Photosensitized efficacy of tetrasulfonated phthalocyanines of zinc, aluminum and nickel (ZnPcS4, AlPcS4 and NiPcS4, respectively) as studied by gramicidin channel (gA) photoinactivation was compared with adsorption of the dyes on the surface of a bilayer lipid membrane as measured by the inner field compensation method. The adsorption of the negatively charged phthalocyanines on diphytanoylphosphatidylcholine (DPhPC) membranes led to formation of a negative boundary potential difference between the membrane/water interfaces. Good correlation was shown between the photodynamic activity and the membrane binding of the three metallophthalocyanines. ZnPcS4 appeared to be the most potent of these photosensitizers, while NiPcS4 was completely ineffective. All of these phthalocyanines displayed no binding and negligible gA photoinactivation with membranes formed of glycerol monooleate (GMO), whereas Rose Bengal exhibited significant binding and photodynamic efficacy with GMO membranes. Gramicidin photoinactivation in the presence of AlPcS4, being insensitive to the ionic strength of the bathing solution, was inhibited by fluoride and attenuated by phosphate ions. A blue shift of the fluorescence peak position of ZnPcS4 dissolved in ethanol was elicited by phosphate, similarly to fluoride, which was indicative of the coordination interaction of these ions with the central metal atom of the phthalocyanine macrocycle. This interaction was enhanced in the medium modeling the water-membrane interface. The results obtained imply that binding of tetrasulfonated metallophthalocyanines to phospholipid membranes is determined primarily by metal-phosphate coordination.  相似文献

Two experiments investigate the effects of adrenalectomy on the capacity of male mice of the BDF1 genotype (C57B1/6Fa♀×DBA/2♂) to retain sexual behaviour following castration. Adrenalectomy is without effect and it is suggested that androgens can play no part in the maintenance of sexual behaviour in these castrates.  相似文献

The participation of H-2I gene products in generating virus-specific proliferative and/or cytotoxic T-lymphocyte (CTL) responses was investigated. Spleen cells from mice infected with vaccinia virus were restimulated secondarily in vitro with syngeneic virus-infected peritoneal exudate cells (PEC) and then restimulated in tertiary cultures with virus-infected PEC from syngeneic and partially histoincompatible strains of mice. Based on the finding that comparable proliferative responses resulted when stimulating the responding cells were histocompatible at the H-2K, I, or D region of the major histocompatibility complex (MHC), the additively enhanced, but not potentiated, proliferation caused by identity at two or three H-2 regions was analyzed. Enhancement of proliferation followed when the H-2K/D components plus virus and the H-2I components plus virus were either on the same, or alternatively on two, stimulating cells. This suggests that H-2K, D, and I plus virus trigger distinct T-cell subsets. A virus-specific CTL response was generated in vitro when spleen cells from virus-primed mice and even unprimed mice were stimulated with cells sharing only H-2K and/or H-2D of the MHC. Identity at the H-2I region did not stimulate a CTL response, nor did it influence the magnitude of the KD restricted response. Nevertheless, the presence of anti-Ia antiserum in cultures of syngeneic stimulators and responders inhibited the cytotoxic response to a great extent. Therefore, H-2I region products seem to participate in the generation of virus-specific CTL in vitro.  相似文献

Distribution of mobile organisms on near-continent islands is mainly shaped by factors operating over ecological rather geological time. However, the phylogeography of single species has the potential to expose historical factors at work. In the present study, West Mediterranean populations of the butterfly Maniola jurtina are studied using geometric morphometrics. The distribution of the two well established lineages ( Maniola jurtina jurtina in the Atlanto–Mediterranean area and Maniola jurtina janira in the Central–Eastern-Mediterranean area) on 12 islands and the adjoining continents are compared. The south-western lineage unexpectedly occurs on islands close to shores occupied by the eastern lineage. We have modelled the distribution of the lineages using three different hypotheses: (1) a contemporary isolation model, which predicts lineage occupancy of islands is linked to relative distances from neighbouring continental areas; (2) a refugial hypothesis, which predicts one lineage to be the ancestral one for the whole region studied, and then successively replaced over part of it; (3) a changing geography hypothesis, which predicts the two lineages to have evolved in their currently occupied areas, continuously sourcing islands subsequent to the Würm maximum glaciation. Of the three models, the refugial hypothesis is most highly correlated with the observed pattern, suggesting that Mediterranean islands may function as refugia during cold periods, much as the three mainland peninsulas of Iberia, Italy and Greece are known to have done. In this study, the cells producing T-cell growth factor (TCGF) in the rat MLR were characterised with respect to the antigens defined by W313, W325, and OX8 monoclonal antibodies. Unfractionated lymphocytes and cells depleted of OX8 positive cells were found to be fully capable of producing TCGF, whereas lymphocytes depleted of W313andW325 positive cells had lost this ability. Parallel experiments demonstrated that cells selected by the fluorescence-activated cell sorter for the expression of W313andW325 defined antigens were potent producers of TCGF. Further studies suggested a functional role for the antigen defined by W325 antibody because the addition of this antibody to a MLR abrogates TCGF production. These findings suggest that the important immunomodulatory functions of W325 positive lymphocytes could be exercised via the synthesis of essential lymphokines.  相似文献

