The Eurasian collared‐dove (Streptopelia decaocto) has recently experienced a population explosion in North America. It is frequently infected with West Nile virus (WNV). To test the hypothesis that the Eurasian collared‐dove is competent to transmit WNV, we experimentally infected two cohorts of doves with two different strains of WNV, CO08, and NY99, respectively. Both virus strains induced a low‐level viremia, capable of infecting a small fraction of vector mosquitoes. We suggest that the Eurasian collared‐dove plays a relatively insignificant role as an amplifying host for WNV, but it may be important where it is locally abundant. 相似文献

Patterns of circadian and ultradian rhythms in the heart rate(HR) are described in a full-term baby with birth asphyxia and convulsions.A 24h HR recording was carried out at the age of 1, 15, 56, 289, and 295 days;West syndrome diagnosis was made when the patient was 3 months old. The HRshowed no circadian rhythm in the follow-up, whereas it is known that thecircadian rhythm appears in healthy infants at the age of 1 month and remainsthereafter. Wolbachia (Rickettsiales: Anaplasmataceae) infects a wide range of arthropods, including several mosquito species. The bacterium is known to induce a plethora of phenotypes in its host, examples being the reproductive phenotype cytoplasmic incompatibility or resistance against infection with arboviruses. The latter is especially relevant when assessing the vector competence of mosquito species for emerging arboviruses. Thus, knowledge of Wolbachia infection status is important for the assessment of vector competence. To facilitate Wolbachia screening in mosquito populations, a quantitative polymerase chain reaction (qPCR) assay was developed to enable high‐throughput analysis of mosquito samples. Using this assay, the Wolbachia infection status of the two most common Culex mosquito species in Germany, Culex pipiens biotype pipiens Linnaeus (Diptera: Culicidae) and Culex torrentium Martini (Diptera: Culicidae), was assessed. About 93% of all tested C. pipiens biotype pipiens individuals were positive for Wolbachia, whereas none of the C. torrentium samples was found to be infected. Furthermore, other applications of the qPCR assay were explored by assessing a potential link between the levels of Wolbachia and West Nile virus (WNV) infections in German C. pipiens biotype pipiens mosquitoes. No relationship was found between the two variables, indicating that a Wolbachia‐induced antiviral phenotype in this mosquito population is not exclusively attributable to the general level of bacterial infection. 相似文献

Recently, Zika virus (ZIKV) emerged as a global public health concern and is distinct from other flaviviruses in many aspects, for example, causing transplacental infection, fetal abnormalities and vector-independent transmission through body fluids in humans. The capsid (C) protein is a multifunctional protein, since it binds to viral RNA in the process of nucleocapsid assembly and plays important roles in virus infection processes by interacting with cellular proteins, modulating cellular metabolism, apoptosis and immune response. Here we solved the crystal structure of ZIKV C protein at a resolution of 1.9 Å. The ZIKV C protein structure contains four α helices with a long pre-α1 loop and forms dimers. The unique long pre-α1 loop in ZIKV C contributes to the tighter association of dimeric assembly and renders a divergent hydrophobic feature at the lipid bilayer interface in comparison with the known C structures of West Nile and dengue viruses. We reported the interaction between the ZIKV C protein and lipid droplets through confocal microscopy analysis. Substitutions of key amino acids in the pre-α1 loop of ZIKV C disrupted the interaction with lipid droplets, indicating that the loop is critical for membrane association. We also recognized that ZIKV C protein possesses broad binding capability to different nucleotide types, including single-stranded and double-stranded RNAs or DNAs. Furthermore, the highly positively charged interface, mainly formed by α4 helix, is proposed to be responsible for nucleotide binding. These findings will greatly enhance our understanding of ZIKV C protein, providing information for anti-ZIKV drug design targeting the C protein. 相似文献

