Abstract Phytophthora cinnamomi (cinnamon fungus) is a pathogenic soil fungus that infects plant communities along the southeastern coast of Australia and the southwestern corner of Western Australia. Infection of native plant communities with P. cinnamomi regularly leads to dramatic changes in both the structural and the floristic characteristics of these communities. This study aimed to assess the effect of P. cinnamomi induced changes in plant community attributes upon the abundance and diversity of invertebrates in an area of open forest in the Brisbane Ranges, Victoria. Pitfall trapping was conducted continuously for 1 week within each season over a 3 year period. Invertebrates were sorted to order level, and abundances were compared between times (season and year) and infection status for taxa with normalized distributions (Coleoptera, Collembola, Dermaptera and ants). Non-parametric comparisons were made for other groups (Aranae, Blattodea, Diptera, Hemiptera, Orthoptera, ant ‘morpho-species’ and unidentified larvae) to assess differences on the basis of infection status. Significant differences on the basis of infection were uncommon and, where identified, elevated abundances were more commonly observed at sites infected by P. cinnamomi. Consistent temporal effects (season and year) were observed in normalized data sets. Abundances both within individual taxa and from pooled counts were generally weakly associated with ground-level habitat features. Overall, the impact of P. cinnamomi on vegetative structure and floristics was not reflected in different abundances of ground-dwelling invertebrates.  相似文献

Pinus nigra (Austrian pine) is a tree widely planted in central and southern Europe in the reforestation of areas with poor soils. One of the major constrains of P. nigra is a fungal disease caused by Sphaeropsis sapinea which causes shoot tip dieback and tree mortality. This paper describes a real‐time quantitative PCR assay based on the partial sequence of the small subunit ribosomal RNA of S. sapinea to detect and quantify DNA of the fungal pathogen in S. sapinea inoculated shoots. The study found that real‐time quantitative PCR was a reliable technique to diagnose this disease and suggests that it can also be used to study fungal behaviour in host tissue and particularly to quantify fungal growth in the latent or endophytic phase.  相似文献

Canopy tree survival and compositional change of the Greefswald forest on the Limpopo River, South Africa, were monitored between 1990 and 2007 in response to a severe drought (cessation of flow in 1991/2), water abstraction commencing in 1991, a mega‐flood in 2000 and increasing impact of elephants since 2000. Aerial photographs confirmed that forest area had not decreased during the 35 years prior to the study. In total, 25% of 428 canopy trees tagged in 1990 had died by 2005. Tree density was reduced from 22.8 to 16.3 trees per hectare. Forest was thus transformed to woodland. Mortality was attributed mainly to drought stress (47%), drought in combination with creeper infestation (30%) and the flood (21%). Of the nine main canopy species, mortality was highest for Acacia xanthophloea (56%) and Faidherbia albida (37%) mainly because of drought‐related stress, and Ficus sycamorus (25%) mainly because of the flood. Water extraction increased drought‐related mortality in the area of extraction by 45%. Creepers rendered microphyllous but not broad‐leafed species more vulnerable to drought‐induced mortality. Elephants were responsible for a further 3% mortality between 2005 and 2007. Composition has shifted towards ‘drought‐tolerant’ species not selected by elephants, namely Philenoptera violacea, Xanthocercis zambesiaca and Schotia brachypetala. Initial concern about water abstraction was eclipsed by a rapid, unpredictable concatenation of a series of rare events that transformed forest to woodland in less than 15 years.  相似文献

Climate change science is increasingly concerned with methods for managing and integrating sources of uncertainty from emission storylines, climate model projections, and ecosystem model parameterizations. In tropical ecosystems, regional climate projections and modeled ecosystem responses vary greatly, leading to a significant source of uncertainty in global biogeochemical accounting and possible future climate feedbacks. Here, we combine an ensemble of IPCC‐AR4 climate change projections for the Amazon Basin (eight general circulation models) with alternative ecosystem parameter sets for the dynamic global vegetation model, LPJmL. We evaluate LPJmL simulations of carbon stocks and fluxes against flux tower and aboveground biomass datasets for individual sites and the entire basin. Variability in LPJmL model sensitivity to future climate change is primarily related to light and water limitations through biochemical and water‐balance‐related parameters. Temperature‐dependent parameters related to plant respiration and photosynthesis appear to be less important than vegetation dynamics (and their parameters) for determining the magnitude of ecosystem response to climate change. Variance partitioning approaches reveal that relationships between uncertainty from ecosystem dynamics and climate projections are dependent on geographic location and the targeted ecosystem process. Parameter uncertainty from the LPJmL model does not affect the trajectory of ecosystem response for a given climate change scenario and the primary source of uncertainty for Amazon ‘dieback’ results from the uncertainty among climate projections. Our approach for describing uncertainty is applicable for informing and prioritizing policy options related to mitigation and adaptation where long‐term investments are required.  相似文献

