We characterized microsatellite loci for the sand skink (Neoseps reynoldsi) for future studies of genetic structure in this threatened taxon. We screened a partial genomic library enriched for microsatellites, designed primers for eight loci and assessed these markers for polymorphism across 11 populations in central Florida. Preliminary analyses indicate deviations from Hardy–Weinberg expectations for most loci, suggesting population genetic structure across the sampled populations; therefore, understanding genetic connectivity is critical for maintaining genetic variation in this species.  相似文献

利用透射电镜观察中国石龙子附睾成熟精子的超微结构。顶体囊前部扁平、由皮质和髓质组成 ,穿孔器中度倾斜、顶端尖 ,穿孔器基板塞子状 ,细胞核长形 ,核内小管缺 ,核前电子透亮区小 ,核肩圆 ,核陷窝锥形。颈段具片层结构 ,近端中心粒和远端中心粒的长轴呈直角 ,9束外周致密纤维与远端中心粒相应的 9束三联微管相联 ,向后与轴丝相应的 9束双联微管相联 ,中央纤维与 2个中央单微管相联。中段短 ,多层膜结构缺 ,含有线状嵴的柱状线粒体 ,不规则卵状致密体组成不连续的环状结构 ,纤维鞘伸入中段 ,具终环。线粒体与环状结构的模式为 :rs1 /mi1 ,rs2 /mi2 ,rs3/mi3,rs4 /mi4。主段前面部分具薄的细胞质颗粒区。纤维 3和 8至主段前端消失。轴丝呈“9 2”型。中国石龙子精子超微结构具有塞子状的穿孔器基板、致密体形成不连续的环状结构和纤维鞘始于ms2等特征与巨石龙子群和蜓蜥 -胎生群不同。没有发现石龙子科精子的独征  相似文献

Caudal autotomy is a defense mechanism used by numerous lizards to evade predators, but this entails costs. We collected 294 adult Chinese skinks (Eumeces chinensis) from a population in Lishui (eastern China) to evaluate energetic and locomotor costs of tail loss. Of the 294 skinks, 214 (c. 73%) had previously experienced caudal autotomy. Neither the proportion of individuals with regenerated tails nor the frequency distribution of locations of the tail break differed between sexes. We successively removed four tail segments from each of the 20 experimental skinks (adult males) initially having intact tails. Lipid content in each removed tail segment was measured, and locomotor performance (sprint speed, the maximal length traveled without stopping and the number of stops in the racetrack) was measured for each skink before and after each tail-removing treatment. Another independent sample of 20 adult males with intact tails was measured for locomotor performance to serve as controls for successive measurements taken for the experimental lizards. Caudal lipids were disproportionately stored along the length of the tail, with most lipids being aggregated in its proximal portion. Tail loss significantly affected sprint speed, but not the maximal length of, or the number of stops during the sprint. However, the adverse influence of tail loss on sprint speed was not significant until more than 51% of the tail (in length) was lost. Our data show that partial tail loss due to predatory encounters or other factors may not severely affect energy stores or locomotor performance in E. chinensis. As tail breaks occurred more frequently in the proximal portion of the tail in skinks collected from the field, we conclude that caudal autotomy occurring in nature often incurs substantial energetic and locomotor costs in E. chinensis.  相似文献

Abstract Reproduction has been shown to be costly in viviparous skinks, both in terms of survival and in terms of growth and future reproductive output. One possible source of such costs could be a reduction in food intake, with an associated reduction in energy available for maintenance and future reproduction. Therefore, I compared food intake in male and female southern water skinks (Eulamprus tympanum), to determine whether food intake was reduced during reproduction in either sex. In addition, I measured the relationship between reduction in food intake and the total volume of offspring or testis. A reduction in food intake proportional to the volume of offspring suggests that costs associated with reduced energy intake may be fecundity dependent. Food intake was estimated using the total volume of stomach contents of individuals. Females ate significantly less (mean volume of stomach contents = 0.13mL) than males (mean = 0.36mL) just before parturition in January, when embryos were largest. After parturition, females ate significantly more than males. Otherwise, there were no significant differences between the food intake of males and females. Females reduced food intake by eating smaller food items. Females with relatively large numbers of ova or embryos ate relatively less, and therefore decreased food intake in females may be a fecundity-dependent cost of reproduction in these lizards. Males did not reduce food intake during periods when testes were enlarged and mating occurred. Decreased food intake during reproduction is probably an important source of reproductive costs to females of these viviparous lizards, but does not appear to be important in males.  相似文献

