Production of clavulanic acid (CA) by Streptomyces clavuligerus ATCC 27064 in shake-flask culture (28 °C, 250 rev min–1) was evaluated, with media containing different types and concentrations of edible vegetable oil. Firstly, four media based on those reported in the literature were examined. The medium containing soybean oil and starch as carbon and energy source gave the best production results. This medium, with the starch replaced by glycerol, and with various soybean oil concentrations (16, 23 and 30 g l–1) was utilized to further investigate CA production. Medium containing 23 g l–1 led to the highest CA productivity (722 mg l–1 in 120 h) and that one containing 30 g l–1 gave the highest CA titre (753 mg l–1 in 130 h). Also, substitution of corn and sunflower edible oils furnished similarly good results in terms of CA titre and productivity. It can be concluded that easily available vegetable oil is a very promising substrate for CA production, since it is converted slowly to glycerol and fatty acids, which are the main carbon and energy source for the microorganism. 相似文献

Anaerobic biodegradation of vegetable oil in freshwater sediments is strongly inhibited by high concentrations of oil, but the presence of ferric hydroxide relieves the inhibition. The effect of ferric hydroxide is not due to physical or chemical interactions with long-chain fatty acids (LCFAs) that are produced as intermediates during metabolism of vegetable-oil triglycerides. The anaerobic biodegradation of canola oil and mixtures of acetic and oleic acids, two important intermediates of vegetable-oil metabolism, were investigated using sediments enriched on canola oil under methanogenic and iron-reducing conditions to determine whether the effect of ferric hydroxide has a biological basis. Sediments enriched under both conditions rapidly and completely converted canola oil to methane when the initial oil concentration was relatively low (1.9 g oil/kg sediments), but the biotransformation was strongly inhibited in sediments enriched under methanogenic conditions when the initial concentration was 19 g/kg (<30% of the oil-derived electron equivalents were transferred to methane in a 420-day incubation period). Sediments enriched under iron-reducing conditions, however, completely transformed canola oil to methane in about 250 days at this initial oil concentration. The anaerobic biotransformation of mixtures of acetate and oleic acid followed a similar pattern: the rate and extent of conversion of these electron-donor substrates to methane was always higher in sediments enriched under iron-reducing than under methanogenic conditions. These results suggest that enrichment on canola oil in the presence of ferric hydroxide selects a microbial community that is less sensitive to inhibition by LCFAs than the community that develops during enrichment under methanogenic conditions. 相似文献

During the last few years, geographical information systems (GIS) have spread as powerful tools for landscape analysis. The main purpose of this work was to use GIS to display an ecological network made up of core areas, key areas and ecological corridors. As an example of the application of this method we refer to the population of deer (Cervus elaphus) and roe deer (Capreolus capreolus) in an alpine area in northwestern Italy. The method provided an overall view of the ecological network of the area, highlighting how linear infrastructures can affect animal populations and consequently, their survival probability. 相似文献

A fungal strain capable of utilizing chlorpyrifos as sole carbon and energy sources was isolated from soil by enrichment cultivation approach. The half-lives of degradation (DT50) for chlorpyrifos at concentrations of 1, 10, and 100 mg l−1 by the fungal strain DSP in mineral salt medium were measured to be 2.03, 2.93, and 3.49 days, respectively. Two cell-free extracts [E (1:10) and E (1:20)] from the fungal strain DSP in bran–glucose medium were prepared and used to enhance chlorpyrifos degradation on vegetables. Compared with the controls, the DT50 of chlorpyrifos were reduced by 70.3%, 65.6%, 80.6%, 80.6%, and 86.1%, and by 53.8%, 43.2%, 66.0%, 54.3%, and 67.7% on E (1:20) and E (1:10) treated pakchoi, water spinach, Malabar spinach, haricot beans, and pepper, respectively. The 7-day residual values (R
7) of chlorpyrifos on E (1:10) treated vegetables were all lower than the corresponding maximum residue levels of European Union (EU MRLs), except that the R

Performance of wheat crops with different chromosome ploidy: root-sourced signals, drought tolerance, and yield performance 总被引:1,自引:0,他引:1
Pot-culture experiments were carried out to estimate the role of non-hydraulic root signals (nHRS) and the relation of these signals to drought tolerance and grain yield formation under drought stress in six wheat varieties. These were two modern hexaploid wheat (Triticum aestivum L., AABBDD) Plateau602 and Longchun8139-2, two diploid wheat (Triticum monococcum L., AB) MO1 and MO4, and two tetraploid wheat (Triticum dicoccum Schuebl L., AABB) DM22 and DM31. In the two diploid relatives, the nHRS was switched on and off at a soil water content (SWC) of approximately 53–45% field water capacity (FWC). In contrast, in the modern hexaploid varieties, Longchun8139-2 and Plateau602 the nHRS occurred between a SWC of about 71 and 35% FWC, a much wider soil moisture range. The two tetraploid relatives, DM22 and DM31, were generally intermediate. The nHRS threshold range in SWC also narrowed as all six varieties went through successive developmental stages from shooting to grain filling. The two hexaploid wheat varieties had the longest duration of survival after the water supply ceased, and the best yield stability under drought stress, similar to with tetraploid wheat varieties; the diploid wheat varieties were least robust. These two parameters were both significantly correlated with the nHRS soil moisture threshold range (r=0.9456** and 0.8608*, respectively). Based on these patterns, we propose a ‘triple Z’ model to describe the features of non-hydraulic stomatal sensitivity versus soil drought in wheat growth. 相似文献

