Since 2006, winter melon plants (Cucumis melo L. var inodorus) showing symptoms of pin‐point yellow spots were noticed in Sicily (Italy). Leaf samples were tested by enzyme‐linked immunosorbent assay to the most important viruses‐infecting cucurbits. Zucchini yellow fleck virus (ZYFV, genus Potyvirus) was the only virus detected. Surveys in 2007 and 2008 revealed an increasing number of sites in Sicily with ZYFV‐infected winter melon plants. To confirm the identity of the virus as ZYFV, two isolates from different locations were sequenced and shown to be approximately 85% identical to the published sequences of isolates previously identified in Italy and France. This is the first report of ZYFV occurring on melon in Italy.  相似文献

Zucchini yellow mosaic virus (ZYMV), Papaya ringspot virus – type W (PRSV‐W) and Zucchini lethal chlorosis virus (ZLCV) cause important diseases on zucchini squash crops in Brazil. ZYMV and PRSV‐W belong to the genus Potyvirus and are transmitted by aphids, whereas ZLCV belongs to Tospovirus and is transmitted by the thrips Frankliniella zucchini. These three viruses may occur simultaneously in the field, and the epidemiology of the corresponding diseases may be determined by interactions among viruses, hosts and vectors. In this work, the progress of the diseases caused by these viruses was studied over a temporal and geographic range for three planting seasons (PS). For the lethal chlorosis (ZLCV), a monomolecular model was found to be the best fit for the data, though only during the third PS. For data collected during the first two PS, the Gompertz model was found to fit the data best. The spatial distribution of disease indicated disease aggregation at the end of the crop cycle. For the yellow mosaic (ZYMV), the model that best fit in the 1st PS was the logistic and in the 2nd and 3rd PS was monomolecular. The spatial pattern of the disease was random when the disease incidence was low but aggregated when the disease incidence was high. The common mosaic (PRSV‐W) showed the lowest incidence in all three PS. An exponential model was the best fit for data collected during all PS, and the spatial pattern of the disease was random. Interactions among the three viruses apparently did not result in changes in the epidemiology of the diseases. Removal of sources of inoculum and planting at an unfavourable time for reproduction of virus vectors are the two main measures recommended for the control of these diseases. The use of insecticide is indicated only for the control of the F. zucchini.  相似文献

Methods to obtain moult data from wild birds have not changed much over the last century and most studies still depend on checking museum specimens or capturing birds. Here we assess the applicability of systematic field photography for detecting and scoring moult in adult Black Skimmers Rynchops niger from southern Brazil. Moult data extracted from photographs have a high within‐ (RGLMM = 0.98) and between‐observer repeatability (RGLMM = 0.97) and show very good fit to current Underhill–Zucchini moult models (R2 = 0.75). Photography offers the advantages of being less invasive, requiring less equipment and human effort, being feasible in areas where captures may not be possible, and causing less disturbance, so enhancing the number of sampled individuals.  相似文献

Analysis of phloem exudates from the fruit of Cucurbitaceae revealed the presence of several compounds with UV-visible absorption spectra identical to that of l-ascorbic acid. In Cucurbita pepo L. (zucchini), the compounds could be isolated from phloem exudates collected from aerial parts of the plant but were not detected in whole tissue homogenates. The compounds isolated from the phloem exudates of C. pepo fruit were eluted from strong anion exchange resin in the same fraction as l-ascorbic acid and were oxidised by ascorbate oxidase (E.C. 1.10.3.3). The major compound purified from C. pepo fruit exudates demonstrated similar redox properties to l-ascorbic acid and synthetic 6-O-glucosyl-l-ascorbic acid (6-GlcAsA) but differed from those of 2-O-glucosyl-l-ascorbic acid (2-GlcAsA) isolated from the fruit of Lycium barbarum L. Parent and fragment ion masses of the compound were consistent with hexosyl-ascorbate in which the hexose moiety was attached to C5 or C6 of AsA. Acid hydrolysis of the major C. pepo compound resulted in the formation of l-ascorbic acid and glucose. The purified compound yielded a proton NMR spectrum that was almost identical to that of synthetic 6-GlcAsA. A series of l-ascorbic acid conjugates have, therefore, been identified in the phloem of Cucurbitaceae and the most abundant conjugate has been identified as 6-GlcAsA. The potential role of such conjugates in the long-distance transport of l-ascorbic acid is discussed.  相似文献

