Summary Investigation of 7 insulin sequences from a bony fish, a bird, and 5 mammalian species showed that guinea pig and coypu insulin, that have a strongly divergent primary and quarternary structure, are not the result of gene duplication in an ancient vertebrate or invertebrate ancestor but that they diverged from the other mammals after divergence of the mammals from the other vertebrates. After this divergence both insulins underwent evolution at a highly increased rate.  相似文献

Aim We investigate the biogeography of Austral Polychaeta (Annelida) using members of the families Eunicidae, Lumbrineridae, Oenonidae, Onuphidae, Serpulidae and Spionidae and Parsimony Analysis of Endemicity (PAE). We determine whether observed polychaete distribution patterns correspond to traditional shallow-water marine areas of endemism, estimate patterns of endemism and relationships between areas of endemism, and infer the biological processes that have caused these patterns. Location The study is concerned with extant polychaete taxa occupying shallow-water areas derived from the breakup of the Gondwana landmass (i.e. Austral areas). Methods Similarity was assessed using a significance test with Jaccard's indices. Areas not significantly different at 0.99 were combined prior to the PAE. Widespread species and genera (155 taxa) were scored for presence/absence for each area of endemism. PAE was used to derive hypotheses of area relationships. Hierarchical patterns in the PAE trees were identified by testing for congruence with patterns derived from cladistic biogeographic studies of other Gondwanan taxa and with geological evidence. Results The polychaete faunas of four area-pairs were not significantly different and the areas amalgamated: South-west Africa and South Africa, New Zealand South Island and Chatham Islands, Macquarie Island and Antipodean Islands, and West Antarctica and South Georgia. Areas with the highest levels of species endemism were southern Australia (67.0%), South-east South America (53.2%) and South Africa (40.4%). About 60% of species and 7.5% of genera occupied a single area of endemism. The remainder were informative in the PAE. Under a no long-distance dispersal assumption a single minimal-length PAE tree resulted (l=367; ci=0.42); under dispersal allowed, three minimal-length trees resulted (l=278; ci=0.56). In relation to the sister grouping of the New Zealand areas and Australia we find congruence between our minimal-length trees and those derived from a biogeographic study of land plants, and with area relationships predicted by the Expanding Earth Model. Main conclusions The polychaete distribution patterns in this study differ slightly from the classical areas of endemism, most notably in being broader, thereby bringing into question the value of using single provincial system for marine biogeographic studies. The Greater New Zealand region is found to be ‘monophyletic’ with respect to polychaetes, that is comprising a genuine biogeographical entity, and most closely related to the polychaete fauna of southern Australia. This finding is consistent with studies of land plants and with the Expanding Earth model, but disagrees with conventional geology and biogeographic hypothesis involving a ‘polyphyletic’ New Zealand. Both vicariance and concerted range expansion (=biotic dispersion) appear to have played important roles in shaping present-day distribution patterns of Austral polychaetes. Shallow-water ridge systems between the Australian and Greater New Zealand continental landmasses during the Tertiary are thought to have facilitated biotic dispersion.  相似文献

In vivo muscle forces are typically estimated using literature-based or subject-specific moment arms (MAs) because it is not possible to measure in vivo muscle forces non-invasively. However, even subject-specific muscle-tendon MAs vary across contraction levels and are impossible to determine at high contraction levels without techniques that use ionized radiation. Therefore, different generic MA functions are often used to estimate in vivo muscle forces, which may alter force predictions and the shape of the muscle’s force-length relationship. The aim of this study was to examine the influence of different literature-based patella tendon MA functions on the vastus lateralis (VL) force-angle relationship. Participants (n = 11) performed maximum voluntary isometric knee extension contractions at six knee flexion angles, ranging from 40° to 90°. To estimate in vivo VL muscle force, the peak knee extension torque at each joint angle was multiplied by the VL’s physiological cross-sectional area (PCSA) relative to the quadriceps’ PCSA (34%) and then divided by the angle-specific patella tendon MA for 19 different functions. Maximum VL force was significantly different across MA functions (p ≤ 0.039) and occurred at different knee flexion angles. The shape of the VL force-angle relationship also differed significantly (p < 0.01) across MA functions. According to the maximum force generated by VL based on its literature-derived PSCA, only the VL force-angle relationships estimated using geometric imaging-based MA functions are feasible across the knee angles studied here. We therefore recommend that an average of these MA functions is calculated to estimate quadriceps muscle forces if subject-specific MAs cannot be determined.  相似文献

