基因枪转化小麦幼胚的再生培养与转基因植株的获得   总被引:20,自引:3,他引:17  
以小麦幼胚为受体,用基因枪法对Trx-S反义基因 目的基因 和Bar基因 标记基因 进行了共转化,以轰击后的小麦幼胚为实验材料,对幼胚培养的基本培养基、分化和生根培养基进行了筛选优化.结果表明:4种基本培养基中,L3培养基的成愈率最高,且增殖速度快;MS培养基次之.以L3为基本培养基,分化培养基中添加NAA1mg·L-1和ZT2mg·L-1配比对愈伤组织诱导分化的效果最好,分化率达到50%以上.1/2MS培养基中添加IAA0.8mg·L-1的生根效果好,且移栽成活率高.以优化的培养方案对来自7个小麦品种的幼胚进行转化与再生培养,多数品种的出愈率都达到90%以上,分化率在40%以上,并在5个品种上获得再生植株,经检测证实在4个品种上获得转基因再生植株.  相似文献

以江西省50种珍稀濒危植物为定量研究对象,采用以濒危系数、遗传价值系数、物种价值系数和生物学特征系数4个指标构成的珍稀濒危植物等级综合评价值的方法对上述植物优先保护顺序进行了定量研究。对50种珍稀植物进行排序的结果基本符合《中国珍稀濒危植物》和《中国植物红皮书》第1册的分析,但也有不同:属一级保护的有长柄双花木、天目木姜子和长序榆等共10种,占研究物种总数的20.00%,其中有浙江楠和华东黄杉等2渐危种;二级保护的有胡豆莲、连香树、伯乐树、银钟花等共26种,占52.00%,其中胡豆莲属于濒危种;三级的有小花木兰、观光木等12种,占24.00%,其中有4种属稀有种,8种属渐危种;此外,八角莲、野大豆等在江西可以暂不列入保护,这两者均属渐危种,占4.00%。  相似文献

Abstract. This study investigates the effects of human disturbance and environmental factors on the distribution of alien plant species on the Georgia Sea Islands (GSI), USA. We sampled the absolute cover of native and alien plant species on two tourist islands (St. Simons Island and Jekyll Island) and on two protected National Wildlife Refuge Islands (Blackbeard Island and Wassaw Island). On each island, vegetation composition and environmental variables (soil properties and salt spray) were measured in two habitats that differed substantially in their degree of environmental stress, the more exposed primary dune and the more sheltered and inland maritime forest. Sites were further stratified within each habitat into areas that had different levels of human disturbance. Many alien species were present on all islands and the absolute cover of alien species was not significantly different among islands even though they varied substantially in their degree of accessibility and overall land use. Alien plant cover was appreciably greater in severely disturbed sites than in less disturbed sites on all islands and within both habitats. However, the difference between disturbance categories was much less pronounced in the primary dunes where human disturbance agents do not mitigate the harsh environmental conditions of this habitat (salt spray and saline soils). Alien plant abundance on the GSI is evidently more dependent upon the availability of disturbed ground than the degree of accessibility or overall island development. It appears that human disturbance increases alien cover in general, but in environments where the stress levels are not mitigated, human disturbance does little to foster alien invasions.  相似文献

Water hyacinth (Eichhornia crassipes (Mart.) Solms.)creates severe problems in the irrigationdistricts of Mexico, particularly in westernSinaloa. Therefore water hyacinth weevils(Neochetina eichhorniae Warner and N. bruchi Hustache), imported from the USA in1993, were used to initiate a biologicalcontrol program. Precautionary screeningrevealed that some were infected with amicrosporidian so disease-free colonies wereproduced by eliminating infected breedinglines. To demonstrate effectiveness prior toopen field releases, weevils were firstreleased in cages at field sites. Weevilintensity increased to 6.3 weevils/plant after320 days when the plants were all dead ordying. More than 8,600 N. bruchi and14,500 N. eichhorniae were then releasedat various sites during January 1995 to August1996. Waterhyacinth coverage declined atBatamote reservoir (134 ha) from 95% to <3%by 1997; at the 12-ha Hilda reservoir from100% in May 1995 to 1% by March 1998; at the42.3-ha Arroyo Prieto reservoir from 100% to1% during the same interval; and at theMariquita reservoir (492 ha), the largestreservoir in the Humaya system, from 394 ha(80%)to 98.4 ha (20%).  相似文献