ABSTRACT. Using the polymerase chain reaction and arbitrarily selected oligonucleotide primers of 10 or 11 bases, we have amplified DNA sequences from Trypanosoma vivax parasites isolated from South America and Africa. On the basis of polymorphisms in the DNA fingerprints generated by three of the primers, the parasites could be separated into two major groups, one comprising T. vivax isolates from Kenya and the second including all the other T. vivax parasites (from Colombia, The Gambia, Nigeria and Uganda). One of these three primers (ILo 525) also gave isolate-specific DNA fingerprints for the parasites for the parasites tested, which will allow the use of this technique both in the species identification and discrimination of T. vivax parasites.  相似文献

Lactoperoxidase-catalyzed cell surface radioiodination was used to study the size distribution and physiological characteristics of cell surface proteins of Cloudman melanoma cells maintained in vivo (An cell) or in vitro (TC cells). Slight qualitative and quantitative differences were observed between detergent-extracted radiolabeled membrane proteins obtained from An and TC cells when the proteins were resolved by polyacrylamide gel electrophoresis in sodium dodecyl sulfate. Striking differences were observed in the membrane residence times of radioiodinated proteins of the An and TC cells when labeled cells were cultured 24 hr. Many labeled surface proteins are lost more rapidly from An cells than from TC cells. The possibility that the release of surface proteins from tumor cell membranes may be an important factor which determines the nature of the host immune response against incipient cancers is considered.  相似文献

Abstract: Estimating components of detection probability is crucial to improving the design of aerial surveys for wildlife populations, and this is especially true for species of marine mammals that are threatened or endangered. To evaluate the probability that Florida manatees (Trichechus manatus latirostris) will be detected by observers during aerial surveys, we conducted 6 series of survey flights, during mornings and afternoons on 14-16 consecutive days over the Tampa Electric Company's (TECO) Big Bend power plant discharge canal in Tampa Bay, Florida, USA (winter 2000 through 2003). Our objective was to understand how our ability to detect manatees at a winter aggregation site affects aerial survey counts, so that we may improve techniques for estimating manatee population size. We estimated the probability that manatees would be present at the warm-water discharge of the plant during winter cold fronts and estimated the overall detection probability of manatees present at the plant and the 2 components that make up the probability of detection (the probability of being available and the probability of being detected given they are available). We used telemetry tags and marker flags (n = 15) to facilitate capture-recapture analyses. The probability that marked manatees would be at the plant varied from 48% to 68% across flight series and was inversely related to the ambient water temperature. Based on sightings of marked animals, estimates of the overall probability of detecting a manatee ranged from 45% to 69% across flight series (x̄ = 58%, n = 6). The probability that a manatee would be available to an observer ranged from 73% to 94% across flight series (x̄ = 83%) but was constant among years (83%, 81%, and 78%; x̄ = 81%). The probability that an available manatee would be detected by an aerial observer was variable across flight series (55-95%) and years (73%, 86%, and 66%, x̄ = 73%). Independent estimates of the probability that a manatee would be available to the observer on one pass were obtained from time-depth data loggers and ranged from 5% to 33% (x̄ = 19%, SE = 3.7%), and the probability that a manatee would be available during ≥1 of 10 passes ranged from 41% to 98% (x̄ = 88%, 95% confidence bounds 0.71-0.95). We adjusted survey counts using measures of detectability. Although corrected counts presented here are site-specific, adjusting counts based on detection probability will greatly improve reliability of population estimates from all aerial surveys. Special sampling to estimate components of detection probability should be built into all aerial surveys to ensure that reliable and unbiased information on species abundance is used to evaluate wildlife populations.  相似文献