Viruses have compact genomes that encode limited number of proteins in comparison to other biological entities. Interestingly, viral proteins have shown natural abundance of either completely disordered proteins that are recognized as intrinsically disorder proteins (IDPs) or partially disordered segments known as intrinsically disordered protein regions (IDPRs). IDPRs are involved in interactions with multiple binding partners to accomplish signaling, regulation, and control functions in cells. Tuning of IDPs and IDPRs are mediated through post-translational modification and alternative splicing. Often, the interactions of IDPRs with their binding protein partner(s) lead to transition from the state of disorder to ordered form. Such interaction-prone protein IDPRs are identified as molecular recognition features (MoRFs). Molecular recognition is an important initial step for the biomolecular interactions and their functional proceedings. Although previous studies have established occurrence of the IDPRs in Zika virus proteome, which provide the functional diversity and structural plasticity to viral proteins, the MoRF analysis has not been performed as of yet. Many computational methods have been developed for the identification of the MoRFs in protein sequences including ANCHOR, MoRFpred, DISOPRED3, and MoRFchibi_web server. In the current study, we have investigated the presence of MoRF regions in structural and non-structural proteins of Zika virus using an aforementioned set of computational techniques. Furthermore, we have experimentally validated the intrinsic disorderness of NS2B cofactor region of NS2B–NS3 protease. NS2B has one of the longest MoRF regions in Zika virus proteome. In future, this study may provide valuable information while investigating the virus host protein interaction networks. 相似文献

BACKGROUND: The objective of current study was to develop and validate a nomogram to predict overall survival in pancreatic neuroendocrine tumors (PNETs). METHODS: The Surveillance, Epidemiology, and End Results (SEER) database was queried for patients with PNETs between 2004 and 2015. Patients were randomly separated into the training set and the validation set. Cox regression model was used in training set to obtain independent prognostic factors to develop a nomogram for predicting overall survival (OS). The discrimination and calibration plots were used to evaluate the predictive accuracy of the nomogram. RESULTS: A total of 3142 patients with PNETs were collected from the SEER database. Sex, age, marital status, primary site, TNM stage, tumor grade, and therapy were associated with OS in the multivariate models. A nomogram was constructed based on these variables. The nomogram for predicting OS displayed better discrimination power than the Tumor-Node-Metastasis (TNM) stage systems 7th edition in the training set and validation set. The calibration curve indicated that the nomogram was able to accurately predict 3- and 5-year OS. CONCLUSIONS: The nomogram which could predict 3- and 5-year OS were established in this study. Our nomogram showed a good performance, suggesting that it could be served as an effective tool for prognostic evaluation of patients with PNETs. 相似文献

Fourteen West Tatra lakes were studied, of which one could be considered to be recently anthropogenically acidified and eight
others classified as acidification-endangered. In the anthropogenically acidified lake, the zooplankton assemblage has been
substantially altered (three mountain-lake crustacean species have been eliminated). Several littoral macrobenthic species
sensitive to acidification have either been eliminated from the acidified and acidification-endangered lakes or occur only
sporadically. The effect of acidification has so far not been observed on the benthic fauna of the lakes medial which is probably
due to the higher pH below the surface of sediment. In comparison with the High Tatra, acid depositions have had a less pronounced

The effects of habitat and timing of growing season fires on resprouting of shrubs were studied in second-growth longleaf pine savannas of the west Gulf coastal plain in the southeastern United States. Within the headwaters of three different drainages of the Calcasieu River in the Kisatchie National Forest in western Louisiana, replicated permanent transects were established that extended from xeric upland longleaf pine savannas into downslope hydric seepage savannas. All shrubs were mapped and tagged, and numbers of stems were counted prior to any fires. Replicated prescribed fires were set early (June) and late (August) during the 1990 growing season; maximum fire temperatures were measured within both upland and seepage habitats within each transect. Shrubs were relocated; stems were recensused two and twelve months after the fires. At least some shrubs of all species resprouted from underground organs; none regenerated solely from seed banks in the soil. There was no reduction in total numbers of stems one year after fires compared to before fires, either in the upland or in seepage savannas. In addition, there was no reduction in total numbers of stems one year after early or late growing season fires. Fire-related mortality was restricted to small shrubs (< 18 stems) and was not associated with high fire temperatures. The rate of resprouting varied among species and between habitats. Resprouting occurred more rapidly in seepage than upland savannas, but more resprouts were produced in upland than seepage savannas one year after fires. In contrast to other upland species, Vaccinium arboreum and V. elliottii delayed resprouting more than two months following fire. Stems of Rhus copallina and Pyrus arbutifolia, species with long rhizomes, increased more after fires in June than fires in August. We suggest that growing season fires may block further recruitment of shrubs into longleaf pine savannas, but reduction in numbers of large shrubs may require additional management. 相似文献
