Summary Anthers and ovaries of six grapevine cultivars (three Vitis vinifera L., two V × Labruscana L. H. Bailey, and one complex hybrid) were extracted from flower buds over 2 yr and cultured on three media reported to promote somatic embryogenesis in Vitis tissues. The highest percent embryogenesis from the hybrid ‘Chancellor’ and V. vinifera ‘Chardonnay’, ‘Merlot’, and ‘Pinot Noir’ occurred on medium C [Nitsch and Nitsch, 1969, basal medium with 3.0% (w/v) sucrose, 0.01% (w/v) inositol. 0.3% (w/v) Phytagel, 2.5 μM 2.4-dichlorophenoxyacetic acid, 2.5μM β-naphthoxyacetic acid, 5.0μM N-(2-chloro-4-pyridyl)-N′-phenylurea, and 0.05% (w/v) glutamine]. Regardless of the media, the labrusca cultivars ‘Concord’ and ‘Niagara’ produced soft non-embryogenic callus that was sometimes mixed with well-developed somatic embryos. Nine vinifera genotypes were further tested for several different years on medium C. Embryogenic cultures suitable for transformation were obtained from all genotypes in more than 1 yr. The average percent embryogenesis from ovaries was 7-fold higher than from anthers. There was significant annual variation in percent embryogenesis, demonstrating the need for media comparisons to be replicated for more than one season. Suspension cultures suitable for use in genetic transformation were initiated from ‘Chardonnay’, ‘Merlot,’ and ‘Pinot Noir’ pro-embryogenic masses. ‘Chardonnay’ suspension cultures plated and grown under conditions developed for recovery of plants after biolistic transformation yielded approximately 500 non-transformed embryos per plate after 4 mo. of culture, with 68.6% of the embryos converting to plants. This is the first reported protocol for embryogenesis from ‘Concord,’ ‘Cabernet Franc,’ and ‘Pinot Noir’ grapevines.  相似文献

This study shows the results of Vitis vinifera pollen season in the atmosphere of the Braga region (Portugal) for the last 5 years (1999–2003). The fluctuations in the airborne pollen concentration, the synchronic rhythms in the annual pollen trends and their relationship to the main meteorological parameters were investigated. The vineyard pollen season extends from late May to early July, presenting always a bimodal pattern. During the studied period several pollen types such as Poaceae, Castanea, Olea, Urticaceae, Quercus, Plantago, Rumex, Asteraceae, Pinaceae, Ericaceae and Eucalyptus were also observed. Airborne pollen concentration is highly influenced by mean temperature, rain, number of days with rain and wind direction.  相似文献

The wild grapevine, Vitis vinifera L. subsp. silvestris (Gmelin) Hegi, is considered to be an endangered taxon in Europe, mainly as a consequence of the introduction of pathogens from North America and of the destruction of its habitat. In the Rhine Valley, nearly all populations disappeared due to river management, the intensi.cation of forestry, and the introduction of phylloxera. After a growing awareness of the need to preserve endangered forest ecosystems, attempts to reintroduce wild grapevine in the Rhine Valley were performed, particularly in the French nature reserves Erstein and Offendorf since 1992. However, regular surveys of the plants indicate the rapid decline of the populations. In 2002, we proposed to summarise the knowledge accumulated after 10 years of experiments. Results indicate that from the initial 91 individuals planted in 1992, only 14 survived in 2002 (2 in Erstein, 12 in Offendorf). The failure of the experiment may be explained by several factors: unsuitable sites (too shady, absence of support for the young plants), absence of monitoring, vandalism or predation. According to these results and recent knowledge of the ecology of the plant and of vines in general, new transplantation experiments are proposed in which the plants will be monitored during their establishment in the forests. The success of this second transplant (50 plants per reserve) will be enhanced by restoration projects of the Rhine River dynamics, with partial re-.ooding. Floods should help to avoid, or at least to reduce, pest and disease expansion on future adult plants.  相似文献

A procedure is described for obtaining allometric regression equations to estimate non-destructively and in a cost-effective manner the current year's above-ground vegetative and reproductive biomass of Vitis vinifera L. Merlot' throughout the growing season. Significant relationships were obtained over a 3-year period (1998-2000) between the dimensions of an individual shoot per vine (i.e. diameter and length) and dry weights of its primary stem, primary leaves and lateral growth. The dry mass of a grape was best estimated from measurements of the basal diameter of the bunch peduncle. Introducing cumulative degree-days as an additional explanatory variable in the equations allowed them to be used irrespective of year and growth stage. Multi-year regressions were used to quantify in detail the seasonal evolution of mature grapevine biomass under the climatic conditions of the Bordeaux area, France, and for differing levels of soil nitrogen.  相似文献

In vivo feeding experiments with [5,5-(2)H(2)]mevalonic acid lactone (MVL) and [5,5-(2)H(2)]-1-deoxy-D-xylulose (DOX) indicate that the novel mevalonate-independent 1-deoxy- D-xylulose 5-phosphate/2C-methyl- D-erythritol 4-phosphate (DOXP/MEP) pathway is the dominant metabolic route for monoterpene biosynthesis in grape berry exocarp and mesocarp and in grape leaves. The highly uneven distribution of the monoterpene alcohols (3S)-linalool and geraniol between leaves, berry exocarp and berry mesocarp can be attributed to a compartmentation of monoterpene metabolism. In grape berries incorporation of [5,5-(2)H(2)]-DOX into geraniol is mainly restricted to the exocarp, whereas (3S)-linalool biosynthesis can be detected in exocarp as well as in mesocarp tissue. The results demonstrate that grape berries exhibit an autonomic monoterpene biosynthesis via the novel DOXP/MEP route throughout the ripening process.  相似文献

