The role of reversing factor (RF) in the regulation of protein synthesis by inhibitory protein kinases that phosphorylate the 38,000-dalton subunit of initiation factor eIF-2 has been examined. Results show that as with the heme-regulated protein kinase (HRI), RF restores protein synthesis in reticulocyte lysates inhibited by translational inhibitors from rat liver, wheat germ, Krebs ascites cell, by oxidized glutathione, the protein kinase activated by double stranded RNA (dRI), and the interferon-induced double stranded RNA activated protein kinase from Ehrlich ascites and Hela cells. These findings suggest that RF plays an important role in eukaryotic protein chain initiation cycle. 相似文献

Total polyadenylated enriched mRNA was prepared from rat liver by guanidine-HCl extraction and oligo(dT)-cellulose chromatography. It was translated in an mRNA-dependent wheat germ system and rabbit reticulocyte lysate system, using radiolabeled leucine or methionine as amino acid precursor. A product, designated preapoE, was specifically precipitated by a rabbit anti-rat apoE serum and accounted for 1.5% of the total radioactive peptides. It migrated as a single band of radioactivity on SDS gels with an apparent molecular weight similar to that of mature plasma apoE. Inclusion of dog pancreatic microsomal membranes in the translation reaction resulted in a slightly smaller product (by 500 daltons). It also converted the preapoE from an endoglycosidase H-resistant to an enzyme-sensitive species. This suggests that processing of preapoE takes place by the cotranslational removal of a signal peptide and core glycosylation of the mature protein. 相似文献

In non fatal and synchronous P. chabaudi rodent malaria, we observed at the stage of parasitaemia peak, an alteration (50 % decrease) in LCAT activity. This decrease could be related partly to hepatic dysfunction, and mainly to circulating inhibitors released into blood from parasitized red blood cells at each end of a schizogonic cycle. This decrease in LCAT activity, at this step of the infection, accounts for part of the dyslipoproteinemia previously observed (i.e., increase in cholesterol and phospholipids into VLDL-LDL and decrease in the EC series and delayed conversion of Tg-rich lipoproteins into LDL-HDL. At a prepatent step of infection and after the parasitaemia peak, the alterations observed in LCAT activity, (respectively, increase and then decrease), would be related to similar changes in levels of cholesterol of HDL associated to complex changes in triacylglyceride transport and metabolism. 相似文献
