Reptilian transferrins: evolution of disulphide bridges and conservation of iron-binding center 总被引:1,自引:0,他引:1
Transferrins, found in invertebrates and vertebrates, form a physiologically important family of proteins playing a major role in iron acquisition and transport, defense against microbial pathogens, growth and differentiation. These proteins are bilobal in structure and each lobe is composed of two domains divided by a cleft harboring an iron atom. Vertebrate transferrins comprise of serotransferrins, lactoferrins and ovotransferrins. In mammals serotransferrins transport iron in physiological fluids and deliver it to cells, while lactoferrins scavenge iron, limiting its availability to invading microbes. In oviparous vertebrates there is only one transferrin gene, expressed either in the liver to be delivered to physiological fluids as serotransferrin, or in the oviduct with a final localization in egg white as ovotransferrin. Being products of one gene sero- and ovotransferrin are identical at the amino-acid sequence level but with different, cell specific glycosylation patterns. Our knowledge of the mechanisms of transferrin iron binding and release is based on sequence and structural data obtained for human serotransferrin and hen and duck ovotransferrins. No sequence information about other ovotransferrins was available until our recent publication of turkey, ostrich, and red-eared turtle (TtrF) ovotransferrin mRNA sequences [Ciuraszkiewicz, J., Olczak, M., Watorek, W., 2006. Isolation, cloning and sequencing of transferrins from red-eared turtle, African ostrich and turkey. Comp. Biochem. Physiol. 143 B, 301-310]. In the present paper, ten new reptilian mRNA transferrin sequences obtained from the Nile crocodile (NtrF), bearded dragon (BtrF), Cuban brown anole (AtrF), veiled and Mediterranean chameleons (VtrF and KtrF), sand lizard (StrF), leopard gecko (LtrF), Burmese python (PtrF), African house snake (HtrF), and grass snake (GtrF) are presented and analyzed. Nile crocodile and red-eared turtle transferrins have a disulphide bridge pattern identical to known bird homologues. A partially different disulphide bridge pattern was found in the Squamata (snakes and lizards). The possibility of a unique interdomain disulphide bridge was predicted for LtrF. Differences were found in iron-binding centers from those of previously known transferrins. Substitutions were found in the iron-chelating residues of StrF and TtrF and in the synergistic anion-binding residues of NtrF. In snakes, the transferrin (PtrF, HtrF and GtrF) N-lobe "dilysine trigger" occurring in all other known transferrins was not found, which indicates a different mechanism of iron release. 相似文献

高原湖泊流域是高原地区人类活动的重要载体,兼具高生态价值和高脆弱性的特点。随着高原湖泊流域城市化和工业化发展加速,湖泊面积萎缩,污染加剧,流域生态环境受损严重,引发了一系列生态环境问题,如水土流失、水污染、湿地退化、生境质量下降等。亟需开展生态修复以平衡经济发展与生态环境保护之间关系,而基于整体保护与系统治理思维诊断并修复生态修复优先区,是科学有序推进国土空间生态保护与修复的重要抓手。基于此,研究以高原湖泊流域典型代表滇池流域为例,利用人类足迹和景观生态风险模型定量评估生态系统所受负向干扰,以最小累积阻力模型和电路理论构建流域生态网络;提取生态网络受负向干扰较高的关键区域为生态修复优先区并提出针对性修复措施。研究表明:(1)滇池流域人类干扰和生态风险整体较高,人类干扰整体呈核心—边缘递减的圈层式分布,中高生态风险占据了绝大部分区域。人类交通网络大幅扩展了人类干扰和生态风险的强度和深度;(2)区域生态网络呈典型湖泊生态网络特点,38条生态廊道呈放射状或环状分布,连通湖区、山区两大生态空间内共23块生态源地,保障区域生态安全;(3)研究共提取生态源地修复优先区73.83km2 相似文献

