A study of the mangrove fish fauna in a bay of Martinique Island (French West Indies) was carried out at different seasons during two consecutive years. fishes were sampled with specific hoop-nets in the coastal areas at 8 stations.A total of 87 species was collected in the bay. Most individuals were represented by small-size specimens and juveniles. The overall species richness varied according to the stations and the sampling periods. The biomass and number of individuals were variable according to the location but remained stable in time. A factor correspondence analysis and a hierarchical clustering with median links were used to follow the evolution of the stations in space and time. Two types of stations were differentiated: the stations characterized by the mangrove and those under the influence of seagrass beds. A seasonal cycle, opposing the dry periods to the others, was observed.Thus, it seems that the use of the mangrove habitat by the fishes is optimized through a complete reorganization of communities in terms of species composition whereas the overall number and biomass remain stable. This model remains valid even for the most constraining biota of the mangrove ecosystem inhabited by a small number of well adapted species.  相似文献

Isolates of novel strains of Kashmir bee virus (KBV) were obtained from field-collected dead adults of Apis mellifera from honey bee colonies in Canada and Spain. They differed from other strains of KBV in their tendency to aggregate in dilute buffer solution and in containing only three proteins when analysed by SDS-polyacrylamide gel electrophoresis compared with five proteins resolved in the type strain of KBV from Apis cerana in India and six proteins in KBV strains from South Australia and New Zealand. Immunodiffusion tests and Western blotting studies indicated that the five virus isolates were serologically related and all were related to acute paralysis virus (APV). The world distribution of KBV strains and their apparent relationship with APV are discussed.  相似文献

The occlusion of capillary vessels results in low oxygen tension in adjacent tissues which triggers a signaling cascade that culminates in neovascularization. Using bovine retinal capillary endothelial cells (BRCEC), we investigated the effects of short-term hypoxia on DNA synthesis, phosphotyrosine induction, changes in the expression of basic fibroblast growth factor receptor (bFGFR), protein kinase C (PKCα), heat shock protein 70 (HSP70), and SH2-containing protein (SHC). The effect of protein tyrosine kinase (PTK) and phosphatase inhibitors on hypoxia-induced phosphotyrosine was also studied. Capillary endothelial cells cultured in standard normoxic (pO2 = 20%) conditions were quiesced in low serum containing medium and then exposed to low oxygen tension or hypoxia (pO2 = 3%) in humidified, 5% CO2, 37°C, tissue culture chambers, on a time-course of up to 24 h. DNA synthesis was potentiated by hypoxia in a time-dependent manner. This response positively correlated with the cumulative induction of phosphotyrosine and the downregulation of bFGFR (Mr ~ 85 kDa). Protein tyrosine kinase inhibitors, herbimycin-A, and methyl 2,5-dihydroxycinnamate, unlike genistein, markedly blocked hypoxia-induced phosphotyrosine. Prolonged exposure of cells to phosphatase inhibitor, sodium orthovanadate, also blocked hypoxia-induced phosphotyrosine. The expression of HSP70, PKCα, and SHC were not markedly altered by hypoxia. Taken together, these data suggest that short-term hypoxia activates endothelial cell proliferation in part via tyrosine phosphorylation of cellular proteins and changes in the expression of the FGF receptor. Thus, endothelial cell mitogenesis and neovascularization associated with low oxygen tension may be controlled by abrogating signaling pathways mediated by protein tyrosine kinase and phosphatases. © 1995 Wiley-Liss, Inc.  相似文献

Immunocytochemistry and Western blotting techniques demonstrated that the nervous system and foot of the pond snail Lymnaea stagnalis are rich sources of tubulin, which can be extracted and assembled in vitro in the presence of taxol. Various broad-spectrum antibodies raised against -tubulin and -tubulin yielded qualitatively similar results. One monoclonal antibody to trypanosome -tubulin, however, labelled -tubulin more strongly on both probed sections and Western blots. Cytochemistry and immunoblotting revealed that tyrosinated tubulin constitutes a large proportion of total -tubulin in locomotor cilia of the foot and in axons of the nervous system. Detyrosinated tubulin also appeared to be abundant in the foot cilia but only a very faint band of detyrosinated tubulin was found on protein blots extracted from the central ganglia, and staining was barely detectable in central ganglia or peripheral nerves. Similarly, acetylated tubulin appeared to be abundant in foot cilia, but Western blotting indicated only low levels of acetylated tubulin in the nervous system. Immunocytochemistry indicated that, while most neurons possessed little or no acetylated tubulin, a small number of axons contained significant amounts of this isoform. Thus, while a large amount of tubulin was expected in the nervous system and locomotor cilia of L. stagnalis, the observed distribution of isoforms was unanticipated. Specifically, neurons of other organisms have generally been reported to contain substantial amounts of both detyrosinated -tubulin and acetylated -tubulin. Our results indicate that such findings cannot be generalized across all species. L. stagnalis, with its well studied nervous system and unusual distribution of tubulin isoforms, may prove to be particularly useful for studying the roles of tubulin isoforms in microtubule function and cell activity.  相似文献

