The phylogeny of Ptychostomum was first spacer (ITS) region of the nuclear ribosomal (nr) DNA DNA rps4 sequences. Maximum parsimony, maximum undertaken based on analysis of the internal transcribed and by combining data from nrDNA ITS and chloroplast likelihood, and Bayesian analyses all support the conclusion that the reinstated genus Ptychostomum is not monophyletic. Ptychostomum funkii (Schwagr.) J. R. Spence (≡ Bryum funkii Schwaigr.) is placed within a clade containing the type species of Bryum, B. argenteum Hedw. The remaining members of Ptychostomum investigated in the present study constitute another well-supported clade. The results are congruent with previous molecular analyses. On the basis of phylogenetic evidence, we agree with transferring B. amblyodon Mull. Hal. (≡ B. inclinatum (Brid.) Turton≡ Bryum archangelicum Bruch & Schimp.), Bryum lonchocaulon Mull. Hal., Bryum pallescens Schleich. ex Schwaigr., and Bryum pallens Sw. to Ptychostomum. 相似文献

Coelomycetous fungi classified in Ascochyta, Phoma, and Phyllosticta have been recorded from spots on leaves and pods of soybeans. Based on the Genealogical Concordance Phylogenetic Species Concept, the authors suggest the re-evaluation of the taxonomic status of Phoma sojicola (syn. = Ascochyta sojicola) and Phyllosticta sojicola. In spite of the former delimitation of Phoma sojicola based on small differences in morphological features, it has proved to be identical to Phoma pinodella. Similarly, it was also confirmed that Phyllosticta sojicola was identical to Phoma exigua var. exigua. The authors supply tools for identification of Phoma-like fungi by combined conventional and molecular methods. Protein-encoding genes (tef1 and β-tubulin) were successfully applied within the Phoma genus to infer phylogenetic relationships. 相似文献

The relationships between dispersal and local transmission rate of parasites are essential to understanding host–parasite coevolution and the emergence and spread of novel disease threats. Here we show that year‐round transmission, as opposed to summer transmission, has repeatedly evolved in malarial parasites (genera Plasmodium and Haemoproteus) of a migratory bird. Year‐round transmission allows parasites to spread in sympatric host's wintering areas, and hence to colonize distantly located host's breeding areas connected by host‐migration movements. Widespread parasites had higher local prevalence, revealing increased transmission, than geographically restricted parasites. Our results show a positive relationship between dispersal and local transmission of malarial parasites that is apparently mediated by frequent evolutionary changes in parasite transmission dynamics, which has important implications for the ecology and evolution of infectious diseases. 相似文献

This paper is a speleomycological report from a former aluminous shale mine in Janowiec, Poland. Fungi were identified morphologically and molecularly. Microclimatic conditions differed significantly between locations of the study. However, the external environment around the mine did not directly increase the community composition and concentration of fungi in the mine. The density of fungi isolated from the air outside the mine was 63.1 colony forming units (CFU) per 1 cm3 of air. Inside the mine, fungal density ranged from 287.5 to 655 CFU per 1 m3 from the air, 28.4 to 131.1 CFU per 1 cm2 from the rock surfaces and 288.1 to 335.1 CFU per 1 cm3 from the water. Pearson correlation analysis showed that the levels of fungi isolated from the air were correlated positively with temperature, relative humidity and CO2 concentration. The concentration of fungi isolated from the rock surfaces showed a positive correlation with air flow. Five species of filamentous fungi were isolated from the sampled external air, 10 species from the internal air, six species from the rock surface and 11 species from the water. The fungi most frequently isolated from the air and water of the mine belonged to Penicillium spp., whereas from the rock surface, Geomyces pannorum was most frequently isolated. Some of the fungi present in the mine can be psychrotolerant and pathogenic for humans and animals, and they can also cause degradation of rocks. 相似文献

To increase the productivity in biopharmaceutical production, a natural step is to introduce integrated continuous biomanufacturing which leads to fewer buffer and storage tanks, smaller sizes of integrated unit operations, and full automation of the operation. The main contribution of this work is to illustrate a methodology for design and control of a downstream process based on integrated column sequences. For small scale production, for example, pre‐clinical studies, integrated column sequences can be implemented on a single chromatography system. This makes for a very efficient drug development platform. The proposed methodology is composed of four steps and is governed by a set of tools, that is presented, that makes the transition from batch separations to a complete integrated separation sequence as easy as possible. This methodology, its associated tools and the physical implementation is presented and illustrated on a case study where the target protein is separated from impurities through an integrated four column sequence. 在基因组中,编码区存在许多高度相似的基因簇或基因群(多拷贝基因),非编码区也存在大量的重复序列。这些重复序列能通过改变染色体的三维结构调控基因的转录,对于生物体的遗传与进化起到了重要的作用。其高度同源的特征使得利用CRISPR/Cas9技术进行基因组编辑时面临更加复杂的状况。如果编辑的片段是二倍体或多倍体,还会产生各条染色单体上的编辑情况不相同的现象。为此本文选择了2个位于同一染色体相距11 kb的高度同源300 bp片段(L1和L2)进行CRISPR介导的DNA片段编辑。采用一对sgRNA(分别共同靶向两片段的上、下游位点)引导Cas9对HepG2细胞两个高度相似的DNA片段进行切割。片段编辑的细胞进一步单克隆化后,对获得的22个L1/L2编辑的CRISPR单克隆细胞株进行详细的基因型鉴定。结果发现除了这两个DNA片段本身被删除外,它们之间的大片段也存在被删除的现象,三个片段的各种反转组合也很频繁。该研究结果对于采用CRISPR/Cas9系统编辑多拷贝基因或重复序列,尤其是对二倍体或多倍体生物进行基因组编辑时具有重要的借鉴和参考价值。 相似文献

We investigated areas at the eastern fringe of Kolkata, Dhapa–a land filled with Municipal Solid Waste (MSW) land of West Bengal (India) for exploring nematode species diversity, abundance and dynamic at three sites practising different vegetable-based cropping sequences. The area sampled at monthly interval and the soil and root populations estimated at laboratory. The plant-parasitic nematode (PPN) species and free-living group (saprozoics) were estimated from soil. No differences in the diversity of nematode species and feeding groups among the sites in the MSW land were found but considerable variations in abundance and dynamics of PPNs were noted. All three sites at MSW, total PPNs outnumbered free-living nematodes in most of the samples but mononchids were less abundant. Saprozoic nematode index (SNI) was determined; SNI value was low (0.33–0.40) in this organic rich production system. 相似文献