Propan-2-ol was used as an acyl acceptor for immobilized lipase-catalyzed preparation of biodiesel. The optimum conditions
for transesterification of crude jatropha (Jatropha curcas), karanj (Pongamia pinnata) and sunflower (Helianthus annuus) oils were 10% Novozym-435 (immobilized Candida antarctica lipase B) based on oil weight, alcohol to oil molar ratio of 4:1 at 50 °C for 8 h. propan-2-ol were 92.8, 91.7 and 93.4% from crude jatropha, karanj and sunflower oils, respectively. Reusability of the lipase
was maintained over 12 repeated cycles with propan-2-ol while it reached to zero by 7th cycle when methanol was used as an acyl acceptor, under standard reaction conditions.

An active Trypanosoma cruzi transmission cycle maintained by wild rodents in the Andean valleys of Cochabamba Bolivia is described. Wild and domestic Triatoma infestans with 60% infection with T. cruzi were found and was evidenced in 47.5% (rodents) and 26.7% (marsupial) by parasitological and/or serologycal methods. Phyllotis ocilae and the marsupial species Thylamys elegans, are the most important reservoirs followed by Bolomys lactens and Akodon boliviensis. In spite of both genotypes (TCI and TCII) being prevalent in Bolivia, in our study area only T. cruzi I is being transmitted. Our data suggest that wild T. infestans and wild small mammals play an important role in the maintenance of the transmission cycle of T. cruzi. Furthermore, the finding of high prevalence of T. cruzi infection in wild T. infestans point to the risk of the dispersion of Chagas' disease. 相似文献

Wild Type (WT) and transgenic tobacco plants expressing isopentenyltransferase (IPT), a gene encoding the enzyme regulating the rate-limiting step in cytokinins (CKs) synthesis, were grown under limited nitrogen (N) conditions. We analyzed nitrogen forms, nitrogen metabolism related-enzymes, amino acids and photorespiration related-enzymes in WT and PSARK∷IPT tobacco plants. Our results indicate that the WT plants subjected to N deficiency displayed reduced nitrate (NO3−) assimilation. However, an increase in the production of ammonium (NH4+), by the degradation of proteins and photorespiration led to an increase in the glutamine synthetase/glutamate synthase (GS/GOGAT) cycle in WT plants. In these plants, the amounts of amino acids decreased with N deficiency, although the relative amounts of glutamate and glutamine increased with N deficiency. Although the transgenic plants expressing PSARK∷IPT and growing under suboptimal N conditions displayed a significant decline in the N forms in the leaf, they maintained the GS/GOGAT cycle at control levels. Our results suggest that, under N deficiency, CKs prevented the generation and assimilation of NH4+ by increasing such processes as photorespiration, protein degradation, the GS/GOGAT cycle, and the formation of glutamine. 相似文献

The predator Tupiocoris cucurbitaceus is frequently found attacking Trialeurodes vaporariorum in greenhouses without pesticide applications in Argentina. The objective of these studies was to evaluate some biological characteristics of this species fed on three types of diet (whitefly nymphs, Sitotroga cerealella eggs and a mix of both) and on two host plants (tomato and tobacco), under controlled experimental conditions. Preimaginal developmental time for female and male bugs was shorter in the presence of whiteflies than with only moth eggs. Females lived longer when they ate only whitefly nymphs compared to a mixed diet or only moth eggs. The amount of adult descendants was greater when bugs could eat whiteflies, regardless of the presence of S. cerealella. Embryonic development time, male longevity and sex proportion were not affected by the diet or the host plant. Prey consumption was evaluated for three T. cucurbitaceus life history stages (fourth/fifth instar nymphs, female and male adults) on two types of prey (whitefly nymphs and S. cerealella eggs). On tomato, females were more voracious than males and nymphs. On tobacco, adults and nymphs consumed more S. cerealella than T.vaporariorum nymphs, but again, bug females preyed more than males and nymphs. Results demonstrate that T. cucurbitaceus can survive, develop and reproduce normally using both T. vaporariorum and S. cerealella eggs as prey on tobacco or tomato plants. This information can be useful for managing this predator against T. vaporariorum through conservative or augmentative biological control strategies. 相似文献
