Summary Thionins are high sulphur basic polypeptides present in the endosperm of Gramineae. In wheat there are three thionins encoded by genes located in the long arms of chromosomes 1A, 1B and 1D. Rye has one thionin encoded by a gene which has been assigned to chromosome 1R after analysis of the Imperial-Chinese Spring rye-wheat disomic addition lines. Commercial varieties and experimental stocks with a 1B/1R substitution carry the thionin from rye ( R) instead of the B thionin from wheat. The R thionin gene is not located in the large chromosomal segment representing most of the short arm of chromosome 1R.  相似文献

Abstract. The onset of prestalk differentiation of Dictyostelium discoideum has been thought to be triggered by differentiation inducing factor (DIF), which is secreted by differentiating cells. We characterized the cell-type specific proteins, wst31 (prestalk and stalk specific) and wst34 (stalk specific), using the mutant HM44 which is defective in DIF-production, and examined the effects of DIF and cAMP on the formation of the proteins. In the mutant HM44, wst34 was formed only in the presence of exogenous DIF as reported for other prestalk/stalk markers (e.g. pDd63 and acid phosphatase-2), which indicates the DIF-requirement for this protein. By contrast, the accumulation of wst31 in this mutant occurred in the presence of cAMP regardless of the presence of exogenous DIF. Thus, we propose a new and distinct state (or stage) in prestalk differentiation, where the expression of wst31 occurs but not that of pDd63 or acid phosphatase-2.  相似文献

A new method allowing control of rhizosphere pH and mineral nutrition was applied to study depletion of various organic and inorganic phosphorus fractions extractable sequentially with 0.5M KHCO3 (pH 8.5), 0.1M NaOH and residual P extractable with 6M H2SO4 from the rhizosphere soil.Soil pH was affected about 2 mm from the root mat. Depletion zones of inorganic P (KHCO3-Pi) extractable with 0.5M KHCO3 extended up to about 4 mm but the depletion zones of all other P fractions were about 1 mm only. The root-induced decrease of soil pH from 6.7 to 5.5 increased the depletion of total P from all fractions by 20% and depletion of KHCO3-Pi and residual P by 34% and 43%, respectively. Depletion of organic P (KHCO3-Po) extractable with 0.5M KHCO3 was not affected by a change in rhizosphere pH. With constant or increased pH, depletion of inorganic P (NaOH-Pi) was 17% and organic P (NaOH-Po) was 22% higher than with decreased pH. Only 54–60% of total P withdrawn from all fractions was from KHCO3-Pi. Substantial amounts of KHCO3-Po and NaOH-Po were mineralized and withdrawn from the rhizosphere within 1 mm from the root mat, as 11–15% of total P withdrawn originated from the organic P fractions. A remaining 11–16% was derived from NaOH-Pi, and 15–18% from residual P fractions likely to be rather immobile. Thus, 40–46% of the P withdrawn near the root mat of rape originated from non-mobile P fractions normally not included in 0.5M NaHCO3 extraction used to obtain an index of plant-available soil P.  相似文献

For in vivo demonstration of acid phosphatase activity in the rhizosphere of soil-grown plants filter papers were treated with a mixture of 1-naphthyl phosphate as substrate and the diazonium salt Fast Red TR as an indicator. After enzymatic hydrolysis, 1-naphthol forms a red complex with Fast Red TR. This method was applied to 8-day old maize plants and 3-year old Norway spruce plants growing in rhizoboxes in soil under non-sterile conditions. The treated filter paper is placed at the surface of roots and soil and acid phosphatase activity is visualized as a red-coloured root print on the filter paper. The method can be used as a qualitative analysis of acid phosphatase in the rhizosphere. It also allows a rough estimate of phosphatase activity in different root zones.  相似文献

Mobilization of soil P as the result of plant-induced changes of soil pH in the vicinity of plant roots was studied. Seedlings of ryegrass were grown in small containers separating roots from soil by a 30-μm meshed nylon screen which root hairs could penetrate but not roots. Two soils were used, a luvisol containing P mainly bound to calcium and an oxisol containing P mainly bound (adsorbed) to iron and aluminum. Plant-induced changes of soil pH were brought about by application of ammonium-or nitrate-nitrogen. After plants had grown for 10 d the soil was sliced in thin layers parallel to the root mat which had developed on the screen, and both soil pH and residual P determined. Mobilization of P was assessed by P-depletion profiles of the rhizosphere soil. Soil pH at the root surface decreased by up to 1.6 units as the result of ammonium N nutrition and it increased by up to 0.6 units as the result of nitrate N nutrition. These changes extended to a distance between 1 and 4 mm from the root surface depending on the type of soil and the source and level of nitrogen applied. In the luvisol, compared to zero-N treatment, P mobilization increased with the NH4-induced decrease in pH, whereas the NO3-induced pH increase had no effect. In contrast, in the oxisol a similar pH decrease caused by NH4 nutrition had no effect, whereas the pH increase caused by NO3 increased markedly the mobilization of soil P. It is concluded that in the luvisol calcium phosphates were dissolved by acidification, whereas in the oxisol adsorbed phosphate was mobilized by ligand exchange.  相似文献

