Field observations in Hong Kong waters have shown the marine macroalga Ulva reticulata Forsskal (Chlorophyta) to be free of fouling. In this study, the presumptive antifouling mechanism in this alga was investigated and attempts were made to distinguish between possible mechanisms. Waterborne algal compounds were analyzed in larval behavioral bioassays with the marine polychaete Hydroides elegans, a major fouling organism in tropical waters around the world. Larval attachment and metamorphosis in this sessile species is induced by specific bacterial strains in natural bio‐organic films. In bioassays with alga‐conditioned seawater, levels of larval metamorphosis were significantly reduced in comparison to controls. The results demonstrate that the inhibitive effect was caused by direct larval deterrence. Following a bio‐assay‐guided isolation procedure, an inhibitive fraction for larval metamorphosis was purified from U. reticulata‐conditioned seawater. Preliminary chemical analysis of the biologically active fraction pointed to polysaccharides, proteins or glycoconjugates with a molecular weight > 100 kD.  相似文献

Abstract

Bioadhesion and surface wettability are influenced by microscale topography. In the present study, engineered pillars, ridges and biomimetic topography inspired by the skin of fast moving sharks (Sharklet AF?) were replicated in polydimethylsiloxane elastomer. Sessile drop contact angle changes on the surfaces correlated well (R2 = 0.89) with Wenzel and Cassie and Baxter's relationships for wettability. Two separate biological responses, i.e. settlement of Ulva linza zoospores and alignment of porcine cardiovascular endothelial cells, were inversely proportional to the width (between 5 and 20 μm) of the engineered channels. Zoospore settlement was reduced by ~85% on the finer (ca 2 μm) and more complex Sharklet AF? topographies. The response of both cell types suggests their responses are governed by the same underlying thermodynamic principles as wettability.  相似文献

Interaction of zoospores of Ulva linza with cationic, arginine-rich oligopeptide self-assembled monolayers (SAMs) is characterized by rapid settlement. Some spores settle (ie permanently attach) in a ‘normal’ manner involving the secretion of a permanent adhesive, retraction of the flagella and cell wall formation, whilst others undergo ‘pseudosettlement’ whereby motile spores are trapped (attached) on the SAM surface without undergoing the normal metamorphosis into a settled spore. Holographic microscopy was used to record videos of swimming zoospores in the vicinity of surfaces with different cationic oligopeptide concentrations to provide time-resolved insights into processes associated with attachment of spores. The data reveal that spore attachment rate increases with increasing cationic peptide content. Accordingly, the decrease in swimming activity in the volume of seawater above the surface accelerated with increasing surface charge. Three-dimensional trajectories of individual swimming spores showed a ‘hit and stick’ motion pattern, exclusively observed for the arginine-rich peptide SAMs, whereby spores were immediately trapped upon contact with the surface.  相似文献

The excessive growth of Ulva rigida C. Agardh, a green seaweed present in the Northern Adriatic Sea, is a problem for the inhabitants and the economy of the region. As information about hormonal control of growth in seaweeds is scarce, our aim was to investigate the presence of endogenous polyamines and their absorption by algal cells and to correlate the findings with terrestrial plants. Free polyamines (putrescine, spermidine, and spermine) were present endogenously in the algal thallus at concentrations ranging from 4 to 134 μM. Putrescine and spermidine were also present in the seawater in which the alga usually grows at concentrations between 0 and 0.9 μM. Uptake of labeled polyamines occurred, but it was inhibited by cations present in the seawater. Uptake was investigated also by incubation in distilled water. In this case, uptake displayed characteristics similar to those observed in higher plant systems. Uptake studies in seawater showed that polyamine accumulation in algal cells occurred and that it followed a concentration gradient and displayed linear kinetics. The mechanism proposed that of a passive uptake, as indicated also by the inability of metabolic inhibitors to block transport. There was evidence for polyamine binding to external cell sites, but polyamine uptake by protoplasts as well as polyamine translocation and secretion by the whole thallus was also demonstrated. Since cultured and actively growing thallus discs displayed a higher uptake ability than freshly collected ones, a role for polyamines in sustaining growth is discussed.  相似文献

Axenic Ulva mutabilis gametes develop parthenogenetically into callus‐like colonies consisting of undifferentiated cells without normal cell walls. From the accompanying microbial flora of established laboratory strains of U. mutabilis with normal morphology, a Roseobacter, a Sulfitobacter, and a Halomonas species were isolated. Each of these microbe species alone induced the development of the Ulva gametes into thalli composed of differentiated cells with characteristic deficiencies. Typical traits of these thalli were: an enhanced rate of cell division not followed by cell expansion, the presence of unusual cell wall protrusions, and the absence of differentiated rhizoid cells. The addition of a Cytophaga species, also derived from the same microbial flora, to either one of the three other strains resulted in the development of normal fast growing thalli with the typical morphology of the algal strain used. These effects are mediated by specific regulatory factors that are excreted into the environment by the bacteria and could be also isolated from the bacterial cell extracts. In contrast with the Cytophaga‐factor, the regulatory factor of the three other bacterial species was also found intracellularly in other bacterial strains not associated with Ulva, but in this case it was not excreted. Functionally, the Roseobacter‐, Sulfitobacter‐, and Halomonas‐factors resemble a cytokinin, while the Cytophaga‐factor acts similar to auxin. Neither factor could be replaced by known phytohormones. The Roseobacter species exhibits a specific chemotactic affinity to the rhizoid cells of U. mutabilis and seems to cooperate with the Cytophaga strain and the alga by chemical communication forming a symbiotic tripartite community.  相似文献

