A water-soluble polysaccharide isolated from the aqueous extract of the corm of Amorphophallus campanulatus was found to contain d-galactose, d-glucose, 4-O-acyl-d-methyl galacturonate, and l-arabinose in a molar ratio 2:1:1:1. Structural investigation of the polysaccharide was carried out using acid hydrolysis, methylation analysis, periodate oxidation study, and NMR studies (1H, 13C, DQF-COSY, TOCSY, NOESY, ROESY, HMQC, and HMBC). On the basis of the above-mentioned experiments the structure of the repeating unit of the polysaccharide was established as:This molecule showed splenocyte activation.  相似文献

RAPD markers linked to beet necrotic yellow vein virus (BNYVV) resistance genes were identified in two Beta vulgaris accessions Holly-1-4 and WB42 using bulked segregant analysis. The polymorphism revealed by the RAPD markers in the F2 generations of WB42 was higher than that of Holly-1-4. The segregation distortion at marker loci was slightly lower in the B. vulgaris × B. maritima cross than in the B. vulgaris × B. vulgaris cross. For Holly-1-4, a RAPD marker was identified in a long distance from the resistance gene of Rz 1 . However, a RAPD marker tightly linked with Rz 2 gene in repulsion phase was detected with an approximate distance of 0.036 rf. This marker was not generation specific and showed high repeatability. The distance between Rz 1 and Rz 2 genes was estimated as 0.464 rf. After the Rz 2 Rz 2 and Rz 2 rz 2 genotypes were identified using ELISA values and repulsion phase RAPD markers, comparison of their ELISA means revealed lack of the gene dosage effects. Nevertheless, under the field or severe infection conditions, the difference between ELISA mean values of the Rz 2 Rz 2 and Rz 2 rz 2 genotypes might be more than that observed in this study and the gene dosage effects of Rz 2 allele might be important.  相似文献

Voltage gated proton channels and NADPH oxidase function cooperatively in phagocytes during the respiratory burst, when reactive oxygen species are produced to kill microbial invaders. Although these molecules are distinct entities, with no proven physical interaction, their presence and activity in many cells appears to be coordinated. Latent TGF-β (LTGF-β) has to be converted to active TGF-β for its activities. Previously, we reported that certain fragments of latency associated peptide (LAP) augmented LTGF-β activation via increase in binding of LTGF-β to the endothelial cell (EC) surface followed by cell-associated proteolysis. By searching for EC membrane proteins crosslinked with the LAP fragment, we identified the molecule bound to LAP fragment as vimentin. Moreover, the LAP fragment-induced LTGF-β activation was attenuated by anti-vimentin antibody. These results indicate that binding of the LAP fragment to vimentin on the cell surface is indispensable for LTGF-β activation by the LAP fragment.

Since an increased serum unconjugated bilirubin (UCB) level has been proposed as an independent protective factor against atherosclerotic disease, we investigated the molecular events at the basis of this effect. HUVEC and H5V cells were treated with TNFα and UCB and the effects assessed on E-selectin, VCAM-1 and ICAM-1. In HUVEC cells, UCB blunted the TNFα-induced gene upregulation of E-selectin VCAM-1 and ICAM-1. The same pattern was observed in H5V cells except for ICAM-1. UCB also inhibited the PMN endothelial adhesion in HUVEC H5V cells. Western blot and FACS analysis confirmed that UCB prevented TNFα-induced over-expression of adhesion molecules proteins in H5V cells. These data contribute to further explain the protective effect of bilirubin against development of atherosclerosis.  相似文献

In a mark–recapture study in 2006, migrating European Anguilla anguilla silver eels were caught, tagged and released in the Baltic Sea and recaptures in commercial pound nets examined for possible effects on migration of infection with the swimbladder parasite Anguillicola crassus . The overall recapture rate was 36%. The prevalence of infection was lowest at the northernmost sampling site. There were no significant differences between infected and uninfected A. anguilla in condition indices, body fat content and estimated migration speeds. Parasite infection intensity levels were significantly negatively correlated with times and distances covered between release and recapture, but did not correlate with migration speed. It appears that more heavily infected A. anguilla were relatively more vulnerable to recapture in pound nets. It is hypothesized that parasite-induced damage to the swimbladder inhibited vertical migrations and infected A. anguilla tended to migrate in shallower coastal waters, relatively close to the shore.  相似文献

