缓激肽对背根节神经元钠通道电流的作用   总被引:1,自引:0,他引:1  
目的:观察缓激肽(bradykinin,BK)对大鼠背根节神经元电压依赖性钠通道电流的作用。方法:采用全细胞膜片钳技术,记录钠通道电流。结果:缓激肽剂量依赖性(0.01~10μmol/L)增高小细胞背根节神经元诱发放电频率;缓激肽剂量依赖性(O.01~10μmol/L)增加小细胞背根节神经元的河豚毒素不敏感(TTX—resistant,TTX—R)钠电流,对TTX敏感(TTX—sensitive,TTX-S)钠电流无明显影响。结论:缓激肽引起炎性痛的机制可能与TTX-R钠通道电流有关。  相似文献

苹果酸脱氢酶(MalateDehydrogenase,MD)是一种氧化还原性酶,参与体内多种能量代谢反应。它可以催化苹果酸氧化脱羧生成丙酮酸和CO2,并使NADP+还原成NADPH,NADPH是脂肪酸合成所必需的载体,棕榈酸可以利用生成的NADPH来合成长链脂肪酸,MD的活性与脂肪酸合成效率之间存在密切的相关,MD也参与体内骨骼肌、心肌的能量代谢,并对肌纤维的生长有一定的调节作用。根据鸡MD基因的5′侧翼区序列设计一对引物,用直接测序的方法在侧翼区检测多态性位点,在235bp(GenBank登录号:U49693)处发现一个SNP位点,此位点是一个限制性内切酶(SphⅠ酶)发生变化的位点。以东北农业大学高低脂双向选择系的第8世代肉鸡和东农F2资源群体为实验材料,用PCR-RFLP的方法进行基因型分析,建立适合的统计模型,进行基因型与生长和体组成性状的相关分析。结果表明:在高低脂系第8世代肉鸡中AA基因型个体的腹脂重和腹脂率显著高于BB基因型个体(P<0.05);BB基因型个体的大胸肌重和大胸肌率显著高于AA基因型个体(P<0.05)。在东农F2资源家系中BB基因型个体的大胸肌重和大胸肌率显著高于AA和AB基因型个体(P<0.05);AA基因型个体的肝脏重和肝脏率显著高于BB基因型个体(P<0.05)。综上所述,MD基因可能是影响鸡生长和体组成性状的主效基因或与控制生长和体组成性状的主效基因相连锁。  相似文献

The Grueneberg ganglion (GG) is a cluster of neurones present in the vestibule of the anterior nasal cavity. Although its function is still elusive, recent studies have shown that cells of the GG transcribe the gene encoding the olfactory marker protein (OMP) and project their axons to glomeruli of the olfactory bulb, suggesting that they may have a chemosensory function. Chemosensory responsiveness of olfactory neurones in the main olfactory epithelium (MOE) and the vomeronasal organ (VNO) is based on the expression of either odorant receptors or vomeronasal putative pheromone receptors. To scrutinize its presumptive olfactory nature, the GG was assessed for receptor expression by extensive RT-PCR analyses, leading to the identification of a distinct vomeronasal receptor which was expressed in the majority of OMP-positive GG neurones. Along with this receptor, these cells expressed the G proteins Go and Gi, both of which are also present in sensory neurones of the vomeronasal organ. Odorant receptors were expressed by very few cells during prenatal and perinatal stages; a similar number of cells expressed adenylyl cyclase type III and G(olf/s), characteristic signalling elements of the main olfactory system. The findings of the study support the notion that the GG is in fact a subunit of the complex olfactory system, comprising cells with either a VNO-like or a MOE-like phenotype. Moreover, expression of a vomeronasal receptor indicates that the GG might serve to detect pheromones.  相似文献

将编码人β神经生长因子(Huβ-HGF)的基因克隆到由T7噬菌体启动子控制的pET11c大肠杆菌表达载体中,重组质粒经鉴定含有Huβ-NGF基因,未解聚的表达产物经SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE),结果显示出二聚体27kD的蛋白带。而完全解聚的表达产物SDS-PAGE显示出一条13β5kD单体带。经凝胶电泳扫描,表达带占菌体总蛋白的14.5%。用兔抗鼠β—NGF的多克隆抗体进行的Western-Blot的结果表明,二聚体同单体都有免疫原性。在生物活性的鉴定中,菌体表达产物可以使小鸡鸡胚的背根神经节产生神经元突起,由此可以证明该表达产物有较高的生物活性。  相似文献