There are various approaches to explain the mechanisms of biological invasions. It is possible (1) to focus on the characteristics of invading species and (2) on those of the ecosystems invaded, (3) to investigate the relationship between these two factors (key–lock approach), or (4) to differentiate the invasion process in time. Each of these approaches may serve to improve the understanding of some aspects of biological invasions, and each of them is in some way suitable for the purpose of predicting invasions. We discuss the usefulness and the limitations of these approaches, focusing on case studies from central Europe. An example of the fourth approach, a model of steps and stages of plant invasions that describes the invasion process in greater detail, illustrates some general limitations in predicting biological invasions. Reverse latitudinal trends in species richness of pitcher-plant food webs   总被引:3,自引:0,他引:3  
Latitudinal patterns in species richness have been well documented for guilds and individual trophic groups, but comparable patterns for entire, multitrophic communities have not been described. We studied the entire food web that inhabits the water‐filled leaves of the pitcher plant Sarracenia purpurea across North America at two spatial scales: among sites and among leaves within sites. Contrary to the expectation, total species richness at both scales increased with latitude, because of increasing species richness at the lower trophic levels. This latitudinal pattern may be driven by a top‐down effect. The abundance of the mosquito Wyeomyia smithii, a ubiquitous top predator in this system, decreases from south to north and may permit greater species richness of prey trophic levels at higher latitudes.  相似文献

Leaf shapes reflect complex assemblages of shape-determining elements, yet evolutionary studies tend to treat leaf shape as a single attribute, for example cordate or linear. As with all complex structures, individual elements of a leaf could theoretically evolve independently and at different rates to the extent permitted by genetic and functional limitations. We examined relative evolutionary lability of shape-determining elements in the highly diverse South African plant genus Pelargonium (Geraniaceae). We used SIMMAP to calculate Bayesian posterior probabilities for ancestral states of leaf-shape characters for major nodes across multiple phylogenetic trees. Trees were derived from a Bayesian analysis of DNA sequence data from four partitions. We found that shape elements differed in rates of character-state transformations across the tree. Leaf base, apex, and overall outline had low rates. Transformations in venation occurred at slightly higher rates and were associated with shifts in venation among major clades. Leaf margin type and overall leaf size showed intermediate rates, whereas high rates were observed in the extent of lamina lobing and functional leaf size. The results indicate that suites of elements characteristic of the recently evolved xerophytic lineage, for example pinnate venation, dissected lamina, and entire margins, were acquired piecemeal over nested levels of the phylogeny.  相似文献

Indigenous medicine is important to rural livelihoods, but lay knowledge and use of medicinal plants has not been extensively studied. Research in KwaZulu-Natal, South Africa, showed that medicinal plants were frequently used by villagers and contributed to their ability to cope with health problems. Knowledge of plants and household remedies was extensive and varied in that households often held different knowledge. Villagers mainly relied on common species, and were generally aware of alternative species for a certain ailment. People were flexible in their use of indigenous and western health care, which were both perceived as beneficial. Improved cooperation between health care systems could improve health standards. Extraction of medicinal plants has been described as unsustainable in the region—a situation not found in the study area. It is argued that conservation policies aimed to restrict access should be differential and potentially not include local consumption, since this may be ecologically unnecessary and entail local hardships.  相似文献

RAPDs and ISSRs were used as molecular markers and combined with the chemical analysis of the volatiles, aiming at characterizing Thymus caespititius from the islands of Corvo, Flores and Graciosa (Azores). Thirty-one individuals were analyzed using 17 RAPD and 11 ISSRs primers, which generated respectively 199 and 127 polymorphic scorable bands. Volatiles were isolated by distillation–extraction and analyzed by GC and GC–MS. Molecular data obtained from both markers discriminated assessments from Corvo and Flores which clustered separately from plants collected in Graciosa. Volatile data grouped plants in a similar way, giving rise to two groups, depending on their oil type; plants collected on Corvo and Flores yielded carvacrol-rich volatiles while those from Graciosa yielded both carvacrol and α-terpineol-rich oils.  相似文献
