This study was designed to identify physiological responses and differential proteomic responses to salinity stress in roots of a salt-tolerant grass species, seashore paspalum (Paspalum vaginatum), and a salt-sensitive grass species, centipedegrass (Eremochloa ophiuroides). Plants of both species were exposed to salinity stress by watering the soil with 300 mM NaCl solution for 20 d in a growth chamber. The 2-DE analysis revealed that the abundance of 8 protein spots significantly increased and 14 significantly decreased in seashore paspalum, while 19 and 16 protein spots exhibited increase and decrease in abundance in centipedegrass, respectively. Eight protein spots that exhibited enhanced abundance in seashore paspalum under salinity stress were subjected to mass spectrometry analysis. Seven protein spots were successfully identified, they are peroxidase (POD, 2.36-fold), cytoplasmic malate dehydrogenase (cMDH, 5.84-fold), asorbate peroxidase (APX, 4.03-fold), two mitochondrial ATPSδ chain (2.26-fold and 4.78-fold), hypothetical protein LOC100274119 (5.01-fold) and flavoprotein wrbA (2.20-fold), respectively. Immunblotting analysis indicated that POD and ATPSδ chain were significantly up-regulated in seashore paspalum at 20 d of salinity treatment while almost no expression in both control and salt treatment of centipedegrass. These results indicated that the superior salinity tolerance in seashore paspalum, compared to centipedegrass, could be associated with a high abundance of proteins involved in ROS detoxification and energy metabolism.  相似文献

Mutation of RAS genes is one of the most common oncogenic alterations in cancer and acquisition of activating RAS mutations has been demonstrated to cause progression of colorectal adenoma to cancer. The aim of this study was to identify changes in the proteome of the intermediate-stage colorectal cancer cell line Caco2, induced by ectopic expression of two distinct RAS proteins, KRAS(V12) and HRAS(V12), in their mutated, constitutively active form. Using 2D-gel electrophoresis, followed by LC-MS/MS we identified almost 200 differentially expressed proteins in pair-wise comparisons of Caco2 vs Caco2-KRAS(V12) and Caco2 vs Caco2-HRAS(V12). Although many of the affected proteins were unique for each pair, there were also substantial similarities. Interestingly, transformation by the mutant KRAS(V12) gene resulted in elevated expression levels and activity of endogenous H-ras protein. Silencing the latter with a specific RNAi reversed several proteomic changes observed in KRAS(V12)-transformed cells, suggesting that oncogenic K-ras partly exerts its effects through endogenous H-ras activation. Alterations in the expression of cytoskeletal and cell adhesion proteins, caused by HRAS siRNA treatment, correlated with a reduction in the invasive properties of Caco2-KRAS(V12) cells. Our data suggest a novel interplay between K-ras and H-ras, with possible implications for colorectal carcinogenesis.  相似文献

天鹅洲迁地保护江豚群体的遗传评估与发展预测   总被引:2,自引:0,他引:2  
文章以2008年春季遭受严重雪灾的天鹅洲白鱀豚国家级自然保护区迁地保护江豚群体为研究对象,利用物种特异性微卫星遗传标记对该群体的遗传多样性进行了评估,并通过软件对其发展趋势进行了模拟和预测。研究结果表明,虽然2008年春季雪灾并未导致该群体遗传多样性的严重损失,灾后群体依然表现出中等水平的微卫星遗传多样性(18个座位的平均等位基因数为4.780,平均观察杂合度和期望杂合度分别为0.748和0.674);但模拟结果显示,该迁地保护江豚群体的遗传多样性将快速下降,100年后仅能保留约79%的等位基因和约84%的遗传杂合度。此外,模拟结果表明,通过增加群体数量和调整群体性比可有效减缓该群体遗传多样性的下降速度。根据研究结果,并结合长江江豚自然种群快速下降的现状,作者提出天鹅洲迁地保护江豚群体当前最重要的保护策略应该是尽早补充一定数量的、合适的野生个体,在加强遗传多样性保护的同时使其群体数量快速增长并形成一个更大规模的有效繁殖群体。  相似文献

采用常规土壤理化分析和Biolog-Eco微孔板鉴定系统,研究了松嫩平原盐碱土地区高羊茅草的生长对原油污染土壤pH、总盐含量和微生物群落的影响.结果表明: 石油污染导致表征土壤微生物代谢活性的平均颜色变化率(AWCD)、物种多样性指数和碳源利用数增加, 微生物碳源利用模式发生改变.高羊茅草对石油污染土壤有较好的修复作用,降低了土壤pH和石油烃(TPH)含量,提高了土壤含水率.高羊茅草根际土壤的AWCD和碳源利用丰富度指数明显高于裸地土壤,为其根际微生物的生长发育提供了适宜的环境.  相似文献

