Two types of time scales – 10-day intervals (D) and phenological phases (P) – were applied to environmental variables for the development of statistical regression models relating to the southwest pampa region of Argentina, with the aim of detecting the effects of weather on wheat yields for the period 1977–1999. The parameters were grouped as meteorological and processed variables and indices. The processed variables used were total soil water availability (SWA) and the ratio of actual evapotranspiration to potential evapotranspiration (), obtained from a water-balance model in which the moisture anomaly index (Z) and the Palmer drought severity index (PDSI) were calculated according to Palmers model. For these parameters it was possible to establish the times of year and the phenological phases with the best correlation to grain yield. The regression equation for meteorological variables on a 10-day scale provides one of the best fits. Using mixed parameters, the two models, D and P, give rise to a standard error of estimate of approximately 200 kg ha–1. Truncated models perform better on a P scale than on a D scale. The use of phenological stages improved yield assessment, particularly for those years with extreme meteorological conditions. The optimum models were tested and root-mean-square errors (RMSE) of 440 kg ha–1 and 470 kg ha–1 were obtained for P and D scales respectively.  相似文献

Front-face fluorescence was used to assess some structural features of proteins in wheat flours, taking advantage of the fact that this technique allowed a direct approach to a number of systems in which most proteins cannot be solubilized without using conditions that strongly modify the structure of proteins in the original material. In this study, we addressed structural changes ensuing from solvation and from mechanical deformation of proteins in wheat flour. Solvation of proteins in the systems under investigation was monitored by taking fluorescence emission spectra of the protein tryptophans (or of protein-bound 1,8-anilino-naphthalene-sulfonate added in appropriate amounts) in mixtures of increasing water content. In separate experiments, changes in overall protein surface hydrophobicity were estimated by titrating flours-mixed with appropriate amounts of water to a dough-like consistency-with increasing concentrations of the fluorescent hydrophobic probe, 1,8-anilino-naphthalene-sulfonate. This approach allowed concomitant determination of the overall binding capacity and of the apparent affinity for the probe of proteins. To test the usefulness of the structural information obtained from these approaches, they were also applied to characterize dough at different levels of mechanical stress, prepared from either wheat flour or semolina.  相似文献

Microenvironmental studies regarding plant oxygen release in a wastewater environment are important to understand the principles of constructed wetlands for wastewater treatment. pH, oxidation reduction potential (ORP), and dissolved oxygen (DO) microprofiles for the lateral and main roots of the bulrush (Scirpus validus) in a vertical flow constructed wetland fed with municipal wastewater were measured using microelectrode techniques. pH was found to be low (6.91-6.98) near the lateral root surface, indicating possible nitrification or H(+) extrusion. The ORP at the lateral root surface was between +250 and +317 mV and gradually reached the bulk solution ORP (+14 to -54 mV) at a radial distance of approximately 4,750 microm. DO values at the lateral root surface varied from 0.64-2.04 mg L(-1) as bulk biochemical oxygen demand (BOD) changed from 24 to 1,267 mg L(-1). DO at the lateral root surface and the thickness of the oxygen layer around the root marginally increased with an increase in bulk BOD, while ORP at the lateral and main root surface decreased. pH and DO values did not change near the main root and had the bulk solution values. The results of this study provide insights into root-induced microenvironments and would be helpful for the quantification of the total amount of oxygen contributed by plants in constructed wetlands.  相似文献

Spatial distributions of root systems of Larix gmelinii (Rupr.) Rupr. trees were examined in two stands in central Siberia: an even-aged stand (ca. 100 yrs-old) and a mature, uneven-aged (240–280 yrs-old) stand. Five larch trees of different sizes were sampled by excavating coarse roots (diameter > 5 mm) in each stand. Dimensions and ages of all first-order lateral roots were measured. Micro-scale conditions of soil temperature and soil water suction (each 10 cm deep) were also examined in relation to earth hummock topography (mound vs. trough) and/or ground floor vegetation types (moss vs. lichens). All larch trees developed superficial root systems, consisting of the aborted short tap root (10–40 cm in soil depth) and some well-spread lateral roots (n= 4-13). The root network of each tree was asymmetric, and its rooting area reached about four times the crown projection area. Lateral roots generally expanded into the upper soil layers of the mounds where summer soil temperature was 1–6°C higher than inside nearby troughs. Chronological analysis indicated that lateral root expansion started successively from lower to upper parts of each aborted tap root, and some lateral roots occurred simultaneously at several decades after tree establishment. The process of root system development was likely to be primarily linked with post-fire dynamics of rhizosphere environment of the permafrost soils.  相似文献

Small-angle X-ray scattering (SAXS) and scanning electron microscopy (SEM) were used to investigate the internal structure of wheat starch granules with different amylose content. Different approaches were used for treatment (interpretation) of SAXS data to assess the values of structural parameters of amylopectin clusters and the size of crystalline and amorphous lamella in different wheat starches. The average values of the semi-crystalline growth rings thickness in starches have been determined and the relationship between structural characteristics and thermodynamic melting parameters is discussed.  相似文献