In the mid-western Himalaya (altitude 1350 m, rainfall 1100 mm), multipurpose trees found as escapees in agricultural fields or naturally growing in the forests, play an important role in providing fuel, fooder and small timber to the farmers. Shoot elogation, and tree architecture of 4 year old trees of Grewia optiva, Robinia pseudoacacia and Celtis australis (early successionals), and Quercus leucotrichophora, Q. glauca and Ilex odorata (late successionals), were analyzed. All the late successional species showed a proleptic type of bud and branch production, while the early successional trees made growth through syllepsis. The shoot elongation differed significantly (P <0.05) with the crown position, and ranged from 11 to 30 cm in different species. Early successional species tended to maintain a comparatively narrow crown and showed a significantly (P <0.05) higher ramification ratio, and multilayered canopy. The late successionals, in contrast, showed a wide crown with monolayered canopy, adapted to the weak light intensity. There was only one flush of leaves in Q. leucotrichophora and Q. glauca while in the rest of the species there were two distinct flush periods. The results are important for the management of agroforestry trees.  相似文献

A combined palynological and phytosociological methodological approach was used in the reconstruction of precultural forest vegetation in a small model area of Bílý Kí-Janík in the Moravskoslezské Beskydy mountains (northwest Slovakia). Two pollen diagrams show the predominance of Fagus sylvatica, Abies alba and Picea abies in past natural mixed forests. The present-day occurrence and distribution of forest herbs allow the differentiation of five past vegetation types with above mentioned forest climax trees covering the area before the beginning of human interference. A map of reconstructed natural vegetation is presented which can be used for revitalization of present secondary spruce plantations, heavily affected by air pollution.  相似文献

Prestained protein molecular weight standards allow easy, direct visual location of electrophoretically transblotted lanes on nitrocellulose. They also provide a simple and accurate means for calibrating the molecular weights of resolved bands. Commercial prestained protein molecular weight standards, however, appear to have significantly different molecular weights from the original unstained proteins. The extent of the negative cooperativity with MgATP of the Ca2+-stimulated ATPase activity of sarcoplasmic reticulum has been studied with various membrane preparations and under various conditions. Preparations studied were fragmented sarcoplasmic reticulum vesicles, deoxycholate-solubilized and fractionated ATPase, triton extracted reticulum, vesicles reconstituted from either detergent, and limited trypsin digests of the reticulum. Conditions studied were suboptimal, optimal, and inhibitory Ca2+ concentrations; temperatures from 13 to 46 °C; 1 or 5 mm MgCl2; 0.1 m KCl, 0.1 m NaCl, or no added salt; and Triton or deoxycholate present in the assay. With preparations in which vesicles could accumulate Ca2+ ion, the ionophore A23187 was added to prevent inhibition by internal Ca2+ ions. Under all circumstances, the negative cooperativity of MgATP was present (Hill coefficient of 0.2 to 0.8), indicating the persistence of the properties of the enzyme molecule and its lipid environment giving rise to kinetic negative cooperativity. Attempts to measure the number of ATP sites by protection against N-ethylmaleimide inactivation and by binding of an analog suggested, but did not prove, that there was only one specific, active ATP binding site below 0.5 mm. These results are interpreted to be consistent with either of two mechanisms for ATP cooperativity of the Ca2+-stimulated ATPase activity of sarcoplasmic reticulum: (a) a single, high affinity ATP active site and a second, lower affinity “allosteric” activator site; or (b) a single ATP site which demonstrates two affinities through some kinetic mechanism such as a substrate-induced, slow transition.  相似文献

An anti-Mos protein monoclonal antibody, 4A6, was used to investigate the distribution of the antigen in the epididymis, in which the c-mos gene is reportedly expressed. The 4A6-reactive antigen was found on the basement membrane and luminal surface of the epithelial cells in the caput epididymis of BALB/c male mice as well as in the proximal corpus epididymis, the cauda epididymis, and the vas deferens. The 4A6 antigen was also found on the luminal surface of the epithelial cells in the epididymis of male germ cell-deficient C57BL/6J-Wv/Wv mice. This confirmed that the 4A6 antigen does not derive entirely from the testicular c-Mos protein but is synthesized in the epididymis. Western blot analysis revealed that the molecular weight of the epididymal 4A6 antigen was 50 kDa, which is unusually high for the c-Mos protein. With its specific distribution in the epididymis, the protein should play a specific role in functions of the epididymis.  相似文献

Within the great expanse of the Brazilian Amazon Basin,Sphagnum has been greatly overlooked, in part because of its scarcity and in part because of the paucity of trained bryological collectors in the area.
