Summary Comparison of different post-plating temperature regimes with a control treatment (27° C) revealed that a short-term cold (8/14°C:2/2 days or 14°C:4 days) as well as a heat treatment (30°C:14 days) increased the production of embryro-like-structures (ELS) from cultured maize anthers. The beneficial effects of short-term cold treatments were magnified 2–3 times when L-proline (PROL) was added to the induction medium (125–500 mg/L). In the best treatment (14°C:4 days, 125 mg/L L-proline) one genotype produced 143.5 ELS/100 anthers. Anthers subjected to high temperature (30°C:4 days, 30°C:7 days, 30°C:14 days) generally showed a lower response than did cold treated anthers, although genotypic differences were observed. The entomopathogenic fungus,Beauveria bassiana (Balsamo) Vuillemin, was applied to whorl-stage (V7) corn,Zea mays L., by foliar application of a granular formulation of corn grits containing conidia or by injection of a conidial suspension. All plants were infested with European corn borer larvae,Ostrinia nubilalis (Hübner), at the V7 (whorl), V12 (late-whorl), or V17 (pretassel) stage of plant development. Plants infested at whorl and late-whorl stages had significantly more European corn borer tunneling than did plants infested at the pretassel stage. The percentage of plants colonized byB. bassiana did not differ significantly among the whorl, late-whorl, and pretassel stages. As the plants matured,B. bassiana was isolated from different plant areas, with the pith more frequently colonized than the leaf collars. Foliar application ofB. bassiana provided immediate suppression ofO. nubilalis in those plants infested at whorl stage. The reduced efficacy ofB. bassiana at the intermediate plant stages relative to efficacy at harvest is discussed. TrilineeIlus clathrocutis n.g., n.sp. is described and illustrated. It was found as an associate of corn (Zea mays) in Stockton, Georgia, USA, and is related to a group of Tylenchorhynchus sensu lato species having three lines in nonareolated lateral fields. This new species is closely related to Tylenehorhynehus divittatus Siddiqi 1961, T. sculptus Seinhorst 1963, and T. triglyphus Seinhorst 1963 (syn. T. chonai Sethi & Swarup 1968) Tarjan 1973. It differs from these species primarily by having longitudinal striae on the body. These four species are differentiated from Tylenchorhynchus sensu stricto by having three lateral lines instead of four. They differ from Uliginotylenchus Siddiqi 1971 by having nonareolated lateral fields, fewer than 25 annules on conoid rounded tails, differently shaped gubernacula, nonattenuated stylets, and other distinctive characters. They differ from Triversus Sher 1973 by having the male tail enclosed by the bursa and by having rounded female tails. SEM observations of T. clathrocutis reveal a cuticle deeply cut by longitudinal and horizontal striae and bearing wide (> 2.0 μm) annules. Trilineellus is proposed to accommodate the new species and the three-incisured species still within Tylenchorhynchus. Tylenchorhynchus is thereby the repository for species within Tylenchorhynchinae having four lines in the lateral field, no conspicuous labial disc, and bursa enclosing the male tail.  相似文献

Summary In a field experiment, more than 22% increase in the grain yield of corn was obtained by the application of 50 kg ZnSO4/ha. Grain yields were also increased by increasing the level of applied phosphorus. Positive relationship was obtained between Zn and P, the phosphorus treatment increased Zn uptake by grains and vice versa.  相似文献

Summary The location of materials containing terminal fucose residues on the surface of axenic and field grown roots of corn has been determined.Binding patterns of FITC-labelled,Lotus purpureus Moench lectin indicate the presence of the fucose residues in the cell walls and mucilage of the peripheral region of the root cap. During development, fucose residues also appear in the outer periclinal walls and overlying mucilage of columnar epidermal cells. Surface material rich in these residues persists between the mature root hairs but is not found on their surface. Fucose-rich mucilage is present on the exposed surface of aerial roots and at the point where they enter the soil. No lectin binding residues are indicated elsewhere in the roots.  相似文献

