Prostacyclin (PGI2) induced a dose-dependent decrease in blood pressure with slight increases in heart rate and body temperature, when administered at the doses of 0.1–100 μg into the lateral cerebral ventricle (i.c.v.) of the urethane-anaesthetised rat. When the same doses were administered intravenously, both the blood pressure and heart rate decreased. Central pretreatment with sodium meclofenamate (1 mg/rat i.c.v.) antagonised the central hypotensive effect of PGI2 but i.c.v. pretreatment of the rats with indomethacin (1 mg/rat) failed to affect the PGO2-induced hypotension. Central pretreatment with two histamine H2-receptor antagonists, cimetidine (500 μg/rat i.c.v.) or metiamide (488 μg/rat i.c.v.), antagonised the blood pressure lowering effect of 0.1 μg dose of PGI2 but failed to affect the hypotension induced by higher PGI2 doses. Therefore the main central hypotensive effect of PGI2 seems not to be associated with the stimulation of histamine H2 -receptors in the brain.The hypotensive effect of i.c.v. administered PGI2 appears to be due to an action upon the central nervous system rather than to a leakage into the peripheral circulation. This assumption is supported by the fact that sodium meclofenamate i.c.v. antagonished the effect of PGI2. In addition, the chronotropic response to i.c.v. PGI2 was opposite to that induced by intravenous administration. The results also suggest that there may be differences in the mode of action between sodium meclofenamate and indomethacin. 相似文献

A highly active inhibitor of the binding of estrone and estradiol-17β to rat alpha-fetoprotein is demonstrated for the first time in embryo, immature and adult rat sera as well as in fetal and adult human sera. The competitive character and the narrow specificity of this inhibition effect is shown. The major compound responsible for this activity is isolated by successive column Sephadex LH20 and thin layer chromatography : it is characterized as a nonpolar, nonphenolic, dialysable and thermostable substance, unreactive towards anti-estrone and anti-estradiol-17β anti-bodies. A major handicap in the development of simple and accurate radioimmunoassay procedures for bile acids has been the lack of a radioactive standard of high specific activity. To provide such a compound, we first synthesized cholylhistamine using the carbodiimide reaction. The hypothesized structure was confirmed by elemental analysis, thin-layer chromatography, infrared and mass spectral analysis. The cholylhistamine was then iodinated with 125I, using the choloramine-T method. The 125I-cholylhistamine was bound by antisera raised against a cholic acid-bovine serum albumin conjugate. The objective of these studies was to determine if prolactin, known to induce its own receptors, alters the prostaglandin (PG) synthesis which could, in turn, modify the fluidity of the membrane and thus alter the functionality of prolactin receptors. Adult male C3H mice were injected subcutaneously with 100 μg of oPRL every 4 h for 0, 24 or 48 h and sacrificed 8 h after receiving the last injection. Liver 100,000 × membrane pellets were used in the measurement of these parameters. The amount of binding of prolactin to these membranes increased with the duration of injections, the values being 179 and 244% of control values after 24 and 48 h of injections, respectively. The amounts of PGF2α and PGE synthesized also increased after these injections, the values being 127 and 270% of control for PGF2α and 634 and 695% of control values for PGE after 24 and 48 h of injections, respectively. Fluorescence polarization, an index of microviscosity, was decreased by 14 and 20% after 24 and 48 h of PRL administration, respectively. Previous studies have demonstrated simultaneous in vitro effects of prostaglandin on both prolactin receptors and membrane fluidity. The current data are in agreement with those observations and suggest that prolactin may modulate its own receptor by increasing the fluidity of the membrane in which it exists by alterations within the PG cascade. Such biochemical changes may then modify existing restraints and allow the hormone receptor to assume a more functional configuration. 相似文献

Various human Burkitt lymphoma and LCL lines established in vitro and their derived somatic cell hybrids were tested for their comparative EBV receptor levels in a virus binding assay. Their graded C3b and C3d complement receptor expression was estimated simultaneously by means of isotope labeled rosette marker cells. The receptor concentration of each cell line was related to Raji as the standard of comparison, K 562, P3HR-1, and YACUT were used as negative controls. In general, the charging curves for EBV and C3d receptors parallelled each other (r = 0.97) while C3b receptor charging showed no correlation (r < 0.60). In the Raji hybrids between the C3b receptor positive Raji parent and various patents that were negative for this receptor, C3b receptor expression was low or negative. In contrast, the C3d negative P3HR-1 line gave rise to hybrids, after fusion with receptor-positive cells, that were intermediate with regard to their C3d receptor expression. The host range restriction of the Epstein-Barr virus is determined at the receptor level. The close relationship between the EBV receptor and the C3d receptor, a B-lymphocyte-specific moiety, suggests that the moderate interaction with EBV with the B lymphocytes may have had a selective advantage, favoring the presence of EBV. Since EBV causes lytic infections after artificial introduction into nonnatural host cells, it may represent a B-lymphocyte-specific host range mutant, derived from an originally lytic herpesvirus with a much broader target cell range. 相似文献

