N-α-biotinylated-neuropeptide Y analogs: Syntheses, cardiovascular properties, and application to cardiac NPY receptor visualization

Two monobiotinylated analogs of neuropeptide Y (NPY) were synthesized by coupling the N-hydroxysuccinimidyl esters of biotin and (6-biotinylamido)-hexanoic acid, respectively, to the free alpha-NH2 group of the side chain protected NPY peptide resin. Crude peptides obtained by HF cleavage were purified by RPLC and their integrities were confirmed by amino acid and mass spectral analysis. As with NPY, both biotinylated analogs inhibited 125I-NPY binding and adenylate cyclase activity of rat cardiac ventricular membranes in a dose-dependent manner. N-alpha-[(6-biotinylamido)-hexanoyl]-NPY exhibited potencies comparable to that of NPY whereas N-alpha-biotinyl-NPY was slightly less potent. In the in vivo experiments, however, both the biotinylated analogs exhibited responses comparable to NPY in increasing arterial blood pressure and decreasing heart rate in anesthetized rats. The responses of the biotinyl analogs were longer lasting than those of NPY. Histochemical studies revealed that N-alpha-[(6-biotinylamido)-hexanoyl]-NPY could label the NPY receptors in rat cardiac ventricular tissues. This labeling was specific since intact NPY inhibited the staining. These studies show that biotinyl-NPY analogs exhibit biological potencies comparable to intact NPY and can therefore be used to further probe the NPY-receptor interaction.     

Microwave irradiation increases recovery of neuropeptides from brain tissues

The effect of focused high energy microwave treatment (MW) on brain concentrations and molecular forms of substance P, neurokinin A, neuropeptide Y, neurotensin, galanin and calcitonin gene-related peptide was investigated. Groups of rats were treated as follows: 1) MW, storage for 60 min at 22°C, 2) Decapitation, storage for 60 min at 22°C, 3) Decapitation, storage for 60 min at 22°C, MW treatment, 4) MW, decapitation, storage for 2 min at 22°C and 5) Decapitation, storage for 2 min at 22°C. Peptide concentrations were in all instances highest in the MW sacrificed groups. MW increased the concentration of intact peptides by rapid inhibition of peptidase activity and increase in peptide solubility/extractability.     

Molecular cloning and restriction enzyme analysis of a long repetitive DNA sequence in rice

Rice long repetitive DNA (9–20 kbp) reassociating at Cot 50 M.s was cloned in pBR325. Out of several recombinants (Cam^r Amp^r Tet^s), only a few were selected randomly for further characterization. The insert size in all these clones was 3–4 kbp. Restriction enzyme analysis showed the absence ofEcoRI andBclI sites, presence of a singlePstI andPvuII site and multiple sites forAluI in 3 clones namely pRLl, pRL7 and pRL10. TheBamHI-PstI fragment of about 0.4 kbp in the pRL7 insert DNA (pRL7-0.4 kbp) was subcloned in M13mpl8 and partially sequenced using Sanger’s dideoxynucleotide chain termination method. Dot matrix comparison of this sequence with rice rDNA sequences revealed low homology with the 25 S rDNA sequence of rice, however, hybridisation did not indicate any homology.

黄连木五倍子的生态研究与利用

一、产地自然概况黄连木五倍子最早发现于河南省南部信阳市郊贤山脚下(东经114°0′,,北纬32°10′)。该地为低山丘陵,海拔90一320m,南部紧连鸡公山厂傍依南湾水库和狮河。     

水分胁迫对小麦叶片光合作用的影响及其与抗旱性的关系

在水分胁迫初期,两个小麦品种叶片光合速率,气孔导度和细胞间隙CO_2浓度降低,气孔限制值增加,光合速率的降低主要是气孔因素的限制。中度到严重水分胁迫使叶片光合速率、气孔导度和气孔限制值降低,细胞间隙CO_2浓度明显增加,且叶圆片放氧能力,叶绿体Hill反应、叶绿素荧光强度和表观量子产额降低,此时光合速率的降低主要是叶肉细胞光合活性的下降引起的。抗旱性弱的郑引一号叶肉细胞光合活性比抗旱性强的丰抗13更容易受到水分胁迫的影响。     

小鼠LAK细胞对红系祖细胞及多向造血祖细胞增殖的影响

小鼠脾细胞经重组人白细胞介素-2(rhIL-2)激活后对YAC-1,LP-3和WEHI-164等肿瘤细胞均有很强的杀伤活性。在CFU-E和BFU-E培养体系中,不同浓度LAK细胞与BMC直接加入或预温育4h后再培养,均能加强CFU-E和BFU-E增殖。低浓度LAK细胞(LAK/BMC为0.5)与BMC直接加入或预温育后再加入CFU-mix培养体系中,均能增强CFU-mix增殖,而高浓度LAK细胞和BMC(LAK/BMC=8.0)直接加入培养体系则抑制CFU-mix增殖;若共温育后再培养则非常明显地抑制CFU-mix增殖,CFU-mix仅为对照的17.6％。小鼠LAK细胞对造血祖细胞体外增殖具有调节作用,这种调节可能包括分泌某些细胞因子以及细胞间直接相互作用两种方式。

“引黄济青”输水沿线水生生物分布与蓄水库富营养化关系的研究

“引黄济青”是国家“七五”重点工程,由黄河下游博兴县境内打渔张闸引水,在渠首沉沙后,再经惠民、东营、潍坊和青岛的10个县区约253km的明渠输送,到达唠山县棘洪滩蓄水库蓄存,实现向青岛市日供水3.0×10~5m~3。明渠输水过程中,来白沿线的营养物质会污染水体,加之输水在库中滞留时间长,会不会发生富营养化,这是社会普遍关注的问题。