Summary Two families each of wheat (Triticum aestivum L.), barley (Hordeum vulgare L.) and maize (Zea mays L.) were studied for mitochondrial heterosis and in vitro mitochondrial complementation. Inbred parents and their hybrids were compared for seedling heights and rate of oxygen uptake by the whole tissue to find out if the hybrids showed greater growth and respiratory activity at the seedling stage. Further comparisons were made by isolating mitochondria from the seedling tissues and measuring their ADP0 ratio, respiratory control ratio and cytochrome c oxidase activity for mitochondrial heterosis. Mixtures of parental mitochondria were similarly compared with parental and hybrid mitochondria for in vitro mitochondrial complementation. No evidence for mitochondrial heterosis or in vitro mitochondrial complementation was found, nor any correlation between the different mitochondrial parameters, seedling heights or rates of oxygen uptake by seedling tissue. The suggested use of mitochondrial heterosis and in vitro mitochondrial complementation for plant breeding is discussed.Data for this paper is taken from the author's dissertation written as a part of Ph.D. degree requirements at the Biology Department, Texas A & M University, College Station, Texas  相似文献

The formation of lysigenous cavities (aerenchyma) in the root cortex of maize, Zea mays L. cv. Capella, under well-aerated conditions has been studied in relation to the composition of the nutrient solutions. Nitrogen, either supplied as nitrate or as ammonium, reduced the cavity formation by the roots. This reduction was most apparent at nitrate concentrations above 2 mM. Cavities were increasingly formed when the nitrate concentration was decreased and they reached their largest dimensions in roots growing in water. Thus, inadequate availability of nitrogen leads, under acrated conditions, to deterioration of cortex cells and cavity formation in the maize roots. It is suggested that cavity formation in these roots is connected with reduced nitrogen assimilation.  相似文献

Summary A method was devised for the biochemical selection of immature, haploid Zea mays embryos using Adh1 and either the Stock 6 or indeterminate gametophyte (ig in W23) high haploid-inducing systems. Haploid (Adh1 ) embryos survived exposure to levels of allyl alcohol which killed diploid (Adh1 +/Adh1 ) embryos. Of the total surviving embryos which were examined cytologically 15% (using ig) and 22% (using Stock 6) were haploid. In two experiments with Stock 6, 100% of the surviving embryos were haploid. To obtain maximum effectiveness of Stock 6 and ig, Adh1 was transferred to stock 6 and W23 backgrounds. Immature, haploid embryos are being used to develop haploid, morphogenic tissue cultures of Zea mays.  相似文献

Genome structure exhibits remarkable plasticity within Zea mays. To examine how haplotype structure has evolved within the Andropogoneae tribe, we have analyzed the bz gene‐rich region of maize (Zea mays), the Zea teosintes mays ssp. mexicana, luxurians and diploperennis, Tripsacum dactyloides, Coix lacryma‐jobi and Sorghum propinquum. We sequenced and annotated BAC clones from these species and re‐annotated the orthologous Sorghum bicolor region. Gene colinearity in the region is well conserved within the genus Zea. However, the orthologous regions of Coix and Sorghum exhibited several micro‐rearrangements relative to Zea, including addition, truncation and deletion of genes. The stc1 gene, involved in the production of a terpenoid insect defense signal, is evolving particularly fast, and its progressive disappearance from some species is occurring by microhomology‐mediated recombination. LTR retrotransposons are the main contributors to the dynamic evolution of the bz region. Common transposon insertion sites occur among haplotypes from different Zea mays sub‐species, but not outside the species. As in Zea, different patterns of interspersion between genes and retrotransposons are observed in Sorghum. We estimate that the mean divergence times between maize and Tripsacum, Coix and Sorghum are 8.5, 12.1 and 12.4 million years ago, respectively, and that between Coix and Sorghum is 9.3 million years ago. A comparison of the bz orthologous regions of Zea, Sorghum and Coix with those of Brachypodium, Setaria and Oryza allows us to infer how the region has evolved by addition and deletion of genes in the approximately 50 million years since these genera diverged from a common progenitor.  相似文献
