The gene of Blomia tropicalis group 5 allergen (Blo t 5) was cloned and expressed in Pichia pastoris KM71. Selected KM71 clones were cultivated in a fed-batch bioreactor feeding first glycerol then followed by methanol. Recombinant Blo t 5 constituted about 30% of the total broth protein after 60 h cultivation. The harvested broth was purified to >95% purity by a two-step anion exchange chromatography. The overall yield was 37 mg Blo t 5 per litre broth.  相似文献

Spider mites are very small phytophagous organisms living on plant leaves. They possess a diverse array of life types. Such diversity in life type has been supposed to be a result of coevolution between predator and prey, although there is little concrete evidence to support this concept. In Yezonychus sapporensis inhabiting the leaves of a dwarf bamboo, Sasa senanensis, all quiescent stages including eggs appear on the tips of the host leaf hairs during the summer season, and this habit is thought to be a kind of predator avoidance trait. To confirm this, the survival rates of Y. sapporensis eggs deposited on natural sites and experimentally manipulated sites were observed for eight co-occurring predator species. The results clearly showed that the oviposition behavior of Y. sapporensis has a function in avoiding predator attack, at least under experimental conditions. The effects of prey mite suitability on several demographic characteristics of phytoseiid predators and the relationship of these effects to the potential of phytoseiid predators to control herbivorous mite populations are well documented. Evidence has also accumulated in the last 20 years demonstrating that phytoseiid predators utilize herbivorous prey mite-induced plant volatiles as olfactory cues in locating their herbivorous mite prey, but less well established is the predictability of reproductive success from the ability of the predators to utilize olfactory cues to locate their prey, and how these processes are related to the success of the predators as biological control agents of the herbivorous mite. In this study, we determined in laboratory no choice experiments, the development, survivorship and fecundity of the two neotropical phytoseiid predators Typhlodromalus manihoti Moraes and T. aripo DeLeon when feeding on three herbivorous mites, including the key prey species Mononychellus tanajoa (Bondar), and the two alternative prey species Oligonychus gossypii (Zacher) and Tetranychus urticae (Koch). Intrinsic rate of increase (rm) of T. aripo was 2.1 fold higher on M. tanajoa as prey compared with T. urticae as prey, while it was almost nil on O. gossypii. For T. manihoti, rm was 2.3 fold higher on M. tanajoa as prey compared with O. gossypii as prey, while reproduction was nil on T. urticae. An independent experiment on odor-related prey preference of the two predator species showed that T. manihoti and T. aripo preferred odors from M. tanajoa-infested leaves to odors from O. gossypii-infested leaves. Moreover, both predator species preferred odors from M. tanajoa-infested leaves over those from T. urticae-infested leaves. As reported here, life history of the two predatory mites matches odor-related prey preference if the key prey species is compared to the two inferior prey species. The implications of our findings for the persistence of T. manihoti and T. aripo and biological control of M. tanajoa in the cassava agroecosystem in Africa are discussed. Inositol deficiency of Schizosaccharomyces pombe did not induce significant change of contents of various cellular components except for phospholipids and inositol. The most remarkable decrease in inositol content by the deficiency occurred in the mitochondrial fraction. Electronmicroscopic observation of the inositol-deficient cells of Sch. pombe showed no remarkable thickening of cell wall as occurred in the inositol-exacting mutant of Saccharomyces cerevisiae Strain A–21–20.

Marked loss of fermentative activity under the aerobic condition was caused from inositol deficiency without significant change of activities of respiration and anaerobic fermentation. This seemed to indicate elevated regulatory control of the fermentative activity by oxygen in the inositol-deficient Sch. pombe.

Phosphorylative activities of intact cells and the isolated mitochondria coupled with oxidation was also remarkably suppressed by inositol deficiency.  相似文献