Abstract: The current primary threats to biodiversity on a global scale are species invasions and habitat modification. Management of vulnerable populations often involves a lengthy sequence of 1) research to identify threats and recommend management strategies, 2) active management, and 3) results monitoring to assess effectiveness of management. The last mainland population of the large, endemic New Zealand skink (Whitaker's skink [Cyclodina whitakeri]) provides an opportunity to test this process in a system where the synergistic effects between invasive species (introduced rodents and grasses) have predicted outcomes. A low abundance of Whitaker's skink at the Pukerua Bay Scientific Reserve in the 1980s prompted management recommendations to remove grazing stock and revegetate the site to simultaneously restore habitat and provide protection against introduced mammalian predators. Since Whitaker's skink have low detectability, it was recommended that sympatric copper skink (C. aenea) be used an indicator species of management effectiveness. Grazing stock were removed in 1987, but efforts to revegetate the site were ineffective. Long-term monitoring (1984-2006) of the Whitaker's skink population and 4 other sympatric lizard species within a 336-m2 area at the site resulted in 1,693 lizard captures over 7,597 trap days. Whitaker's skink represented 2.8% of all captures in 1984-1988, but declined relative to other species to represent only 0.2% of captures in 2000-2006 (representing 2 individuals). Congeneric copper skink showed a similar decline with capture rates also approaching zero by 2006. Removing grazing stock did not result in an increased abundance of Whitaker's skink or copper skink through improved habitat quality, as was intended by the management recommendation. Instead, reduced grazing has allowed introduced seeding grasses to proliferate, which may have led to periodic rodent irruptions, supporting a guild of introduced mammalian predators and depleting populations of Whitaker's skink and copper skink. In this instance, attempted protection may have driven a vulnerable population towards extinction. We recommend investigating the feasibility of constructing a mammal-proof fence around the core Whitaker's skink habitat, as the last remaining management option to salvage the population.  相似文献

The inbreeding avoidance hypothesis predicts that organisms that often encounter relatives as potential mates should evolve behaviours to avoid incestuous matings. Avoidance behaviours have practical importance for small populations because deleterious genetic processes may be less imminent than otherwise expected from genetic models that assume random mating. I used genetic techniques to investigate the extent of inbreeding and inbreeding avoidance behaviours in rare lizards from southern New Zealand. Grand skinks, Oligosoma grande, live in small patchily distributed groups, and have low rates of inter-group dispersal (ca. 3–20% disperse). I used data from 15 microsatellite loci to test the hypothesis that adults are likely to encounter kin as potential mates and will inbreed. These data showed that adult skinks usually inhabited rock outcrops with adult relatives of the opposite sex – up to 35% of potential mates were of equivalent relatedness as half-sibs and 17% were equivalent to full sibs. However, skinks did not preferentially breed with less related mates, and 18.2% of matings were between individuals of equivalent relatedness as full-sibs. Instead, skinks mated with partners of all levels of relatedness, and were promiscuous – almost half of adult females and nearly three quarters of adult males reproduced with multiple partners. In addition, inbreeding had no effect on survival of offspring in their first year. Two other putative mechanisms of inbreeding avoidance, sex-biased and natal dispersal, were not pronounced in this species. This study adds to a growing list of species that inbreed despite the risks.  相似文献

We isolated 10 polymorphic microsatellite markers for the skink Mabuya affinis from genomic libraries enriched for (AAGG)n and (AAAG)n repetitive elements. The number of alleles ranged from eight to 13 per locus with the observed heterozygosity ranging from 0.60 to 0.92. These markers will be useful for analysis of questions concerning population genetic structure and models of speciation.  相似文献
