Cicer canariense is a threatened perennial wild chickpea endemic to the Canary Islands. In this study, rhizobia that nodulate this species in its natural habitats on La Palma (Canary Islands) were characterised. The genetic diversity and phylogeny were estimated by RAPD profiles, 16S-RFLP analysis and sequencing of the rrs, recA, glnII and nodC genes. 16S-RFLP grouped the isolates within the Mesorhizobium genus and distinguished nine different ribotypes. Four branches included minority ribotypes (3–5 isolates), whereas another five contained the predominant ribotypes that clustered with reference strains of M. tianshanense/M. gobiense/M. metallidurans, M. caraganae, M. opportunistum, M. ciceri and M. tamadayense. The sequences confirmed the RFLP groupings but resolved additional internal divergence within the M. caraganae group and outlined several potential novel species. The RAPD profiles showed a high diversity at the infraspecific level, except in the M. ciceri group. The nodC phylogeny resolved three symbiotic lineages. A small group of isolates had sequences identical to those of symbiovar ciceri and were only detected in M. ciceri isolates. Another group of sequences represented a novel symbiotic lineage that was associated with two particular chromosomal backgrounds. However, nodC sequences closely related to symbiovar loti predominated in most isolates, and they were detected in several chromosomal backgrounds corresponding to up to nine Mesorhizobium lineages. The results indicated that C. canariense is a promiscuous legume that can be nodulated by several rhizobial species and symbiotypes, which means it will be important to determine the combination of core and symbiotic genes that produce the most effective symbiosis. 相似文献

Genetic diversity at 28 microsatellite loci was studied in a natural population of Triticum dicoccoides at the Ammiad microsite, north of the Sea of Galilee, Israel. This microsite was subdivided into four major habitats, North,
Valley, Ridge and Karst, and further subdivided into nine subhabitats. The units thus defined showed strong and highly significant
differentiation in ecological factors; in particular with respect to cover, proximity and height of rocks, and surface soil
moisture after early rains. The results showed that allele distributions at microsatellite loci were nonrandom and associated with habitats. Significant genetic differentiation and variation in repeat number were found among subpopulations
in the four major habitats and nine subhabitats. Habitat-specific and -unique alleles and linkage disequilibria were observed in the Karst subpopulation. The subpopulations dwelling in drier habitats
and subhabitats showed higher genetic diversities at microsatellite loci. These results suggest that natural selection, presumably
through aridity stress, acts upon microsatellite divergence predominantly on noncoding sequences, thereby contributing to
differences in fitness.

The genetic structure of 35 populations of wild relatives of cultivated wheats, all collected in Syria and Lebanon, was assessed
using ten isozymes. The populations consisted of diploid goat grass, Aegilops speltoides, diploid wild wheats, Triticum monococcum spp. aegilopoides and T. urartu, and tetraploid wild wheat, T. turgidum ssp. dicoccoides. The majority of the populations were polymorphic (P=0–70%) having low within-population mean genetic diversity (Hep=0.05–0.10) and relatively high within-species genetic diversity (Hes=0.14–0.31). The linkage between loci did not seem to be one of the causes for the observed polymorphism. All four species
showed significant inbreeding at both the population (0.31–0.64) and species (0.77–0.96) levels, and the extent of inbreeding
did not correlate with mating systems. Despite their apparent common ecological and evolutionary history, between-population
or between-species level genetic identity was low (I=0.43–0.86). Among the diploid species, populations of Ae. speltoides clustered distinctly from those overlapping clusters of T. monococcum ssp. aegilopoides and T. urartu. The tetraploid species T. turgidum ssp. dicoccoides had relatively less genetic diversity (Hes=0.14) and was highly homozygous (F=0.96). The results suggest that these wild progenitors of cultivated wheats have undergone
extensive local differentiation and inbreeding. We discuss the implications of our results on the management of wild wheat
and goat grass populations.