Temperature, dissolved oxygen and dissolved methane profiles were measured during autumn and summer, in a shallow floodplain lake in south-eastern Australia to determine the effects of water-column stability on methane and oxygen dynamics. The water column was well mixed in autumn. Strong thermal stratification developed in the late afternoon in summer, with top-to-bottom temperature differences of up to 6 °C. Methane concentrations in surface waters varied over a daily cycle by an 18-fold range in summer, but only by a 2-fold range in autumn. The implication of short-term temporal variation is that static chambers deployed on the water surface for short times (less than a day) in summer will significantly underestimate the diffusive component of methane emissions across the water–atmosphere interface. There was a marked diel variation in dissolved oxygen concentrations in summer, with the highest oxygen values (commonly 5–8 mg l–1) occurring in the surface waters in late afternoon; the bottom waters were then devoid of oxygen (< 0.2 mg l–1). Because of high respiratory demands, even the surface water layers could be nearly anoxic by morning in summer. The concentration of dissolved oxygen in the surface waters was always less than the equilibrium value. When the water column became thermally stratified in summer, the dissolved oxygen and methane maxima were spatially separated, and planktonic methanotrophy would be limited to a moving zone, at variable depth, in the water column. In summer the whole-wetland rates of oxygen production and respiration, calculated from long-term (5 h) shifts in dissolved oxygen concentrations over a diel period, were approximately 6–10 and 3–6 mmol m–3 h–1, respectively. Until now, additions of lime have been used to restore the buffering capacity of acidified lakes, but an alternative method
which is more effective in the treatment of lakes with organogenic sediments has recently been applied in a full-scale experiment.
The method, called CONTRACID, is based on the cation exchange properties of lake sediment. A sodium carbonate (soda ash) solution
is injected into the sediment (by a harrow), so that the sediment becomes sodium stocked. A reverse exchange occurs during
subsequent acidification. Liming has a limited effect on humic lakes, since Ca-humates have a reduced reverse exchange ability
and also the lime, which remains undissolved, is rendered inactive.
Ionic exchange processes and nutrient transport were studied in water/sediment cores andin situ enclosures after additions of soda ash-, lye- and lime solutions with subsequent re-acidification. Sodium carbonate additions
in laboratory systems resulted in a sorption to the sediment of 42–62% of the added sodium ions (5 eq m−2) and a release of 14–78 mg Pm−2 sediment. Similar results were obtained in the enclosures where phosphorus release stimulated algal growth. Sediment pH,
elevated by the sodium base addition, was lowered by re-acidification. Limed systems released no phosphorus and only about
25% of the added lime remained active for future neutralization. With the injection of the sodium carbonate solution into
the sediment, only about 12% of the added sodium was recovered in lake water by spring circulation. Lake water alkalinity
was then 0.12 meq l−1 and pH 6.7. Total phosphorus had been raised by 0.007 mg P l−1 causing an increase in phytoplankton biomass.
Observations indicate that manipulations of acidic lake sediment according to the CONTRACID method create a long-lasting neutralizing
capacity and a biological stimulation (through phosphorus release), which makes the method an attractive alternative to frequent

1. The distribution of zooplankton in shallow lakes is negatively related to macrophyte density. However, the abundance of their food along density gradients of macrophytes is unknown. A common but untested assumption is that food quantity and quality for pelagic zooplankton is poor in the littoral zone owing to the deleterious influence of macrophytes on phytoplankton. 2. We tested this assumption with a combination of a field survey and laboratory experiments. We collected seston samples from the littoral and pelagic zones of four shallow temperate lakes and related food quantity (phytoplankton biovolume) and quality to macrophyte abundance (per cent volume infested). Seston food quality was assessed in three ways: N/C and P/C ratios, polyunsaturated fatty acid content and phytoplankton community composition. In the laboratory, we measured the growth and reproduction of Daphnia pulicaria on diets consisting of seston from the littoral and pelagic zones in one lake. 3. In our four study lakes, food quantity was not significantly influenced by macrophyte abundance, and food quality was generally high. Laboratory experiments showed increased juvenile growth, but no significant change in D. pulicaria reproduction, when feeding on littoral resources compared to pelagic resources. 4. Our results suggest that there is no nutritional cost to pelagic zooplankton inhabiting the littoral zone. 基于巢湖水体2002~2007年水质监测资料,对叶绿素a浓度的分布、动态及与TN、TP的关系进行了统计分析。巢湖叶绿素a浓度与TN、TP的浓度分布存在明显的空间差异,西半湖叶绿素a浓度全年高于20μg/L,TN为1.94~3.84mg/L,TP为0.20~0.42mg/L;东半湖叶绿素a浓度全年小于5.5μg/L,TN为0.95~1.83mg/L,TP为0.08~0.14mg/L。在东半湖,叶绿素a含量与TN呈不明显的正线性关系,当TP浓度较低时,叶绿素a随TP的增加小幅上升,但是当TP>0.15mg/L时,叶绿素a随TP的增加而明显上升;在西半湖,当水体TN<5.8mg/L或者TP<2.0mg/L时,叶绿素a含量与TN、TP关系为正线性关系,当TN在5.8~9.4mg/L或者TP介于0.2~0.3mg/L间时,叶绿素a含量与TN、TP关系为不显著的负线性关系,当TP浓度>0.3mg/L时,叶绿素a含量与TP关系又为正线性关系。西半湖叶绿素a浓度的变化可能是藻类生物活动与沉积物及水体中营养盐的相互作用结果。在治理巢湖富营养化时,应优先控制西半湖的磷元素。 相似文献

