Brindley P. J. and Dobson C. 1982. Nematospiroides dubius in mice selected for liability to infection: modification of parasite biology through host selection. International Journal for Parasitology12: 573–578. Mice selected as liable (L) and refractory (R) over ten generations voided significantly more and less Nematospiroides dubius eggs compared with randomly mated (Rd) mice after primary infections with 100 larvae. There was little difference between the number of parasite eggs voided g?1 faeces (epg) by individual mice on day 14 compared with day 15 after infection.However there was a significant diurnal variation in the egg values for individual mice but the mean differences observed between the R, Rd and L mice were maintained over a 24 h period. There was a strong correlation between both the total number and the number of female worms, surviving 21 days after infection, and the mean epg 14 and 15 days after infection. Female N. dubius produced more eggs in L mice and fewer eggs in R mice compared with worms in Rd mice. Similarly, worms grew longer in L mice and were shorter in R mice compared with parasites in Rd mice.  相似文献

Summary Dominant genes are the main cause of the heterosis induced by fasciated mutants of different lines of Pisum sativum. Most of these cases were originally interpreted by different authors as examples of monogenic overdominance. Several not-closely-linked genes appear to have mutated simultaneously in most of the fasciated lines. Although fasciation itself is recessive, other mutant characters, such as lateness, increased stem length (number and length of internodes) and, in part, seed production per plant, show dominant inheritance. The latter two features are, however, to a considerable extent suppressed in the fasciated lines by unfavourable gene-interactions (epistasis). Crossing these lines with non-fasciated ones shows that the epistatic genes are recessive and the dominant genes are then no longer hindered in their action. By eliminating the epistatic genes from the genomes of fasciated lines by recombination, the heterosis phenomenon has been fixed on six independent occasions for different lines. The fasciata genes themselves were found to be the most probable cause of these cases of recessive epistasis. The question whether different kinds of fasciation affect heterosis differently is examined. Recessive epistasis and dominance explain most of the quantitative distinctions between the different hybrids. In addition, one example of heterosis between non-fasciated lines is given and the possible meaning of the overall results for plant breeding and population genetics is mentioned.  相似文献

The incorporation of radioactive N-acetylglucosamine into murein and lipopolysaccharide of synchronized cells of Escherichia coli K 12 was followed over 100 min in the presence of antibiotics. At 20 min intervals cell walls were prepared. Lipopolysaccharide and murein sacculi were isolated and the radioactivity was quantified in both polymers. Labelled, newly synthesized murein was characterized according to murein subunits linked to lipoprotein, and the degree of crosslinkage. Furthermore, murein subunits containing anhydromuramic acid were determined, permitting the calculation of the average glycan chain length. The results indicated that penicillin G at 30 g/ml stimulated the incorporation of new murein subunits into sacculi followed by a sudden increase in lipopolysaccharide incorporation into the outer membrane. The degree of crosslinkage in murein synthesized in the presence of 30 g/ml penicillin G was higher than in the control, and almost twice as high as in murein synthesized in the presence of 20 g/ml nalidixic acid. Both antibiotics inhibited cell division at the concentrations indicated. Murein synthesized in the presence of 2 g/ml mecillinam also showed higher crosslinkage. However, about twice as much anhydromuramic acid-containing subunits were observed as in the control.   1. A model is presented to estimate the mosquito density and radius (r) of the attraction range of man as the bait.
  2. Man walked to a spot in the woods and sampled mosquitoes flying to him with a net continuously. The arrival pattern was constant in each mosquito species, and in all species, the number of mosquitoes sampled per five min became constant after ten minutes sampling.
  3. The mosquitoes gathered to man were composed of D: those waiting alight, I: those intruding the attraction range and F: those following man to the spot. They were estimated by means of continuous sampling sets and densities of D and I were calculated.
  4. r s of female and male of A. albopictus and A. japonicus were 4.0, 5.6 and 9.0 m respectively. In these species, most individuals were D, although, in T. bambusa all individuals were I, flying to search for hosts.