Effects of N sources (ammonium, nitrate and ammonitrate) and VA mycorrhizae (Glomus intraradices) on rhizosphere soil characteristics (pH, exchangeable acidity, exchangeable cations, inorganic N concentrations) growth and nutrient acquisition of coffee seedlings (Coffea arabica L. cv guatemala) were investigated in a pot study with an acid soil (Red Bluff Loam) sterilized by autoclaving. Ammonium addition decreased rhizosphere pH while nitrate and ammonitrate additions both increased rhizosphere pH. Mycorrhizae induced a higher pH, a lower exchangeable acidity and higher values of exchangeable cations in the rhizosphere. Ammonium addition resulted in a lower mycorrhizal infection than the two other N sources. Mycorrhizal plants grew better and accumulated more N, Ca and Mg than non-mycorrhizal plants.  相似文献

The laboratory germination (criterion radicle emergence) of seven seed lots of winter wheat cv. Slejpner was similar. However, they differed in vigour as demonstrated by differences in germination after controlled deterioration carried out at a range of seed moisture contents, at two temperatures and for different times. A vigour assessment for each lot was quantified by calculating a value for the seed lot constant Ki, of the viability equation. Germination in lower water potentials reduced the uptake of water, radicle and coleoptile emergence and radicle and coleoptile extension. There was no difference in the water uptake of seed lots differing in vigour. However, seed lots of lower vigour showed less radicle emergence, less coleoptile emergence and shorter radicles than higher vigour seed lots in low water potentials. Similarly, controlled deterioration resulted in reduced radicle and coleoptile emergence and growth compared to unaged seed, and also to a greater sensitivity to low water potentials. The implications for field establishment are discussed.  相似文献

Summary Seven isolates belonged toA. brasilense and 3 belonged toA. lipoferum. Isolates having more denitrifying capacity fixed less nitrogen in nitrogen free semi-solid malate medium. One strain ofA. lipoferum having high nitrogen fixing capacity with negative test for denitrification was tested as inoculant to supplement the nitrogen need of a wheat crop in field condition with different doses of N with and without the inoculant. While control without nitrogen yielded 1260 kg/ha the yield in inoculated treatment was 2070 kg/ha resulting in significant increase. In a treatment receiving 40 kg N/ha the grain yield was 2370 kg/ha as against yield of 3110 kg/ha in a similar treatment receiving fertiliser plus inoculant. Thus increase in yield was about 30%. Further the treatment receiving 80 kg N/ha yielded 2970 kg/ha as against yield of 4150 kg/ha in a treatment receiving inoculant alongwith the above dose of the fertiliser. Thus increase in yield due to application of inoculant was about 36%. Similarly, the uptake of N in different treatments was augmented due to inoculation of seeds with the culture.  相似文献

Summary Studies revealed that the application of fertilizer nitrogen brought a significant increase in grain and straw yield of wheat. The significant effect was also noticed on such yield contributing characters like number of effective tillers per metre row length, spike length, and number of grains per spike. The increase of nitrogen level from 40 kg/ha to 80 kg/ha also brought a significant increase in yield and yield contributing characters. The application of entire dose of nitrogen at the time of sowing was as good as its split application. The application of nitrogen also influenced the nitrogen concentration of grains whereas, the other treatments did not influence the nitrogen concentration in grains or straw. The nitrogen treatments did not influence the moisture content of soil. The application of mulch or mulch+Kaolin resulted in significantly higher content of soil moisture in 0–15 cm soil depth as compared to control or Kaolin spray alone which was simultaneously reflected in yield and yield contributing characters.  相似文献

Summary The C-banding patterns of wheat chromosomes in 7 hexaploid triticale and 7 wheat genotypes are described and compared. All 14 wheat chromosome pairs were individually identified in the triticales and a tetraploid wheat, and all the B and two A genome chromosome pairs in the hexaploid wheat genotypes. Little variation was found between genotypes in the distribution of C-bands but considerable variation was found in their size, total number and total length.  相似文献