We studied the effect of lipids on the activity of a neutral cysteine proteinase, calpain, in subcellular fractions obtained from the rat brain. Extraction of nearly 23% of membrane cholesterol from the coarse mitochondrial fraction did not result in modifications of specific activity of calpain in this fraction. Detergents (digitonin or Triton Х-100) used in 0.3% concentration enhanced the activity of calpain in the coarse mitochondrial fraction. Examination of the effects of preparations of different phospholipids on the activity of calpain in the cytoplasm demonstrated that only phosphatidylcholine, but not phosphatidylserine and/or cardiolipin, insignificantly increased the activity of calpain (independently of the size and structure of phospholipid vesicles). We hypothesize that the mechanisms underlying interaction between calpain and lipids are not universal; in native cells and model experiments, they can differ noticeably from each other and are modified depending on the corresponding conditions. Staphylokinase (SAK) is a promising thrombolytic agent for treating blood-clotting disorders. Recombinant SAK (rSAK) was produced after integration of the gene into Pichia pastoris genome. The recombinant Pichia carrying multiple insertions of the SAK gene yielded high-level (~1 g/l) of extracellular glycosylated rSAK (~18 kDa) with negligible plasminogen activation activity. Addition of tunicamycin during the induction phase resulted in expression of non-glycosylated and highly active rSAK (~15 kDa) from the same clone. Two simple steps of ion-exchange chromatography produced an homogenous rSAK of >95% purity which suitable for future structural and functional studies.  相似文献

In order to explore the apoptotic and anti-tumor activities of metallo-salens, we synthesized several Mn(III)-salen derivatives (compds. 1-9) and analyzed their effects on cultured human cancer and non-cancer cells. Our results demonstrated that Mn(III)-salen derivatives affect cell viability, induce nuclear condensation and fragmentation in breast cancer cells (MCF7). Mn(III)-salen derivatives also induced caspase-3/7 activation and release of cytochrome-c from the mitochondria to cytosol suggesting that Mn(III)-salen derivatives induce apoptosis in human cells via mitochondrial pathway. Importantly, the nature of the substituent and the bridging spacer between diimino groups on the salen ligand play critical roles in determining the apoptotic activities of Mn(III)-salen derivatives. The IC50 values for the active Mn(III)-salen derivatives lie within the range of 11-40 μM in MCF7 cells. Most importantly, several Mn(III)-salen derivatives showed preferential cytotoxicity (2- to 5-fold) toward malignant breast cells (MCF7) over a non-malignant breast epithelial cell line (MCF10). Notably, the level of cytotoxicity and selectivity of the Mn(III)-salen derivatives towards MCF7 and MCF10 cells are very similar to cisplatin which indicate that Mn(III)-salens are potential novel anti-tumor agent.  相似文献

病毒感染与癌症的关系是生物医学领域中非常重要的研究方向, 估计当前全世界至少20% 人类肿瘤的发生与病毒感染有密切联系。本文对反转录病毒诱发肿瘤的各种作用机制进行了详细阐述。根据致病速度的快慢, 反转录病毒被分为两大类: 能迅速诱导肿瘤产生的急性转化反转录病毒和缓慢诱导肿瘤产生的非急性转化反转录病毒。急性转化反转录病毒通过其自身携带的癌基因快速诱导肿瘤产生, 而细胞原癌基因的插入激活则是非急性转化反转录病毒引起肿瘤的主要机制。对反转录病毒致瘤机制的研究揭示了肿瘤发生过程中的一些重要原理和机制, 包括原癌基因激活以及肿瘤抑制基因失活等, 对探讨非病毒引起的人类肿瘤的发生机制同样具有重要的参考意义。  相似文献
