Summary Negative-staining of crude cytoplasmic extracts from cells of the green algaErnodesmis verticillata reveals the presence of numerous microfilaments. Rabbit skeletal muscle heavy-meromyosin binds to the microfilaments (in the absence of ATP) in typical arrowhead arrays. These results demonstrate that actin is present in this alga and it is suggested that actin may be involved in cytoplasmic contractions effecting wound healing, since cytoplasmic streaming does not occur in this organism. 相似文献

The ecological significance of rapid wound-induced changes in plants: insect grazing and plant competition 总被引:1,自引:0,他引:1
Summary This paper investigates the hypothesis that a rapidly induced phytochemical response to grazing damage, such as that seen in tomato, serves to deflect insect herbivores away from leaves soon after damaging them (the grazing dispersal hypothesis). As a result, grazing damage is more dispersed than it otherwise would be, and young leaves, which may be of particular importance to a plant in competition for light, are not damaged excessively. In the first experiment, artificial removal of c. 15% of leaf area led to a significant reduction in plant performance compared with undamaged controls, but only when the plants were grown together in competition for light. The second experiment demonstrated that the distribution of grazing damage within the plant was an important factor in the outcome of competition; in those plants in which grazing was applied to the lower leaves there was no effect of damage upon performance compared with undamaged controls, whereas grazing to the upper leaves significantly reduced plant performance. A third experiment provided some insight into how this interaction between damage and competition comes about. It was shown that damage to leaves led to a rapid drop in the rate of extension growth of the main shoot, especially when the upper leaves were damaged, and normal rates of growth were not resumed for at least 3 days. It is argued that in a rapidly growing canopy, such an effect may mean that a damaged plant loses its position in the height hierarchy. The final experiment showed that previous damage to plants can affect the distribution of subsequent grazing by larvae of Spodoptera littoralis, apparently through a wound-induced reduction in leaf palatability. Plants which had been artificially damaged 48 h previously were grazed significantly less than controls, and the avoidance effect was greatest in the young leaves. These results are consistent with the grazing dispersal hypothesis, and suggest that rapid wound-induced responses may be of greatest significance in species characteristic of fertile environments where competition for light is particularly intense. 相似文献

Diabetes diminishes bone healing and ossification. Reduced bone formation in intramembranous ossification is known, yet the mechanism(s) behind impaired intramembranous bone healing are unclear. Here we report the formation of a fibrotic matrix during healing of intramembranous calvarial bone defects that appears to exclude new bone growth. Our histological analyses of 7-day and 14-day calvaria bone healing tissue in chemically-induced diabetic mice and non-diabetic mice showed the accumulation of a non-mineralized fibrotic matrix, likely as a consequence of unresolved hematomas under diabetic conditions. Elevated mRNA and enzyme activity levels of lysyl oxidase on day 7 in diabetic bone healing tissues also supports that the formation of a fibrotic matrix occurs in these tissues. Based on these findings, we propose that elevated fibroblast proliferation and formation of a non-mineralized fibrotic extracellular matrix in diabetes contributes to deficient intramembranous bone healing in diabetes. A greater understanding of this process has relevance to managing dental procedures in diabetics in which successful outcomes depend on intramembranous bone formation. 相似文献

The use of active ingredients in wound management have evolved alongside the pharmaceutical agents and dressings used to deliver them. However, the development of gauzes, dressings with specific properties, still remains a challenge for several medical applications. A new methodology for the controlled release of active components for the healing of burn wounds is proposed herein. Cotton and non‐woven bandages have been cationised to promote the attachment of protein microspheres. The active agents, piroxicam and vegetable oil, were entrapped into the microspheres using ultrasound energy. Active agents were released from the microspheres by a change in pH. Wound healing was assessed through the use of standardised burn wounds induced by a cautery in human full‐thickness skin equivalents (EpidermFT). The best re‐epithelialisation and fastest wound closure was observed in wounds treated with proteinaceous microspheres attached to gauzes, after six days of healing, in comparison with commercial collagen dressing and other controls. Furthermore, the ability of these materials to reduce the inflammation process, together with healing improvement, makes these biomaterials suitable for wound‐dressing applications. 相似文献

Nitric oxide (NO) production and free amino acid fluxes at the wound side during the first 3 days following cutaneous wound were investigated. Experiments were performed on Albino Oxford rats (n = 18) underwent cutaneous implantation of polyvinyl sponges. Intact animals (n = 6) were controls. Nitrites, nitrates, free amino acids and urea were measured both in plasma and wound fluids. Inducible nitric oxide synthase (iNOS) gene expressions at wound site were analyzed, too. The highest levels of both iNOS gene expression and its activity (increased wound fluid citrulline and nitrites) were at the first day. Wound fluid nitrates were significantly above plasma levels throughout the whole period, while molar nitrate to nitrite ratio steadily increased. It was associated with gradual increase of both ornithine and urea as well as steadily decreases of arginine and increases of phenylalanine at the wound site. Gradual decrease in glycine to branched-chain molar ratio was observed both in plasma and wound fluids. In conclusion, an early locally induced alterations in Arg metabolism, due to increased NO formation followed by increased arginase activity, produces relative lack of Arg at the wound site and disturbs nutritional status of the whole body almost within early healing period following cutaneous wound in rats. It is likely that NO autoxidation at the wound side is influenced by substrate availability. 相似文献

Gap junctions and their connexin components are indispensable in mediating the cellular coordination required for tissue and organ homeostasis. The critical nature of their existence mandates a connection to disease while at the same time offering therapeutic potential. Therapeutic intervention may be offered through the pharmacological and molecular disruption of the pathways involved in connexin biosynthesis, gap junction assembly, stabilization, or degradation. Chemical inhibitors aimed at closing connexin channels, peptide mimetics corresponding to short connexin sequences, and gene therapy approaches have been incredibly useful molecular tools in deciphering the complexities associated with connexin biology. Recently, therapeutic potential in targeting connexins has evolved from basic research in cell-based models to clinical opportunity in the form of human trials. Clinical promise is particularly evident with regards to targeting connexin43 in the context of wound healing. A strong immune defense is vital for honey bee health and colony survival. This defense can be weakened by environmental factors that may render honey bees more vulnerable to parasites and pathogens. Honey bees are frequently exposed to neonicotinoid pesticides, which are being discussed as one of the stress factors that may lead to colony failure. We investigated the sublethal effects of the neonicotinoids thiacloprid, imidacloprid, and clothianidin on individual immunity, by studying three major aspects of immunocompetence in worker bees: total hemocyte number, encapsulation response, and antimicrobial activity of the hemolymph. In laboratory experiments, we found a strong impact of all three neonicotinoids. Thiacloprid (24 h oral exposure, 200 μg/l or 2000 μg/l) and imidacloprid (1 μg/l or 10 μg/l) reduced hemocyte density, encapsulation response, and antimicrobial activity even at field realistic concentrations. Clothianidin had an effect on these immune parameters only at higher than field realistic concentrations (50–200 μg/l). These results suggest that neonicotinoids affect the individual immunocompetence of honey bees, possibly leading to an impaired disease resistance capacity. 相似文献