The hypothesis that diatom carbon (C) produced during the spring peak supported spring zooplankton production and, ultimately, the growth of Coregonus lavaretus early larval stages from March to May 2006 in Lake Annecy, France, was tested using gut content analyses and fatty acid biomarkers. Gut content results showed that C. lavaretus larvae from stages 1 to 4 preferentially fed on copepods with Daphnia sp. only a minor proportion of larval diet. The levels of diatom-marker fatty acids (C16:1n-7 and C20:5n-3) were high in Daphnia sp., but lower in both copepods and C. lavaretus larvae from stages 0 to 4. These results indicated that the spring diatom biomass was actually grazed by Daphnia sp., but, contrary to what was expected, the spring bloom was not the only C source supporting copepods secondary production and, consequently, the growth of C. lavaretus early larval stages. In contrast, levels of terrestrial fatty acid marker (C24:0) were low in Daphnia sp. but high in copepods and C. lavaretus larvae, indicating a significant contribution of terrestrial carbon to copepods and, ultimately, to the growth of C. lavaretus early larval stages. 相似文献

1. In view of the paucity of data on the response of warm shallow lakes to reductions in nutrient loading, this paper presents a long‐term limnological data set to document changes in the food‐web of a shallow Mediterranean lake (Lake Albufera, Valencia, Spain) that has experienced reductions in phosphorus (P) (77%) and nitrogen (N) (24%) loading following sewage diversion. 2. Nine years after sewage diversion, P concentration in the lake was reduced by 30% but remained high (TP = 0.34 mg L?1), although the mean water retention time in the lake was only 0.1 years. Nitrate concentrations did not significantly change, probably because the lake continued to receive untreated effluents from ricefields. 3. Chlorophyll a concentration was reduced by half (annual mean of 180 μg L?1). Cyanobacteria abundance remained high but its composition changed towards smaller species, both filamentous and chroococcal forms. 4. Cladocera abundance increased and reached peaks twice a year (December to March and July to September). After nutrient reduction, short‐term clear‐water phases (up to 5 weeks) occurred during February to March in several years, concomitant with annual flushing of the lake and lower fish densities. The abundance of Cladocera in winter contrasted with the spring peaks observed in northern restored shallow lakes. The zooplankton to phytoplankton biomass ratio remained lower than in northern temperate shallow lakes, probably because of fish predation on zooplankton. 5. Improvement of the water quality of Lake Albufera remained insufficient to counteract littoral reed regression or improve underwater light allowing submerged plants re‐colonise the lake. 6. Sewage diversion from Lake Albufera impacted the food web through the plankton, but higher trophic levels, such as fish and waterfowl, were affected to a lesser degree. Although the fish species present in the lake are mainly omnivorous, long‐term data on commercial fish captures indicated that fish communities changed in response to nutrient level and trophic structure as has been observed in restored shallow lakes at northern latitudes. 7. Phosphorus concentrations produced similar phytoplankton biomass in Lake Albufera as in more northern shallow lakes with abundant planktivorous fish and small zooplankton. However, in Lake Albufera, high average concentrations were maintained throughout the year. Overall, results suggest that nutrient control may be a greater priority in eutrophicated warm shallow lakes than in similar lakes at higher latitudes. 相似文献