The objective of proteomics is to get an overview of the proteins expressed at a given point in time in a given tissue and to identify the connection to the biochemical status of that tissue. Therefore sample throughput and analysis time are important issues in proteomics. The concept of proteomics is to encircle the identity of proteins of interest. However, the overall relation between proteins must also be explained. Classical proteomics consist of separation and characterization, based on two-dimensional electrophoresis, trypsin digestion, mass spectrometry and database searching. Characterization includes labor intensive work in order to manage, handle and analyze data. The field of classical proteomics should therefore be extended to also include handling of large datasets in an objective way. The separation obtained by two-dimensional electrophoresis and mass spectrometry gives rise to huge amount of data. We present a multivariate approach to the handling of data in proteomics with the advantage that protein patterns can be spotted at an early stage and consequently the proteins selected for sequencing can be selected intelligently. These methods can also be applied to other data generating protein analysis methods like mass spectrometry and near infrared spectroscopy and examples of application to these techniques are also presented. Multivariate data analysis can unravel complicated data structures and may thereby relieve the characterization phase in classical proteomics. Traditionally statistical methods are not suitable for analysis of the huge amounts of data, where the number of variables exceed the number of objects. Multivariate data analysis, on the other hand, may uncover the hidden structures present in these data. This study takes its starting point in the field of classical proteomics and shows how multivariate data analysis can lead to faster ways of finding interesting proteins. Multivariate analysis has shown interesting results as a supplement to classical proteomics and added a new dimension to the field of proteomics.  相似文献

The capability of plants to promote the microbial degradation of pollutants in rhizosphere soil is a principal mechanism of phytoremediation of PAH-contaminated soil. The formation of a specific rhizosphere microbocenosis with a high degradative potential toward contaminants is largely determined by plant species. The comparative PAH-degradation in unplanted soil and in soil planted with reed (Phragmites australis) and alfalfa (Medicago sativa) was studied in pot experiments during 2 years. Both alfalfa and reed successfully remediated contaminated soil by degrading 74.5 and 68.7% of PAHs, respectively. The study of the rhizosphere, rhizoplane, and unplanted-soil microflora in experimental pots showed that alfalfa stimulated the rhizosphere microflora of PAH-contaminated soil more effectively than did reed. Alfalfa clearly enhanced both the total number of microorganisms (1.3 times, according to fluorescence microscopy data) and the rate of the PAH-degrading population (almost seven times, according to plate counting). The degradative potential of its rhizosphere microflora toward PAHs was higher than the degradative activity of the reed rhizosphere. This study provides relevant information for the successful application of alfalfa to phytoremediate PAH-contaminated soil.  相似文献

Wheat plants (Triticum aestivum L. 'Lyallpur'), limited to a single culm, were grown at day/night temperatures of either 18/13 degrees C (moderate temperature), or 27/22 degrees C (chronic high temperature) from the time of anthesis. Plants were either non-droughted or subjected to two post-anthesis water stresses by withholding water from plants grown in different volumes of potting mix. In selected plants the demand for assimilates by the ear was reduced by removal of all but the five central spikelets. In non-droughted plants, it was confirmed that shading following anthesis (source limitation) reduced kernel dry weight at maturity, with a compensating increase in the dry weight of the remaining kernels when the total number of kernels was reduced (small sink). Reducing kernel number did not alter the effect of high temperature following anthesis on the dry weight of the remaining kernels at maturity, but reducing the number of kernels did result in a greater dry weight of the remaining kernels of droughted plants. However, the relationship between the response to drought and kernel number was confounded by a reduction in the extent of water stress associated with kernel removal. Data on the effect of water stress on kernel dry weight at maturity of plants with either the full complement or reduced numbers of kernels, and subjected to low and high temperatures following anthesis, indicate that the effect of drought on kernel dry weight may be reduced, in both absolute and relative terms, rather than enhanced, at high temperature. It is suggested that where high temperature and drought occur concurrently after anthesis there may be a degree of drought escape associated with chronic high temperature due to the reduction in the duration of kernel filling, even though the rate of water use may be enhanced by high temperature.  相似文献

In contrast to cereals or other crops, legumes are known to acidify the rhizosphere even when supplied with nitrates. This phenomenon has been attributed to N2 fixation allowing excess uptake of cations over anions; however, as we have found previously, the exposure of the shoot to illumination can cause rhizosphere acidification in the absence of N2 fixation in cowpea (Vigna unguiculata L. Walp). In this study, we examined whether the light-induced acidification can relate to photosynthetic activity and corresponding alterations in cation-anion uptake ratios. The changes of rhizosphere pH along the root axis were visualized using a pH indicator agar gel. The intensity of pH changes (alkalization/acidification) in the rhizosphere was expressed in proton fluxes, which were obtained by processing the images of the pH indicator agar gel. The uptake of cations and anions was measured in nutrient solution. The rhizosphere was alkalinized in the dark but acidified with exposure of the shoots to light. The extent of light-induced acidification was increased with leaf size and intensity of illumination on the shoot, and completely stopped with the application of photosynthesis inhibitor. Although the uptake of cations was significantly lower than that of anions, the rhizosphere was acidified by light exposure. Proton pump inhibitors N,N'-dicyclohexyl carbodimide and vanadate could not stop the light-induced acidification. The results indicate that light-induced acidification in cowpea seedlings is regulated by photosynthetic activity, but is not due to excess uptake of cations.  相似文献