利用湿地水生植被生长旺盛、光谱反射较强、光谱信息比较丰富的8月份中分辨率Landsat TM和ETM+多光谱遥感影像,采用面向对象的分类方法,进行野鸭湖湿地水生植被的提取。研究表明:在提取过程中,通过对原始影像进行主成分变换和穗帽变换,将主要信息与噪声分离,不仅减小了数据冗余和波段间的相关性,而且增大了影像上湿地水生植被与其他地物类型光谱和空间信息的差异性,并结合野外水生植被光谱特征分析,选择归一化植被指数NDVI与归一化水体指数NDWI辅助分类,构建特征波段或波段组合,然后,确定适当的隶属度函数和阈值范围,构建分类决策树,完成湿地水生植被的自动分类,提高了影像分割与面向对象分类的精度,取得了较为理想的湿地水生植被提取结果。2002年和2008年两景影像的总体分类精度分别达到86.5%和85.44%,表明中分辨率TM影像可以满足湿地水生植被提取的需要,又因为其具有较高的波谱分辨率、极为丰富的信息量、相对较低的价格、长时间序列,可以作为近20a湿地水生植被提取和动态变化监测的主要数据源。 相似文献

基于光谱指数的植物叶片叶绿素含量的估算模型 总被引:13,自引:0,他引:13
叶绿素是光合作用能力和植被发育阶段的指示器,是监测湿地植被生长健康状况的重要指标之一;高光谱遥感技术可以为植物叶绿素含量的定量化诊断提供简便有效、非破坏性的数据采集和处理方法。为保证被探测叶片面积相同,消除背景反射、叶片表面弯曲造成的光谱波动及叶片内部变异造成的影响,研究采用Field Spec 3光谱仪加载手持叶夹式叶片光谱探测器,测定野鸭湖湿地典型植物的叶片高光谱反射率数据,同时通过分光光度计室内测定相应叶片的叶绿素含量。采用相关性及单变量线性拟合分析技术,建立二者的关系模型,包括叶绿素含量与"三边"参数的相关模型以及比值光谱指数(SR)模型和归一化差值光谱指数(ND)模型,并采用交叉检验中的3K-CV方法对估算模型进行模型精度检验。结果表明:植物叶片叶绿素含量与"三边"参数大多都呈极显著相关,相关系数最大达到0.867;计算光谱反射率组成的比值(SR)和归一化(ND)光谱指数与叶绿素含量的决定系数,总体相关性比较高,较好的波段组合均为550—700nm与700—1400nm以及550—700nm与1600—1900nm,与叶绿素含量相关性最好的指数分别是SR(565nm,740nm)和ND(565nm,735nm)。并通过选取相关性最佳的光谱特征参数,分别基于"三边"参数和ND模型指数构建了植物叶片叶绿素含量的估算模型。其中,基于红边位置(WP_r)光谱特征参数和ND(565nm,735nm)光谱指数建立的叶绿素含量估算模型,取得了较好的测试效果,检验拟合方程的决定系数(R2)都达到0.8以上,估算模型分别为y=0.113x-78.74,y=5.5762x+4.4828。通过3K-CV方法进行测试和检验,植物叶绿素含量估算模型均取得了较为理想的预测精度,预测精度的分别为93.9%及90.7%。高光谱遥感技术对植被进行微弱光谱差异的定量分析,在植被遥感研究与应用中表现出强大优势,为植物叶绿素含量诊断中的实际应用提供了重要的理论依据和技术支持。 相似文献

采用线粒体DNA 细胞色素b 基因片段为遗传标记, 对黄海南部和东海银鲳(Pampus argenteus)群体的遗传结构进行了分析。在所分析的6 个采样点116 个个体中, 共检测到40 种单倍型。6 个群体均呈现出高单倍性多样性(0.647-0.895)和低核苷酸多样性(0.0008-0.0026)的特点。单倍型邻接关系树的拓扑结构简单,未呈现明显的地理谱系结构。分子方差分析和Fst 显示银鲳的遗传变异来自群体内个体间, 而群体间无显著遗传分化。Exact 检验表明单倍型在两两群体间分布频率的差异不显著。研究结果表明, 黄海南部、东海银鲳群体间具有高度的基因交流, 是一个随机交配群体。较强的扩散能力, 黄海、东海的海洋环流以及近期的群体扩张可能是造成黄海南部、东海银鲳群体间遗传同质性较高的原因。