Biosynthesis of both ascorbic acid (AsA) and peroxidase activity were induced by light in cv. Sultana grapevine leaves. Induced peroxidase activity mainly involved basic isoenzymes of pI 9.8 and 9.6 and catalyzed the oxidation of flavonoids like quercetin and kaempferol and derivatives of hydroxycinnamic acids such as ferulic and p-coumaric acids, but not AsA. However, the peroxidase-dependent oxidation of ferulic acid and quercetin was temporarily suppressed by AsA as long as it remained in the reaction medium. Kinetics and spectroscopic results indicated that AsA was oxidized to dehydroascorbic acid only in the presence of phenols or flavonoids, and did not interfere with the catalytic activity of the peroxidase. Ascorbate peroxidase isoenzymes (APx), whose activities are widely considered central for detoxification of H(2)O(2) in most plant cells, were not detected in grape leaves extracts. The significance of light stimulus on peroxidase activity and leaf AsA content is discussed in terms of a flavonoid-redox cycle proposed as an alternative system to detoxify H(2)O(2) in grapevine leaves.  相似文献

Three EST-derived microsatellite loci from Vitis vinifera were amplified and sequenced across eight species of Vitaceae from four different genera. Phylogenetic analysis of the microsatellite’s flanking regions produced informative results in congruence with previous studies. Generic relationships were respected and the data produced sufficient inter-specific variation to distinguish between Cayratia acris and Cayratia saponaria, two very closely related species. Overall, the sequence alignments showed that priming sites were conserved, whereas microsatellite repeats were present in most cases but structurally variable. The sequence data provided information on the evolutionary patterns of various microsatellite repeats and their correlation to evolutionary relationships among taxa. Downwardly-growing grapevine shoots have smaller and more frequent vessels than upwardly-growing ones and, as a consequence, a lower hydraulic conductivity. Here, grapevine (Vitis vinifera L.) shoot growth orientation was manipulated to test whether downward shoot orientation negatively affects vessel growth in the apex via a shortage of water and nutrients. The orientation of the central vine shoot portion was inverted by two consecutive 135 degrees bends, resulting in double-bent N-shaped vines; the central downward shoot portion was of different lengths in the experimental treatments to induce increasing reductions of shoot conductivity. These treatments reduced shoot conductivity and water flow, but had no effects on vessel development and frequency in the apex. In a second experiment, auxin concentration was assessed in shoots of upwardly- and downwardly-growing plants. IAA concentration at the apical internodes was higher in downwardly-oriented shoots than in shoots growing upwards. In addition, a higher density and a lower vessel diameter were observed in the lower, than the upper side, of the downwardly-oriented shoot, suggesting increased accumulation of auxin in the lower side. These results suggest that the downward orientation induces accumulation of auxin in the apex, which in turn affects the density and the size of the xylem vessels, causing reduction of hydraulic conductivity.  相似文献

A reliable and efficient system for transformation and regeneration of 'Chardonnay' (Vitis vinifera L.) plants via microprojectile bombardment was developed. Improvements over the previous biolistic transformation system included: (1) the use of gold particles for bombardment; (2) step-wise selection at 10 then 15 mg/l kanamycin; and (3) embryo induction at 27°C. Embryogenic cell cultures were either bombarded with pBI426, which contains the reporter gene gus (uidA) coding for -glucuronidase (GUS), or were co-bombarded with pSAN237 carrying the npt-II (neomycin phosphotransferase II) selectable marker gene, and a second plasmid with an antimicrobial peptide gene. A large number of transient (7,883±1,928) and stable (46±32) blue spots per plate at 2 and 95 days after bombardment, respectively, were obtained according to GUS expression analyses. A total of 447 putative transgenic embryos was harvested from 84 bombarded plates. From these embryos, 242 (54%) were regenerated into plants within the first year of the experiment. Southern blot analyses confirmed integration of the transgenes into the grape genome. Co-transformation was tested with four separate antimicrobial constructs. The co-transformation frequency of unlinked genes was 48% as measured by polymerase chain reaction (PCR), and 56% as estimated by dot blot hybridization. Range expansions through human introductions have increased with global commerce and have led to the extinction of native species, alterations in community structure and pest status of the invasive species. Inferring the evolutionary history of invasive species can help to build a firmer footing for management tactics. This study used mitochondrial DNA (mtDNA) sequence comparisons of samples collected from the native and introduced ranges of a pest herbivore of cultivated grapes, grape phylloxera (Daktulosphaira vitifoliae Fitch, Phylloxeridae) to infer the sources and pattern of introductions into worldwide viticulture. Introductions into viticulture from its native North American range first occurred in the mid-19th century. The pattern of spread has suggested a focus of introduction into France, but independent introductions may have occurred elsewhere. The results show that the introduced population represents a limited subsample of the native genetic diversity. The data suggest that most grape phylloxera in viticulture, including all European, have originated in the northeastern USA where the grape species Vitis riparia dominates. There was evidence for independent introductions into South Africa and California. Most California haplotypes were most closely related to native grape phylloxera from the Atlantic Coast on V. vulpina. It is likely that subsequent spread from California into Australia, New Zealand and Peru has occurred.  相似文献