A record of simulated removal trapping of 50 samples drawn from N=50 under a design has been considered as to how it does contribute to practical censuses of small mammals. It follows that, for ordinary-density populations so far experienced, the removal census based on the theory may frequently lead to quite wrong estimates from single-plot data by the simplest method, and that the maximum likelihood method is doubtless better, but even this method may produce right estimates in about half of the cases, their standard error being of limited availability. Therefore pooling of data from many plots are desirous for such populations.  相似文献

ABSTRACT

In the biopharmaceutical environment, controlling the Critical Quality Attributes (CQA) of a product is essential to prevent changes that affect its safety or efficacy. Physico-chemical techniques and bioassays are used to screen and monitor these CQAs. The higher order structure (HOS) is a CQA that is typically studied using techniques that are not commonly considered amenable to quality control laboratories. Here, we propose a peptide mapping-based method, named native peptide mapping, which could be considered as straightforward for HOS analysis and applicable for IgG4 and IgG1 antibodies. The method was demonstrated to be fit-for-purpose as a stability-indicating assay by showing differences at the peptide level between stressed and unstressed material. The unfolding pathway induced by a heat stress was also studied via native peptide mapping assay. Furthermore, we demonstrated the structure–activity relationship between HOS and biological activity by analyzing different types of stressed samples with a cell-based assay and the native peptide mapping. The correlation between both sets of results was highlighted by monitoring peptides located in the complementary-determining regions and the relative potency of the biotherapeutic product. This relationship represents a useful approach to interrogate the criticality of HOS as a CQA of a drug.  相似文献

The use of sodium bisulfate for field preservation of soil samples collected for the analysis of volatile organics (VOCs) can result in the formation of acetone. The work was performed at Camp Edwards, Massachusetts, as part of a multi-year investigation of the effects of training activities on the local environment. Preservation with sodium bisulfate was initiated in 1999 under regulatory mandate. The frequency of acetone detection and the quantities reported increased immediately thereafter. Correlation of duplicate sample results and a side-by-side comparison of six sample handling methods involving two analytical laboratories suggest the interaction of sodium bisulfate with naturally occurring organic matter in samples from Camp Edwards cause the formation of acetone. The data also indicate that a higher content of natural organic matter results in a greater production of acetone, and more acetone is formed the longer the sodium bisulfate is in contact with the soil.  相似文献

This paper extends the work of KODLIN (1967), who proposed a method for analyzing patient survival data wherein the hazard rate was linearly related to the survival time. The present paper extends Kodlin's model to permit maximum likelihood estimation of the parameters so that covariate effects are included and the slope and intercept parameters are allowed to change over fixed intervals of the time domain of study. An illustration of the method using multiple myeloma data is given and the results are compared with those of Kodlin's model, the Feigl-Zelen, Zippin-Armitage model, the exponential model, and Cox's proportional hazards model.  相似文献

We present methods for solving and making statistical inferences about marginal attack rates based on observed death rates for contemporaneous mortality factors. The general method of solution involves solving a system of nonlinear equations which depend in part on competition coefficients that express the outcome when more than one agent attacks the same host individual. For two factors, we present a detailed analysis of the effect of varying this competition coefficient. Statistical inferences are illustrated using standard large sample approximations (the delta method) and the bootstrap, which is a resampling technique. We also extend the results to allow inferences for k-values.  相似文献
