The volvocalean microalga Haematococcus pluvialis is used as a sourceof the ketocarotenoid astaxanthin for applications in aquaculture and thepharmaceutical and cosmetic industries. This green alga accumulatesastaxanthin, mostly esterified, canthaxanthin and echinenone in lipid vesiclesoutside the chloroplast. This accumulation process normally but notexclusively accompanies formation of the resting state in the developmentalcycle. With regard to increased bioavailability of the accumulated secondarycarotenoids, the fragility of the extracellular matrix makes the flagellatedstate of H. pluvialis an interesting alternative to the thick-walledaplanospore state. A two-step batch cultivation scheme was developed thatleads to accumulation of secondary carotenoids in flagellated cells of H. pluvialis (strain 192.80, Göttingen, Germany). Germination ofgreen aplanospores during the first step of cultivation proceeded optimallyunder 30 mol photon m-2 s-1 of whitefluorescent light at 20 °C. For optimal induction and enhancementof carotenoid biosynthesis, the flagellated cells formed were then exposedto a decreased level of nitrate (0.4 mM KNO3) and to enhancedirradiance (150 mol photon m-2 s-1). Under theseconditions, which still permitted cell division and chlorophyll synthesisduring the first two days of exposure, carotenoid accumulation in theflagellated cells reached 2° of dry mass at the fourth day of exposure. Asa mixotrophic carbon source, addition of acetate at a concentration nothigher than 10 mM increased carotenoid synthesis only slightly whereaspartial or complete phosphate deficiency or salt stress (40 mM NaCl) didnot. 相似文献

Four genera of the Phacotaceae (Phacotus, Pteromonas, Wislouchiella, Dysmorphococcus), a family of loricated green algal flagellates within the Volvocales, were investigated by means of transmission electron
microscopy and analysis of the nuclear encoded small-subunit ribosomal RNA (18S rRNA) genes and the plastid-encoded rbcL genes.
Additionally, the 18S rDNA of Haematococcus pluvialis and the rbcL sequences of Chlorogonium elongatum, C. euchlorum, Dunaliella parva, Chloromonas serbinowii, Chlamydomonas radiata, and C. tetragama were determined. Analysis of ultrastructural data justified the separation of the Phacotaceae into two groups. Phacotus, Pteromonas, and Wislouchiella generally shared the following characters: egg-shaped protoplasts, a single pyrenoid with planar thylakoid double-lamellae,
three-layered lorica, flagellar channels as part of the central lorica layer, mitochondria located in the central cytoplasm,
lorica development that occurs in mucilaginous zoosporangia that are to be lysed, and no acid-resistant cell walls. Dysmorphococcus was clearly different in each of the characters mentioned. Direct comparison of sequences of Phacotus lenticularis, Pteromonas sp., Pteromonas protracta, and Wislouchiella planctonica revealed DNA sequence homologies of ≥98.0% within the 18S gene and 93.9% within the rbcL gene. D. globosus was quite different from these species, with a maximum of 92.9% homology in the 18S rRNA and ≤86.6% in the rbcL gene. It
showed major similarities to the 18S rDNA of Dunaliella salina, with 95.3%, and to the rbcL sequence of Chlamydomonas tetragama, with 90.3% sequence homology. Additionally, the Phacotaceae sensu stricto exclusively shared 10 (rbcL: 4) characters which were present neither in other Chlamydomonadales nor in Dysmorphococcus globosus. Different phylogenetic analysis methods confirmed the hypothesis that the Phacotaceae are polyphyletic. The Phacotaceae sensu stricto form a stable cluster with affinities to the Dunaliellaes and possibly Haematococcus pluvialis. Dysmorphococcus globosus represented an independent lineage that is possibly related to Chlamydomonas moewusii and C. tetragama.

formation.
Male and female gametes fused after being induced by changing the medium. The anterior end, including the flagellar base,
of the male gamete entered the anterior region of the female gamete. Fusion of the two protoplasts proceeded laterally and
posteriorly. The male gamete bore a slender cytoplasmic protrusion at the base of the flagella. This structure has not been
previously described in the male gamete ofEudorina, and may participate in plasmogamy.
A biflagellate gone cell swam from the germinating zygote and secreted a gelatinous envelope. It then divided to form a gone
colony within the gelatious envelope, which moved during colony formation by means of the two flagella which were retained

