Induced differentiation in three human cell lines altered their sensitivity specifically to human natural killer (NK) cells by affecting their expression of NK target antigens. Differentiation of HL-60, a promyelocytic leukemia cell line, and the erythroleukemic cell line K562 was accompanied by a concomitant decrease in susceptibility to NK-mediated lysis whereas induction of MeWo melanoma cells resulted in an enhanced sensitivity to lysis. Our findings suggest that target cell susceptibility to NK-mediated lysis may in part be dependent on the stage of differentiation of the tumor cell target.  相似文献

对野败型水稻不育系(珍汕97A)、保持系(珍汕97B)和F1杂种(汕优63)线粒体DNA进行了AFLP比较,从M/PA引物对选择扩增产物中找到了不育系与保持系差异条带ZA1、ZA2和ZA3。Northern分析表明,片段ZA1黄化苗期无转录产物,可能是非编码序列;而ZA2、ZA3两片段在不育系、保持系和F1杂种中的转录则有差异,其中片段ZA2在黄化苗期保持系转录,不育系和F1杂种无转录产物;片段ZA3在黄化苗期保持系和F1杂种转录,表明ZA3转录受核恢复基因Rf影响.  相似文献

The spleens of mice bearing transplanted methylcholanthrene (MCA)-induced fibrosarcomas (MCA-1425 and MCA-1460) were shown to contain cells capable of suppressing the generation of cytolytic T lymphocytes (CTL) in mixed leukocyte cultures (MLC). The suppressive activity was first detected 21 days after tumor transplantation. No suppression was seen with lymph node cells taken at the same time as the spleen cells. The cells responsible for the suppressive activity were adherent to nylon wool and plastic dishes and they were not lysed by anti-T-cell serum plus complement. The suppressor cells were phagocytic and were resistant to irradiation (3000 rads) in vitro. Spleen cells from tumor-bearing nude mice were as suppressive as were spleen cells from tumor-bearing conventional mice. We conclude from these findings that T cells were not involved either as inducers or as effectors of the suppression observed, although the responsible adherent cells may have exerted their effect by interacting with a T-suppressor cell population in the MLC mixtures. While spleen cells of tumor-bearing mice were suppressive when added at any time during the first 4 days of a 5-day MLC, they showed no effect on the cytotoxicity of fully differentiated CTL. Indomethacin reversed suppression, suggesting that prostaglandins may have been involved.  相似文献

Muscle and adipose tissue were obtained from steers and dairy cows following subcutaneous administration of [14C] progesterone. Following extraction, purification and separation by column, thin layer and gas-liquid chromatography, various radioactive residues from these tissues were identified by their Chromatographic mobility, crystallization to constant specific activity and mass spectra. Progesterone constituted 54% of free radioactivity extracted from muscle and 69 and 73% of radioactivity in the free and conjugated portions of extracts, respectively, from fat. Metabolites identified were: 5α-pregnane-3,20-dione, 9%, 0%, 0%, 20β-hydroxy-4-pregnen-3-one, 8%, 11%, 3%; 3α-hydroxy-5β-pregnan-20-one, 13%, 2%, 2%; 3α-hydroxy-5α-pregnan-20-one, 3%, 3%, 6%; 20α-hydroxy-5α-pregnan-3-one, 0%, 2%, 3%; of radioactivity in muscle (free) and fat (free and conjugated fractions), respectively. Tentatively identified in fat extracts by chromatographic mobility were: 20α-hydroxy-4-pregnen-3-one, 1%, 1% and 3β-hydroxy-5β-pregnan-20-one, 0%, 2% of radioactivity in free and conjugated fractions, respectively. The average concentration of steroid in these animals due solely to treatment, calculated from the specific activity of the [14C] progesterone administered, was 3.4 and 18.1 ng/g in muscle and subcutaneous fat, respectively.  相似文献

