymbiont of a diverse range of annual Trifolium (clover) species. Strain WSM1325 is an aerobic, motile, non-spore forming, Gram-negative rod isolated from root nodules collected in 1993 from the Greek Island of Serifos. WSM1325 is produced commercially in Australia as an inoculant for a broad range of annual clovers of Mediterranean origin due to its superior attributes of saprophytic competence, nitrogen fixation and acid-tolerance. Here we describe the basic features of this organism, together with the complete genome sequence, and annotation. This is the first completed genome sequence for a microsymbiont of annual clovers. We reveal that its genome size is 7,418,122 bp encoding 7,232 protein-coding genes and 61 RNA-only encoding genes. This multipartite genome contains 6 distinct replicons; a chromosome of size 4,767,043 bp and 5 plasmids of size 828,924 bp, 660,973 bp, 516,088 bp, 350,312 bp and 294,782 bp.  相似文献