We studied xylem anatomy and hydraulic architecture in 14 transgenic insertion events and a control line of hybrid poplar (Populus spp.) that varied in lignin content. Transgenic events had different levels of down-regulation of two genes encoding 4-coumarate:coenzyme A ligase (4CL). Two-year-old trees were characterized after growing either as free-standing trees in the field or as supported by stakes in a greenhouse. In free-standing trees, a 20 to 40% reduction in lignin content was associated with increased xylem vulnerability to embolism, shoot dieback and mortality. In staked trees, the decreased biomechanical demands on the xylem was associated with increases in the leaf area to sapwood area ratio and wood specific conductivity (k(s)), and with decreased leaf-specific conductivity (k(l)). These shifts in hydraulic architecture suggest that the bending stresses perceived during growth can affect traits important for xylem water transport. Severe 4CL-downregulation resulted in the patchy formation of discoloured, brown wood with irregular vessels in which water transport was strongly impeded. These severely 4CL-downregulated trees had significantly lower growth efficiency (biomass/leaf area). These results underscore the necessity of adequate lignification for mechanical support of the stem, water transport, tree growth and survival.  相似文献

Traumatic spinal cord injury causes an inflammatory reaction involving blood-derived macrophages and central nervous system (CNS)-resident microglia. Intra-vital two-photon microscopy enables the study of macrophages and microglia in the spinal cord lesion in the living animal. This can be performed in adult animals with a traumatic injury to the dorsal column. Here, we describe methods for distinguishing macrophages from microglia in the CNS using an irradiation bone marrow chimera to obtain animals in which only macrophages or microglia are labeled with a genetically encoded green fluorescent protein. We also describe a injury model that crushes the dorsal column of the spinal cord, thereby producing a simple, easily accessible, rectangular lesion that is easily visualized in an animal through a laminectomy. Furthermore, we will outline procedures to sequentially image the animals at the anatomical site of injury for the study of cellular interactions during the first few days to weeks after injury.  相似文献

Nitrogen and phosphorus are both important life elements. N, P and combined N-P fertilizers were added to the declining population Spartina anglica Hubbard in coastal China. Some growth parameters and eco-physiological responses of S. anglica to different fertilizer treatments (N, P and combined N-P fertilizer addition with high, medium and low levels, respectively) were measured. The fertilizer addition had a highly significant effect on the dynamics of its height-growth, number of leaves, number of roots and total biomass. Only N addition had a significant effect on leaf area and leaf thickness in all fertilizer treatments. On the dynamics of its height-growth, the effect of N addition was the most apparent, and the effect of N-P addition was not greater than those of N and P addition separately. The photosynthesis rate was enhanced and the yield was the highest with the highest N, the highest N-P and the medium P addition. The rates were higher than those of CK by 19.08 μmol·m?2·s?1, 15.47 μmol·m?2·s?1 and 11.23 μmol·m?2·s?1, respectively. The activity of SOD and POD increased with the treatments after freshwater stress for 14 days. Effects of medium N and P addition were significant for SOD activity. However, POD activity was significantly higher with the treatment of higher N and higher N-P addition. In a word, fertilizer addition improved the growth of the declining population S. anglica. The results indicated that the decline of S. anglica was correlated with the nutriment deficiency in soil, especially with the lack of N.  相似文献

Hymenoscyphus fraxineus causes a lethal disease known as “ash dieback” in the common ash, Fraxinus excelsior, in Europe. It is hypothesized that the fungus originated from East Asia. This fungus is found on the leaf litter of the Manchurian ash, Fraxinus mandshurica, in Japan and is reported to produce apothecia on pseudosclerotial plates formed mainly on decomposing rachises. However, dieback disease has not been reported in Japan, and little is known about the life cycle of H. fraxineus. This study was conducted to explore the behavior and life cycle of this fungus. It was revealed that, after infection by ascospores, H. fraxineus endophytically inhabits the living leaves of F. mandshurica. On fallen leaves, the fungus behaves saprophytically, producing apothecia on pseudosclerotial plates formed mainly on the decomposing rachises. Analysis by real-time quantitative polymerase chain reaction (qPCR) revealed that the amount of H. fraxineus DNA sharply increased in rachises, while such sharp increase of DNA was not found in leaflets.  相似文献