Group I introns were reported for the first time in the large subunit of Rubisco (rbcL) genes, using two colonial green algae, Pleodorina californica and Gonium multicoccum (Volvocales). The rbcL gene of P. californica contained an intron (PlC intron) of 1320 bp harboring an open reading frame (ORF). The G. multicoccum rbcL gene had two ORF-lacking introns of 549 (GM1 intron) and 295 (GM2 intron) base pairs. Based on the conserved nucleotide sequences of the secondary structure, the PlC and GM1 introns were assigned to group IA2 whereas the GM2 intron belonged to group IA1. Southern hybridization analyses of nuclear and chloroplast DNAs indicated that such intron-containing rbcL genes are located in the chloroplast genome. Sequencing RNAs from the two algae revealed that these introns are spliced out during mRNA maturation. In addition, the PlC and GM1 introns were inserted in the same position of the rbcL exons, and phylogenetic analysis of group IA introns indicated a close phylogenetic relationship between the PlC and GM1 introns within the lineage of bacteriophage group IA2 introns. However, P. californica and G. multicoccum occupy distinct clades in the phylogenetic trees of the colonial Volvocales, and the majority of other colonial volvocalean species do not have such introns in the rbcL genes. Morphological details of sexual reproduction in Carteria eugametos (Volvocales, Chlorophyta) were studied under controlled laboratory conditions. Protoplasts of the two pairing, flagellate cells were released from cell walls to become isogametes. Such gametes were nonmotile and soon fused to form a completely immobile zygote. The zygote then secreted a cell wall to enter the dormant period. After dark treatment, the zygote produced four, eight or 16 quadriflagellate germ cells, and a transparent vesicle enclosing all the germ cells was released from the zygote wall. This type of zygote germination and aplanogamy in C. eugametos is unique and may be related to its peculiar phylogenetic position within the Volvocales (Chlamydomonadales). 相似文献

Volvox barberi W. Shaw is a volvocalean green alga composed of biflagellated cells. Vovocales with 16 cells or more form spherical colonies, and their largest members have germ‐soma separation (all species in the genus Volvox). V. barberi is the largest Volvox species recorded in terms of cell number (10,000–50,000 cells) and has the highest somatic to reproductive cell ratio (S/R). Since they are negatively buoyant, Volvocales need flagellar beating to avoid sinking and to reach light and nutrients. We measured V. barberi swimming speed and total swimming force. V. barberi swimming speeds are the highest recorded so far for volvocine algae (~600 μm · s?1). With this speed, V. barberi colonies have the potential to perform daily vertical migrations in the water column at speeds of 2–3 m · h?1, consistent with what has been reported about Volvox populations in the wild. Moreover, V. barberi data fit well in the scaling relationships derived with the other smaller Volvox species, namely, that the upward swimming speed Vup∝N0.28 and the total swimming force FS∝N0.77 (N = colony cell number). These allometric relationships have been important supporting evidence for reaching the conclusion that as size increases, colonies have to invest in cell specialization and increase their S/R to increase their motility capabilities to stay afloat and motile. 相似文献

Phylogenetic analyses of 19 strains representing five species of Eudorina, one strain of Pleodorina indica, and seven strains of Yamagishiella unicocca were carried out by sequencing the internal transcribed spacer region (ITS 1 and ITS 2) of the nuclear ribosomal DNA (rDNA) repeats. The sequence data resolved five phylogenetic groups, one consisting of Y. unicocca and the other four encompassing all the Eudorina species. Two isolates, Eudorina sp. (ASW 05157) and Pleodorina indica (ASW 05153), were of uncertain affiliation. Whereas one monophyletic group included strains of E. elegans only, the other strains of E. elegans appeared alongside E. cylindrica, E. illinoisensis, and E. unicocca var. unicocca in the other Eudorina clades. The distribution pattern of the carotenoid loroxanthin ([3R,3'R,6'R]-β,ε-carotene-3,19,3'-triol), a systematically useful biochemical marker within chlorophycean flagellates, was shown to match the evaluated molecular data. Whereas it was either totally absent or universally present in six of the deduced phylogenetic lines, it occurred randomly in the E. elegans clade containing only E. elegans isolates. The results substantiated the current hypothesis that the unique vegetative morphology of E. elegans has independently arisen at various times during evolution and that it is not a marker of a monophyletic group. 相似文献

The unicellular green alga Chlamydomonas reinhardtii has two actin genes, one encoding a conventional actin (90% amino acid identity with mammalian actin), the other a highly divergent actin (64% identity) named novel actin-like protein (NAP). To see whether the presence of conventional and unconventional actins in a single organism is unique to C. reinhardtii, we searched for genomic sequences related to the NAP sequence in several other species of volvocalean algae. Here we show that Chlamydomonas moewusii and Volvox carteri also have, in addition to a conventional actin, an unconventional actin similar to the C. reinhardtii NAP. Analyses of the deduced protein sequences indicated that the NAP homologues form a distinct group derived from conventional actin. 相似文献