报道了虎纹捕鸟蛛(Ornithoctonus huwena)食道下神经节的解剖以及神经细胞的分离培养方法。为开展蜘蛛神经生物学实验方法的研究,创立了一种蜘蛛神经细胞解剖方法——去胸甲解剖法。神经节细胞培养基为NaCl2 23 mmol/L;KCl6.8 mmol/L;CaCl28 mmol/L;MgCl2 5.1 mmol/L;Sucrose 5 mmol/L;Hepes 10 mmol/L;谷氨酰胺1mmol/L;青霉素200IU/ml;链霉素200μg/ml;小牛血清20%;pH7.4。在温度(27±2)℃的培养箱中培养2~4h。实验结果表明与传统方法相比较,去胸甲解剖法具有取材简便、准确、快捷和高效的特点。改进的培养基非常适合蜘蛛离体神经细胞的培养。培养的细胞状态好、数量多,细胞体呈椭圆形,细胞形状近似汤勺,有一个长的单极突起,其大小为10~30μm。  相似文献

In some arthropods, females learn in the context of reproduction to refine their mate choices and avoid males displaying traits associated with impotency. Previous studies have shown that males of the Asian citrus psyllid, Diaphorina citriKuwayama (Hemiptera: Psyllidae), associate female odor with copulatory rewards. However, it is unclear whether females similarly learn about male traits. We compared mate choice in females previously mated to either blue or orange males and found that females may associate male color with reproductive success, and avoided blue males after previous experience. Orange males mated more frequently than blue males and appeared to be more sexually aggressive in their mating attempts. In addition, females mated to orange males laid approximately twice as many eggs as those mated to blue males. We dissected male psyllids and measured the size of their reproductive organs to determine whether abdominal color was associated with reproductive development. Our morphometric analysis indicated that blue males may not be physiologically immature compared with orange males. Rather, blue males displayed larger testes and seminal vesicles than orange males, suggesting that differences in reproductive output may be behavioral in nature. Based on our data, we suggest that females learn about the appearance and mating behaviors of blue and orange males from early mating experiences, and subsequently avoid blue males in future matings. Although the roles of blue males within a population are unclear, it is possible that blue morphs may specialize in other functions such as dispersal.  相似文献

Our previous study found that co-culture with human vascular endothelial cells (HMVECs) is beneficial for dorsal root ganglion cells (DRGCs). The goal of the present study is to investigate whether co-culture with HMVECs could promote the development of DRGCs, and whether this effect is induced by the secretion of BDNF by HMVECs. DRGCs were mono-cultured, co-cultured with HMVECs or co-cultured with HMVECs that pre-transfected with BDNF siRNA, the expression of neurite formation and branching factors were determined. The results showed that transfecting with BDNF siRNA inhibited BDNF expression and reduced BDNF secretion. Co-culture with HMVECs increased the expression of Etv4, Etv5, FN-L, FN-M, and GAP-43 in DRGCs that accompanied by the activation of ERK pathway. However, these changes were all reversed by the inhibition of BDNF in HMVECs. In conclusion, our data demonstrate that HMVECs potentiated DRGCs development at least partly by the secretion of BDNF in the co-culture system.  相似文献

The autonomic nervous system consists of sympathetic and parasympathetic nerves, which functionally antagonize each other to control physiology and homeostasis of organs. However, it is largely unexplored how the autonomic nervous system is established during development. In particular, early formation of parasympathetic network remains elusive because of its complex anatomical structure. To distinguish between parasympathetic (cholinergic) and sympathetic (adrenergic) ganglia, vesicular acetylcholine transporter (VAChT) and choline O‐acetyltransferase (ChAT), proteins associated with acetylcholine synthesis, are known to be useful markers. Whereas commercially available antibodies against these proteins are widely used for mammalian specimens including mice and rats, these antibodies do not work satisfactorily in chickens, although chicken is an excellent model for the study of autonomic nervous system. Here, we newly raised antibodies against chicken VAChT and ChAT proteins. One monoclonal and three polyclonal antibodies for VAChT, and one polyclonal antibody for ChAT were obtained, which were available for Western blotting analyses and immunohistochemistry. Using these verified antibodies, we detected cholinergic cells in Remak ganglia of autonomic nervous system, which form in the dorsal aspect of the digestive tract of chicken E13 embryos. The antibodies obtained in this study are useful for visualization of cholinergic neurons including parasympathetic ganglia.  相似文献