The competition-density (C-D) effect for given times and self-thinning over time in even-aged, natural, pure stands of Pinus densiflora Sieb. et Zucc. were analyzed with the reciprocal equation of the C-D effect in self-thinning stands, and the equation describing the time-trajectory of mean stem volume and stand density. The C-D effect and self-thinning were consistently well explained by the two equations. Differences in mean stem volume and in stand density among the stands tended to merge with increasing stand age. The self-thinning line with a slope of approximately –3/2 was reached by the higher density stand prior to the medium and lower density stands. The skewness of tree height distribution showed positive values, which means that the distribution is more or less L-shaped, and in addition the skewness decreased with increasing mean tree height, which indicates that smaller trees died as the stands grew. This trend is consistent with the asymmetric (one-sided) competition hypothesis that self-thinning is driven by competition for light. The tree height distribution was analyzed using the Weibull distribution. The location parameter h min of the Weibull distribution increased with increasing stand age, and the scale parameter a tended to increase slightly with increasing stand age. The range of the shape parameter b of the Weibull distribution corresponded to that of the skewness.  相似文献

Haemonchus contortus is a blood-feeding nematode which parasitizes the abomasum of sheep and represents a serious constraint to sheep production. Anthelmintics are currently the most common method of worm control but the worldwide development of multiple-drug resistance and issues of residues in the food chain make alternatives to anthelmintics a priority. Biotechnology-driven solutions to parasitism include vaccines and silencing of genes regulating nematode development. To pursue gene targets that may be suitable for parasite control, a two stage differential-display PCR (dd-PCR) approach was developed to observe differential gene expression between Haemonchus from immune and control sheep. Twenty-four reproducible differentially-expressed bands were identified in 60 pairs of dd-PCR comparisons. The first of these cloned and sequenced corresponded to the H. contortus 60S ribosomal protein L35A. The remaining bands are being cloned and validated and may provide new targets for parasite control.  相似文献

Cold exposure and β3-adrenergic receptor agonist (CL316,243) treatment induce the production of beige cells, which express brown adipocytes(BA)-specific UCP1 protein, in white adipose tissue (WAT). It remains unclear whether the beige cells, which have different gene expression patterns from BA, express BA-characteristic fatty acid oxidation (FAO) proteins. Here we found that 5 day cold exposure and CL316,243 treatment of WAT, but not CL316,243 treatment of primary adipocytes of C57BL/6J mice, increased mRNA levels of BA-characteristic FAO proteins. These results suggest that BA-characteristic FAO proteins are induced in beige cells in a different pathway from UCP1.  相似文献

A lectin that agglutinates human blood group B erythrocytes but not blood group A and O erythrocytes was isolated from eggs of Ayu sweet fish (Plecoglossus altivelis). The lectin also agglutinates Ehrlich ascites carcinoma cells but not rat ascites hepatoma AH109 or rat sarcoma 150 cells tested. The lectin agglutination was most effectively inhibited by monosaccharides with the first type of configuration, i.e., L-rhamnose, L-mannose and L-lyxose at a concentration of 0.03 mM. The lectin agglutination was moderately inhibited by monosaccharides with the second type of configuration, i.e., D-galactose, D-fucose and D-galacturonic acid at a concentration of 0.4 mM. However, the agglutination was not inhibited by various other monosaccharides and oligosaccharides that have other types of configuration. The basis for an apparent B-specific hemagglutination may be due to the steric similarity of the C2 and C4 of the galactosyl series, the B-specific determinant, and the L-rhamnosyl series, which are the best inhibitors of the lectin activity. The lectin was affinity purified on an L-rhamnosyl-Sepharose column and was characterized as a homogeneous low molecular weight protein (Mr 14 000) with an abundance of hydrophobic amino acids and dicarboxylic amino acid.  相似文献

Intracerebroventricular beta-endorphin increases food intake of rats   总被引:1,自引:0,他引:1  
B-Endorphin (B-END), met-enkepalin (M-ENK), and DAla2NMe5-met-enkephalinamide were administered intracerebroventricularly to rats and effects on the ingestion of a liquid diet were examined. B-END significantly increased food intake in a half-hour test at a dose of 200 ng/rat. Lower or higher doses did not affect food intake. Neither M-ENK or the synthetic enkephalin analog affected ingestion of the liquid diet. These findings demonstrate rapid action of an endorphin on food intake administered at a lower dose than has previously been reported and suggest a specificity for B-END in the endorphinergically mediated hyperphagic response.  相似文献