We obtained antisera to each of the five subunits (α, β, γ, δ, and ?) of the F1 portion of the proton-translocating ATPase from Escherichia coli (ECF1). No cross-reaction between the antiserum to a given subunit and any of the other four subunits was observed by Ouchterlony immunodiffusion. The α antiserum reacted only with the denatured α chain. Antibodies to either subunit β or subunit γ inhibited the ATPase activity of the enzyme. The ATPase activity of the holoenzyme in the everted membrane vesicles was just as sensitive as purified ECF1 to inhibition by the anti-β or anti-γ serum. A prolonged digestion of ECF1 with trypsin removed intact γ from ECF1, but did not alter the sensitivity of the ATPase to inhibition by the anti-γ serum. Proteolytic fragments were isolated from the trypsinized enzyme. They gave an immunoprecipitation band with the anti-γ serum, but none of the other subunit antisera. The antiδ serum detached ECF1 from everted membrane vesicles and completely blocked both the ATP- and respiration-dependent pyridine nucleotide transhydrogenase, an energylinked membrane function. The δ antiserum had no effect on the ATPase activity of the ECF1. The e antiserum stimulated the ATPase activity of purified ECF1 as shown previously (P. P. Laget and J. B. Smith, Arch. Biochem. Biophys.197, 83, 1979), but strongly inhibited the holoenzyme in membrane vesicles. The α antiserum completely blocked the ATP-driven transhydrogenase. The same antiserum maximally inhibited the respiratory chain-driven reaction by only 35%. These observations indicate that the antiserum selectively affected energy transduction mediated by the ATPase. The protonmotive force generated by substrate oxidation was probably not dissipated by the ? antiserum. Adsorbing the δ or ? antiserum with everted membrane vesicles selectively removed those antibodies that reacted with membrane-bound ATPase. The adsorbed sera still reacted strongly with purified ECF1, and prevented it from restoring ATP-dependent proton translocation in ECF1-depleted vesicles. Therefore, it appears that more of the δ and the ? subunit is exposed in the purified ECF1 molecule than in the membrane-bound enzyme.  相似文献

Primary cilia are antenna‐like organelles required for signalling transduction. How cilia structure is mechanistically maintained at steady‐state to promote signalling is largely unknown. Here, we define that mammalian primary cilia axonemes are formed by proximal segment (PS) and distal segment (DS) delineated by tubulin polyglutamylation‐rich and ‐poor regions, respectively. The analysis of proximal/distal segmentation indicated that perturbations leading to cilia over‐elongation influenced PS or DS length with a different impact on cilia behaviour. We identified septins as novel repressors of DS growth. We show that septins control the localisation of MKS3 and CEP290 required for a functional transition zone (TZ), and the cilia tip accumulation of the microtubule‐capping kinesin KIF7, a cilia‐growth inhibitor. Live‐cell imaging and analysis of sonic‐hedgehog (SHH) signalling activation established that DS over‐extension increased cilia ectocytosis events and decreased SHH activation. Our data underlines the importance of understanding cilia segmentation for length control and cilia‐dependent signalling.  相似文献

Fine root distribution, quantities, dynamics and composition were studied in a sessile oak coppice stand in the French Ardennes on an acidic soil (< pH-H2O 4.5), one to five years after lime or gypsum applications. Fine root biomass and length increased and specific root length decreased after lime or gypsum treatments. The treatment responses were strongest four to five years after the applications, but the tendencies after one year were similar. The effects were pronounced in the top 15 cm but also at 30–45 cm four to five years after liming. The latter effect suggests an indirect positive feedback from the aerial parts of the trees into the deeper soil layers. Sequential sampling for two years revealed large differences in total fine root length between the years, and also indicated that fine root turnover was lower after liming or gypsum applications than in the control. This seemed to be related to a lower fine root mortality and higher longevity rather than to increased fine root production. The improved nutrient status of the fine roots corroborates this and coincides with improved foliar nutrition and tree growth. Moderate doses of lime and gypsum appeared effective in enhancing root system uptake function, resulting in increased above ground growth.  相似文献

he segregation ratio of RFLP markers in an F2 population from indica "Zhaiyeqing 8” and japonica "Jingxi 17' of rice (Oryza sativa L., 2n= 24) was studied using 54 RFLP markers distributed on 12 chromosomes. Distorted segregation was found in 25.9% of the marker tested, which was indicated by significant deviation from the expected Mendelian segregation ratio ( I: 2: 1) at 5% or 1% level. Among the three RFLP genotypes of the F2 population “Zhaiyeqing” 8 genotype was significantly more than the expected, and its gene frequency was up to 52.1 %. Three positions for distorted segregation were found on chromosome 3 (RG227-RG369), 7 (RG678-RG511-RG528) and 12 (RG463-RG323). These positions could be related to gametophyte loci responsible for the distortion.  相似文献

An understanding of the level, structure and origin of genetic variation within and among populations of tropical trees is essential for devising optimum management strategies for their sustainable utilization and conservation. Here, amplified fragment length polymorphism (AFLP) analysis was used to partition genetic variation within and among nine populations of the predominantly riverine tree, Calycophyllum spruceanum , sampled across a wide geographical range along river tributaries of the Peruvian Amazon Basin. Analysis of molecular variance ( AMOVA ) employed 65 AFLP markers and revealed most variation among individuals within populations (91%), although variation among populations was highly significant ( P < 0.001). Calculation of genetic distances and nested AMOVA indicated a degree of structuring among populations based on geographical proximity, although clustering did not depend on geographical distance alone. No firm evidence was obtained for unidirectional seed dispersal by water playing an important role in determining genetic structure over the geographical range sampled. Implications of data for optimising genetic management of the species are discussed and areas for further study identified.  相似文献