海草是海洋沉水高等植物,属典型的根茎克隆植物.根状茎直径和分株重分别是其个体大小的第一、二表征指标,个体大小是海草重要的种间识别特征.对海草6个形态构件指标和18个生长指标的综合分析表明:果实大小、单株叶面积、分株重具有显著的种特异性;分株发出的时间差、水平根状茎分枝率、叶年产量、分株寿命和垂直茎分枝率是海草种特异性最强的5个生长动态指标,海草生长动态的差异主要体现在克隆生长能力强弱和分株生活史长短上;大海草趋于游击型克隆构型,而小海草则趋于密集型,但小海草Cymodocea nodosa例外;大海草基株水平扩展能力较小海草差.海草个体大小与生长特征的相关生长关系表明:随个体的增大,海草在有机构件生长上表现出两相邻叶、相邻分株、相邻节发出的时间差延长,分株、叶、茎寿命延长的特点;在克隆生长水平上表现出根状茎节间长变短、延伸速率降低,分枝率和根状茎上年产分株数降低,分枝角度变小和间隔子增大的趋势;在克隆片段水平上表现为生理整合性增强;在种群层面则表现出生物量增大和种群密度降低的特点.因此,海草个体大小对其形态、生长特征、克隆构型、种群密度和生产力起到了决定作用.大小海草不同的形态、生长动态和克隆构型特征导致它们的生存策略及生态功能也不同,这一点可能对海草场修复基础理论研究具有一定的指示作用.  相似文献

物种多样性格局是国际生物多样性科学前沿领域热点问题.本文以松嫩平原破碎化羊草草甸退化演替系列(6种植物群落、144个斑块)为研究对象,系统地探讨了其α、β和γ多样性空间格局及其机理.结果表明:在羊草草甸退化演替系列中共发现87种植物,但没有一种能分布于所有斑块;羊草+鸡儿肠群落或羊草群落的α、β和γ多样性较高,多稀有种和特有种;碱地肤群落最低,少稀有种,无特有种;γ多样性与α多样性显著正相关,但与β多样性无相关性.各植物群落的α多样性与单个斑块面积呈显著幂函数关系,β多样性(相似性指数Sjk)仅羊草+鸡儿肠群落呈显著幂函数关系;斑块平均面积和总面积与α、γ多样性呈显著正相关,与β多样性无相关性.群落的物种丰富度越高,稀有种和特有种就越多,物种在局域斑块上灭绝的可能性越大;β多样性在物种多样性格局中的重要性与生境破碎化程度有关.  相似文献

运用8种网目规格的成套浮性刺网作为鱼类采样工具,于2005年夏季在长江中游浅水草型湖泊牛山湖进行鱼类定量采样,通过比较不同茂密程度黄丝草生境中的小型鱼类组成、数量和大小结构,探讨此类湖泊小型鱼类的空间分布特征及其与沉水植被的关系.采样期间共捕获13种1124尾鱼,依据其等级丰度和出现频次,鳖和红鳍原鲌为该湖优势上层小型鱼类.在调查的沉水植物生物量范围内,鱼类物种丰富度和Shannon多样性指数与沉水植物生物量之间呈现倒抛物线关系;两种优势小型鱼类的种群丰度均与沉水植物生物量有着显著的线性正相关关系,且其平均个体大小在裸地生境较高、沉水植被茂密区较低,幼鱼更倾向群聚于厚密的黄丝草生境中;其他生境因子(水深和离岸距离)对鳖和红鳍原鲐空间分布的影响不显著.黄丝草植被生境是牛山湖两种优势小型鱼类的重要保护生境,应加强对黄丝草等沉水植被的保护及恢复.  相似文献