This paper reports on ectoparasitic chigger mites found on small mammals in Yunnan Province, southwest China. Data were accumulated from 19 investigation sites (counties) between 2001 and 2009. A total of 10 222 small mammal hosts were captured and identified; these represented 62 species, 34 genera and 11 families in five orders. From the body surfaces of these 10 222 hosts, a total of 92 990 chigger mites were collected and identified microscopically. These represented 224 species, 22 genera and three subfamilies in the family Trombiculidae (Trombidiformes). Small mammals were commonly found to be infested by chigger mites and most host species harboured several species of mite. The species diversity of chigger mites in Yunnan was much higher than diversities reported previously in other provinces of China and in other countries. A single species of rodent, Eothenomys miletus (Rodentia: Cricetidae), carried 111 species of chigger mite, thus demonstrating the highest species diversity and heaviest mite infestation of all recorded hosts. This diversity is exceptional compared with that of other ectoparasites. Of the total 224 mite species, 21 species accounted for 82.2% of all mites counted. Two species acting as major vectors for scrub typhus (tsutsugamushi disease), Leptotrombidium scutellare and Leptotrombidium deliense, were identified as the dominant mite species in this sample. In addition to these two major vectors, 12 potential or suspected vector species were found. Most species of chigger mite had a wide range of hosts and low host specificity. For example, L. scutellare parasitized 30 species of host. The low host specificity of chigger mites may increase their probability of encountering humans, as well as their transmission of scrub typhus among different hosts. Hierarchical clustering analysis showed that similarities between different chigger mite communities on the 18 main species of small mammal host did not accord with the taxonomic affinity of the hosts. This suggests that the distribution of chigger mites may be strongly influenced by the environment in which hosts live.  相似文献

To determine selective effectiveness for specific pesticides on biological control species we evaluated the contact toxicity of 16 insecticides, 2 acaricides, 3 fungicides, and 5 biopesticides. Targeted species included 3 predatory mites (Phytoseiulus persimilis Athias-Henriot, Amblyseius swirskii Athias-Henriot, and Neoseiulus californicus McGregor), 5 hymenopteran parasitoids (Diglyphus isaea Walker, Aphidius colemani Viereck, Encarsia formosa Gahan, Eretmocerus eremicus Rose and Zolnerowich and E. mundus Mercet), and 2 hemipteran predators (Orius laevigatus Fieber and Nesidiocoris tenuis Reuter) in laboratory condition. In addition, residual toxicity was evaluated on P. persimilis, E. formosa, and O. laevigatus. For contact toxicity, five insecticides (spinetoram, spinosad, lepimectin, chlorfenapyr, and dinotefuran + spinetoram) showed high toxicity to predatory mites. Most pesticides tested were highly toxic to all hymenopteran species except for D. isaea which showed low susceptibility to 11 pesticides. Bistrifluron + flubendiamide and B. valismortis were less toxic to A. colemani, and only B. valismortis was safe to both E. mundus and E. eremicus. Insect growth regulators (methoxyfenozide and bistrifluron), chlorantraniliprole, and bistrifuron + flubendiamide were not toxic to hemipteran predators. Fungicides and biopesticides were safe to hemipteran predators except for two biopesticides (B. subtilis and P. fluorescens). Most pesticides had low residual toxicity to P. persimilis, with the exception of chlorfenapyr whose toxicity persisted over 7 days. One insecticide (cyantraniliprole), 2 acaricides (spiromesifen and fenpyroximate), 1 fungicide (metrafenone), and 4 biopesticides (B. subtilis, P. polymyxa, P. fluorescens, and B. valismortis) showed a much lower residual toxicity to E. formosa. Eight insecticides, 2 acaricides, 3 fungicides, and 5 biopesticides showed low residual toxicity to O. laevigatus.  相似文献

Varroa jacobsoni reproduces in honey bee brood cells. Here the behavioral activity and use of space by infestingVarroa females and progeny were quantified in transparent artificial brood cells. The time-activity budget of both infesting and developing mites converged toward a stable pattern which was established during the bee prepupal stage of the infesting mites and the protonymphal stage of mite progeny. The pattern was such that infesting females and offspring eventually divided their activity between the fecal accumulation on the cell wall, which served as the rendezvous site for newly molted individuals, and the feeding site prepared on the pupa by the foundress. Other parts of the cell wall were used for oviposition and molting, away from the fecal accumulation on which activity of mobile stages was concentrated. Space structuring and the time-activity budget inVarroa probably evolved to enhance the number of fertilized females produced within the capped brood, where space and time are limiting factors. These behavioral adaptations parallel those of other mite species which show group behavior within cavities.  相似文献

本文记述我国腺水螨科Lebertiidae、腺水螨属Lebertia二新种。该属腺毛分布以E1位移于1基节板前端背方为显著特点,基于对两新种的详细观察,描述了腺毛分布体位。三叉腺水螨Lebertia(Piloleberia)trifurcillaJin,sp.nov.本新种近似于印度种Lebertia(P.)carmamayaCook1967,但其E1具发达的毛状表皮突、殖吸盘板后缘刚毛少而短等特征明显别于后者。刷电腺水螨Lebertia(Pilolebertia)ciliaaJin,sp.nov.本新种近似于非洲种L.algeriensisLundblad1942,其以颚底背突长而尖、腹突尖锐等特征显著别于后者。  相似文献