Rat heart ventricular cells, purified by Percoll density gradient centrifugation, were incubated in the presence of 1.3 mM CaCl2. After 20 min incubation, samples of the cells were lysed in medium containing 0.3 mM digitonin, ruthenium red and EGTA, and a mitochondrial fraction was isolated at intervals thereafter. Extrapolation of the mitochondrial 45Ca2+ contents to zero time enabled the endogenous 45Ca2+ to be estimated at the time of cell lysis. The lysis conditions yielded essentially complete release of lactate dehydrogenase from the cells, but caused negligible damage to the mitochondria as judged by their retention of glutamate dehydrogenase, and their ability to accumulate and retain Ca2+ in the absence of ruthenium red and EGTA. The data indicate that about 13% of total cell Ca2+ only may be mitochondrial in vivo. 相似文献

A field experiment was conducted to investigate if carbon isotope (13C) discrimination () measured at the vegetative stage of spring wheat (Triticum turgidum L. var. durum) is related with the yield and water use efficiency (WUE) at ripening. A line source sprinkler irrigation system exposed the wheat genotypes to different watering regimes, from rainfed to full irrigation and thereby increased the range in yield and WUE attainable in the four genotypes studied. The results indicated that values measured at the late stem elongation stage 60 days after planting (DAP), showed strong positive correlation with total dry matter yield (r=0.732***), and a highly significant negative correlation with WUE (r=–0.755***) measured at ripening 105 DAP. The data suggest that the imprints of measured at vegetative growth stage persists throughout the entire growth period, until maturity. Subject to confirmation from additional studies in other crops and locations, early measurements of may prove a useful tool for rapid and early screening of cultivars, for high yield and high WUE. 相似文献

We previously demonstrated that the oxysterol potentiation of arachidonic acid release and prostaglandin biosynthesis induced by foetal calf serum activation of normal rat kidney (NRK) cells (fibroblastic clone 49F) was not related to a direct effect of oxysterols on cell free Ca2+ level. Since both Ca2+ variations and protein C are involved in arachidonic acid release in some models, we looked for a possible modulation by protein C in the oxysterol effect on arachidonic acid release. We show that when the phorbol ester 12-O-tetradecanoyl-phorbol-13acetate (TPA), a protein kinase C activator, was added to the culture medium, the oxyterol effect on arachidonic acid release and prostaglandin synthesis clearly increased. Moreover, the effect of TPA was dose-dependent and TPA EC50 (4 × 10−9 M) was unchanged in the presence of the oxysterol. Preincubation of cells with TPA for 24 h prevented the arachidonic acid release induced by TPA alone, whereas the oxysterol effect was decreased but not abolished. In the absence of serum, TPA and ionomycin added together induced the same noticeable (arachidonic acid) release and PGE2 synthesis as serum alone. Nevertheless, the potentiating effect of cholest-5-ene-3β,25-diol was much higher when serum itself was used to activate NRK cells than it was in the present serum-mimicking experimental conditions. Thus, the presence of growth factors is probably required to obtain a full oxysterol effect. We conclude that the oxysterol effect was synergistic with, but not fully dependent on, protein kinase C and Ca2+ ion fluxes, therefore oxysterols could affed earlier events triggered by serum growth factor binding to their cell membrane receptors. 相似文献

α-Tropomyosin from rat cardiac muscle was shown by two-dimensional gel electrophoresis to become phosphorylated when tissue slices were incubated in Eagle's medium supplemented with 32Pi. In the adult rat and mouse heart the level of phosphorylation was ~30%, but the level was much higher in the foetal heart (60–70%). A similar developmental trend was observed in skeletal muscle from the rat and mouse, where phosphorylated forms of both α- and β-tropomyosins were observed. When rat cardiac cells were grown in tissue culture in the presence of 32Pi, radioactivity was incorporated into the region of the gel containing tropomyosin. 相似文献