通过幼苗萌发法和样方调查相结合的方法对三江平原不同演替恢复阶段的种子库特征及其与植被的关系进行了研究。将开垦湿地、不同演替恢复阶段湿地以及天然湿地不同土壤层次(0-5、5-10 cm和根茎)的种子库在两种水分条件下(湿润、淹水10 cm)进行萌发处理。结果表明: 随着演替恢复阶段的进行, 种子库的结构和规模逐渐扩大, 地表群落表现出由旱生物种占优势的群落逐渐演变成以小叶章(Calamagrostis angustifolia)占优势的湿生群落的演替趋势。恢复7年湿地、恢复14年湿地、天然湿地土壤种子库萌发物种数分别为24种、29种、39种, 植被物种数为21种、25种、14种。湿地类型、水分条件和土壤层次均显著影响种子库萌发的物种数及幼苗数(p < 0.01)。种子库具有明显的分层现象, 天然湿地0-5 cm土层种子库种子萌发密度是5-10 cm土层的4倍左右, 而恢复湿地仅1.3倍左右, 且土层间萌发物种相似性系数较低。湿润条件下的萌发物种数显著高于淹水条件, 且两种水分条件下萌发物种的生活型不同。由于恢复时间较短, 不同演替恢复阶段的种子库与植被相似性维持在30%以下。湿地中根茎分蘖出大量的湿地物种, 对于小叶章等优势物种的繁殖具有重要作用。研究表明, 在开垦湿地退耕后的次生演替阶段, 种子库能够保持大量的湿地物种, 通过对湿地种子库与植被的关系研究, 能够为三江平原湿地群落演替与湿地恢复提供策略指导。  相似文献

Difficulty in species identification of Sargassum (Sargassaceae, Fucales) is partly attributed to the high polymorphism among its individuals and populations. This study aimed at assessing morphological and genetic variations in two varieties, var. hemiphyllum J. Agardh and var. chinense J. Agardh, of Sargassum hemiphyllum (Turner) C. Agardh, a widely distributed species in the northwestern Pacific. We investigated 26 measurable, five numerical, and 33 categorical morphological parameters associated with different branching levels of specimens from each of six localities within its distribution range using cluster analysis (CA) and principal coordinate analysis (PCoA). Leaf size of the primary and secondary branching levels and the vesicle size of the secondary branches of the specimens examined were determined to be the most important morphological parameters that were significantly different among populations. Change in leaf and vesicle length of individuals among the six populations followed a latitudinal gradient, with smaller leaves and vesicles associated with northern populations and larger ones in the southern populations. The possible influence of the gradual change in sea surface temperatures (SSTs) along this gradient in the northwestern Pacific on leaf and vesicle morphologies of this species was suggested. PCR‐RFLP analysis of the RUBISCO spacer in the chloroplast genome revealed two distinct and highly homogenous clades, a China clade and a Japan‐Korea clade, which corresponded to var. chinense and var. hemiphyllum, respectively. The formation of refugia along the “Paleo‐coast” in the East China Sea during glacial periods is suggested to have led to the vicariance of ancestral populations of S. hemiphyllum and thus to have promoted genetic differentiation. The massive freshwater outflow of the Yellow and Yangtze rivers may continue to act as a barrier, prolonging the allopatric distribution of the two varieties.  相似文献

Abstract The population of emperor geese (Chen canagica) in western Alaska, USA, declined by >50% from the 1960s to the mid-1980s and has increased only slightly since. Rates of population increase among arctic geese are especially sensitive to changes in adult survival. Improving adult survival in seasons or geographic areas where survival is low may be the best means of increasing the emperor goose population. We monitored fates of 133 adult female emperor geese that were radiomarked with surgically implanted very high frequency or satellite radiotransmitters from 1999 to 2004 to assess whether monthly survival varied among years, seasons, or geographic areas. Because of uncertainties in determining whether a bird had died based on the radio signal, we analyzed 2 versions of the data. One version used conservative criteria to identify which birds had died based on radio signals and the other used more liberal criteria. In the conservative version of the data we detected 12 mortalities of emperor geese, whereas in the liberal interpretation there were 18 mortalities. In both versions, the models with greatest support indicated that monthly survival varied seasonally and that compared to most seasons estimated monthly survival was lower (φ = 0.95–0.98) in May and August when emperor geese were mainly on the Yukon-Kuskokwim Delta. From 44% to 47% of annual mortality occurred in those months. Estimated monthly survival was higher (φ = 0.98–1.0) from September through March when emperor geese were at autumn staging or wintering areas and in June and July when birds were nesting, rearing broods, or molting. Estimated annual survival was 0.85 (95% CI = 0.77–0.92) in the best-supported model when we used conservative criteria to identify mortalities and 0.79 (95% CI = 0.74–0.85) under the best model using liberal mortality criteria. Lower survival in August and May corresponded to periods when subsistence harvest of emperor geese was likely highest. Managers may be able to most effectively influence population growth rate of emperor geese by reducing subsistence harvest on the Yukon-Kuskokwim Delta in May and August.  相似文献