Four-component xerogel films consisting of 1 mole-% n-octadecyltrimethoxysilane (C18) and 50 mole-% tetraethoxysilane (TEOS) in combination with 1–24 mole-% tridecafluoro-1,1,2,2-tetrahydrooctyltriethoxysilane (TDF) and 25–48 mole-% n-octyltriethoxysilane (C8) and a 1:49:50 mole-% C18/TDF/TEOS were prepared. Settlement of barnacle cyprids and removal of juvenile barnacles, settlement of zoospores of the alga Ulva linza, and strength of attachment of 7-day sporelings (young plants) of Ulva were compared amongst the xerogel formulations. Several of the xerogel formulations were comparable to poly(dimethylsiloxane) elastomer with respect to removal of juvenile barnacles and removal of sporeling biomass. The 1:4:45:50 and 1:14:35:50 C18/TDF/C8/TEOS xerogels displayed some phase segregation by atomic force microscopy (AFM) pre- and post-immersion in water. Imaging reflectance infrared microscopy showed the formation of islands of alkane-rich and perfluoroalkane-rich regions in these same xerogels both pre- and post-immersion in water. Surface energies were unchanged upon immersion in water for 48 h amongst the TDF-containing xerogel coatings. AFM measurements demonstrated that surface roughness on the 1:4:45:50 and 1:14:35:50 C18/TDF/C8/TEOS xerogel coatings decreased upon immersion in water.  相似文献

This is the first report on the antitumor and immunostimulating activities ofUlva lactuca. Using the WSM (water-soluble fraction of a methanol extract fromUlva lactuca), a concentration of 140 g/mL was found to inhibit 50% of the human leukemia cell line U937 in growth, while splenocyte growth was stimulated up to a concentration of 100 μg/mL. In addition, NO production by a macrophage cell line (RAW 264.7) and alkaline phosphatase activity in mouse splenocytes were both stimulated with 10 μg/mL of WSM. Dose-dependent patterns were observed on all three cell-lines. Accordingly, the current results indicate thatUlva lactuca may be useful as a natural antitumor and immunostimulating agent.  相似文献

Marine invertebrate grazing on temperate macroalgae may exert a significant “top-down” control on macroalgal biomass. We conducted two laboratory experiments to test (1) if consumption by the omnivorous mud snail Ilyanassa obsoleta (Say) on the macroalga Ulva lactuca Linnaeus was a function of food quality (nitrogen content) and (2) if grazing on benthic macroalgae occurred at significant rates in the presence of alternative food sources in the sediment (detritus, larvae, benthic microalgae). Grazing rates were higher for N-enriched macroalgae; however, all snails lost weight when grazing on macroalgae alone, indicating that U. lactuca was a poor food source. The presence of sediment from two sites, a sandy lagoon and an adjacent organic-rich muddy tidal creek, did not affect consumption of macroalgae in microcosm experiments, and the grazing snails were capable of significantly reducing macroalgal biomass associated with both sediment types. Grazing rates by this omnivore were as high as 10.83 mg wet weight·individuals 1·d 1 and were similar to those recorded for herbivorous species. In situ loss rates calculated from average grazing rates per individual and snail abundances (up to 3.5 g dry weight·m 2·d 1) also were comparable with those calculated for herbivorous species. This level of grazing could remove up to 88% of new macroalgal growth at the lagoon site where the N supply was relatively low but had a much smaller effect (18% of new growth) at the high-nutrient creek site. Snails facilitated macroalgal growth at both sites by increasing tissue N content by 40%–80%. Consumption and digestion of macroalgae aided in the recycling of nutrients temporarily bound in the algae and resulted in enrichment of surficial sediments. Increased N sequestration in the sediments also was associated with an interruption of snail burrowing behavior due to persistent anoxia in sediments rich in decaying algal material. Our data suggest that in shallow lagoons where mud snails and benthic macroalgae coexist, grazing may influence N retention in macroalgal biomass.  相似文献

于2009年5-7月定点采集了江苏海域绿潮藻类,测定、分析了这些藻类核糖体rDNA ITS序列,并进行分类鉴定。研究结果显示,ITS+5.8S序列片段长度为552-578 bp,其中ITS1序列部分长度为179-182 bp,5.8S序列全长为155-158 bp,ITS2序列全长为180-196 bp,序列的平均GC含量(contents)为61.6%-63.3%,不同ITS序列存在不同的插入/缺失位点。扩增得到27个序列中有7个不同序列,依据NCBI数据库相似性查找、系统进化关系及遗传距离等分析结果,确定有4个种类:浒苔(Ulva prolifeya),缘管浒苔(U.linza),石莼属(Ulva sp.)1种及盘苔属(Blidingia sp.)1种。分析表明,ITS序列可作为石莼科种类鉴定的标记。  相似文献