A flexuous filamentous virus 750 nm in length was isolated from field-grown melons (Cucumis melo L.) and squirting cucumber (Ecballium elaterium L.). Infection of melon seedlings of most commercial varieties resulted in a lethal wilting, but late infections gave dieback. The virus infected systemically all economically important cucurbit crops. Its coat protein consists of a single polypeptide, 34 500 D in size, encapsidating a single stranded RNA genome of 10.1 kb. Antiserum against the Italian zucchini yellow fleck virus (ZYFV) reacted specifically with the Israeli isolate in both ISEM and Western blot. The virus was aphid transmissible in a non-persistent manner. Based on host range, host reaction and serological data, it is suggested that the virus described is an Israeli variant of ZYFV.  相似文献

研究了来自中国大陆9个小西葫芦黄化花叶病毒(ZYMV)分离物的基因组3′末端核苷酸序列及所推导的外壳蛋白(CP)氨基酸序列以及3′末端非编码区(UTR)序列,并与其它地区所报道的16个ZMYV分离物进行了同源性比较。ZYMV CP基因核苷酸序列具有一定的寄主相关性和地域相关性,但总体上其关联程度不明显;同时,CP氨基酸序列的寄主适应性程度明显高于地域相关性。25个ZYMV分离物的CP氨基酸序列根据其变异程度分为2个区: N端约41个氨基酸为高度变异区,CP核心区和C端氨基酸序列为保守区。研究结果初步揭示了ZYMV作为单链RNA病毒通过与寄主相互作用而表现寄主适应性变异的趋势。  相似文献

Summary Zucchini yellow mosaic virus (ZYMV) and Aphis gossypii Glover are two components of a recently identified plant-parasite system that provides an excellent opportunity to study interrelations between a virus and a vector that share the same host, but have no direct physiological interaction. In a field experiment we documented numbers of alate and apterous A. gossypii on healthy Cucurbita pepo and on plants inoculated with virus 0, 7, 14, and 21 days before aphid infestation. When plants were inoculated and infested simultaneously, more than twice as many alatae were produced after 20 days of colony growth than on any other treatment. This indicates that properties unique to the early stages of viral infection somehow stimulated wing formation. Because it is spread by the activities of alatae, virus dispersal would be greater as a result of these properties. Developmental rate, total numbers of aphids, and numbers of alatae and apterae decreased as the time between virus inoculation and aphid colonization increased.  相似文献

在广西临桂罗汉果花叶畸形病株上获得了一个线状病毒分离物LGL-1,寄主范围、蚜传能力测定、病毒粒子形态和细胞病理特征研究表明,它是马铃薯Y病毒科成员.采用马铃薯Y病毒科特异性简并引物做PCR扩增,并测定了分离物LGL-1的基因组3′-末端序列,序列分析表明它是小西葫芦黄花叶病毒(ZYMV).系统进化树分析揭示,世界范围内的ZYMV分离物主要可分为3大群体,分离物LGL-1为中国特有群体Group Ⅲ成员.原核表达制备了分离物LGL-1的外壳蛋白抗血清,明确了ZYMV是引起广西罗汉果病害的主要病毒,并且比较了原核表达法和提纯病毒法制备的抗血清,在病样的间接ELISA法检测中结果的差异.  相似文献

通过鉴别寄主反应、病毒部分序列测定确定了采自广州白云区表现花叶、斑驳症状的节瓜上的病毒为ZYMV。采用RT PCR方法扩增和克隆了该病毒的外壳蛋白基因 ,连接到原核表达载体pET 2 2b( )上。获得的重组子pET ZCP转化大肠杆菌BL2 1(DE3)后 ,用IPTG进行诱导表达。SDS PAGE和Westernblot分析表明 ,CP基因在大肠杆菌中获得了高效表达 ,融合蛋白分子量约为 33 0kD。将融合蛋白纯化后免疫兔子 ,获得了特异性较高的抗血清。ELISA测定其效价为 1 4 0 96  相似文献
