Dietary fibres from Ulva lactuca (L.) Thuret (sea lettuce) and Enteromorpha compressa (L.) Grev. (A.O. nori) were measured according to a ‘standard’ method and a ‘physiological’ protocol simulating the gastric and intestinal environments. U. lactuca contained 15.8–8.0% soluble and 24.2–32.6% insoluble fibres according to the ‘standard’ and ‘physiological’ methods, respectively. For E. compressa, these values were 14.9–15.9 and 21.6–28.7%, respectively. For both algae, the composition suggests that the soluble fibres were xylorhamnoglycuronans sulphates and insoluble fibres were essentially composed of glucans. No marked chemical compositional variation was observed between soluble fractions extracted under the simulated gastric and intestinal conditions. Fibres in both algae are hydrophilic but the water holding capacities were higher after extraction of soluble fibres (5.5–9.5 g g−1 for the dry algae; 14.0–16.0 g g−1 for the standard insoluble fibres). Water soluble fibres demonstrated low intrinsic viscosities at 37 °C in buffers, particularly those from E. compressa (36.0–36.5 ml g−1), and was affected by pH for those of U. lactuca (147.5 ml g−1 at pH 3.0 and 175.0 ml g−1 at pH 7.3).  相似文献

Proteins have been extracted from the edible seaweeds Ulva rigida Agardh and Ulva rotundata Bliding using classical or enzymatic procedures. The protocols using NaOH under reductive conditions or a two-phase system (PEG/K2CO3) produced the best protein yields. The cleavage or the limitation of the linkages between proteins and polysaccharides caused by these experimental conditions probably explains the efficiency of these protocols. In SDS PAGE, the protein fraction obtained after NaOH extraction from U. rotundata is characterised by the presence of three major bands with apparent molecular weights of 45 600, 31 800 and 18 600. The protein fraction from U. rigida presents two specific bands with apparent molecular weights of about 27 000 and 12 000. These fractions are mainly rich in aspartic and glutamic acids, alanine, glycine and contain few hydroxyproline residues (0.91–2.44% total amino acid content). The use of cellulase does not significantly improve the extraction of algal proteins in comparison with the blank procedure (without enzymes). The weak accessibility of the substrates in the intact cell wall could explain these experimental data. The improvement of protein yield after the use of the polysaccharidase mixture (-glucanase, hemicellulase, cellulase) partially confirms this hypothesis.  相似文献

Synchronous zooid formation in Ulva pertusa Kjellman was induced in excised disks maintained in sterilized seawater at 20°C, 12:12 h L:D cycle and fluorescent light at 100 μmol photons m 2s 1. Zooids were reieased from mature disk tissue on the morning of the second or the third day after excision. The degree of zooid formation was found to be dependent on disk size and the region of the mother thalius from which the disk tissue was excised. Zooid formation was induced in more than 90% of small disks (0.9 mm in diameter) which were taken from the margins of the Ulva thalli. When disks were incubated together with a perforated mother thalius, the disks remained sterile. The presence of maturation inhibitors in vegetative thalli is suggested.  相似文献

A destructive disease affecting tank-cultivated Ulva rigida C. Agardh. was characterized by green spots, initially located at the base of the thalli, that spread through the host thallus and gradually caused perforations of the frond. The endophytic filamentous green alga Acrochaete geniculata (Gardner) O'Kelly was identified as the causative agent of the disease. The effects of this infection on the host varied from minor injuries to gross frond destruction. Advanced stages of infection resulted in frond wrinkling and severe tissue loss. These symptoms were caused by a primary compaction of the host cells due to a direct effect of the endophyte followed by secondary bacterial infections that became part of the degradative process initiated by the algal pathogen. Co-cultivation of healthy and diseased fronds resulted in transfer of the infection within 2 weeks; however, attempts to infect the host in the laboratory with actively reproducing suspensions of the endophyte were unsuccessful.  相似文献

Growth of the seaweed Ulva rigida C. Agardh was investigated in relation to biomass densities, internal nutrient pools and external nutrient supply. Research was carried out from 23 March to 5 July 1994 in the Sacca di Goro (Po Delta, Northern Italy), whose south-eastern part was covered by extensive mats of Ulva rigida. Two types of field experiments were conducted by incubating Ulva thalli inside large cages. In the first experiment, beginning on 23 March, 100 g of wet thalli were placed into the cages, allowed to grow for two weeks, then collected and replaced. This procedure was repeated 8 times over the study period. In the second experiment, Ulva thalli were left inside the cages and collected at selected time intervals (14, 27, 41, 64 and 76 days) in order to simulate the effects of increased density on growth and nutrient storage.We recorded specific growth rates (NGR) ranging from 0.025 to 0.081 d–1 for a period up to two months in the repeated short-term experiments performed at relatively low initial algal densities (300–500 g AFDW m–3). These NGR resulted significantly related to dissolved inorganic nitrogen (DIN) in the water column. Tissue concentrations of total Kjeldahl nitrogen (TN) were almost constant, while extractable nitrate decreased in a similar manner to DIN in the water column. Total phosphorus showed considerable variation, probably linked to pulsed freshwater inflow.In the long-term incubation experiment, NGR of Ulva was inversely related to density. Internal concentrations of both total P and TN reached maximum values after one month; thereafter P concentration remained almost constant, while TN decreased below 2% w/w (by dry weight). The TN decrease was also accompanied by an abrupt decrease in nitrate tissue concentration. The biomass incubated over the two month period suffered a progressive N limitation as shown by a decreasing NY ratio (49.4 to 14.6). The reciprocal control of Ulva against biogeochemical environment and viceversa is a key factor in explaining both resource competition and successional stages in primary producer communities dominated by Ulva. However, when the biomass exceeds a critical threshold level, approximately 1 kg AFDW m–3, the macroalgal community switches from active production to rapid decomposition, probably as a result of selfshading, biomass density and development of anaerobic conditions within the macroalgal beds.  相似文献

In situ and in vitro nitrate reductase (NR) activity assays designed for use in the field on Enteromorpha sp., Ulva sp., and Gelidium sp. are described. In optimizing each assay, a variety of compounds and assay conditions were tested for their ability to extract NR and preserve its activity. Enteromorpha sp. had similar levels of in vitro NR activity after exposure to the in situ assay buffer, demonstrating that neither NR induction nor activation likely occurs during the in situ assay. Storing freshly collected Enteromorpha sp. led to a reduction in NR activity over time. However, the use of liquid nitrogen to freeze algal tissue on site and subsequent storage at ?80° C preserved NR activity and allowed for later laboratory use of the optimized in vitro assay. Application of the in situ and in vitro assays to stands of Enteromorpha sp., Ulva sp., and Gelidium sp. in the field consistently found NR activity. In situ NR activity over 9 consecutive days in January demonstrated that Enteromorpha sp. responds to increases in nitrate availability. The influence of light on diel patterns of in vitro NR activity in the field was demonstrated for the first time as well. For the three species tested, these two assays provide a reliable tool for field investigation of the interaction between environmental signals (e.g. nutrient levels) and physiological signals (e.g. tissue metabolite levels) on nitrate reduction.  相似文献

Protoplasts were isolated from thalli of Ulva pertusa using a mixed enzyme solution of 2.0% Cellulase Onozuka R-10, 2.0% Macerozyme R-10, and 2.0% Driselase. Isolated protoplasts regenerated cell walls, developed into thalli, and propagated in large numbers under aeration in the preparative scale-culture system. Typical bioflavor compounds produced from the regenerated plants, as well as from field-collected plants, were found to be long chain aldehydes, which gave a typical seaweed odor. The long chain aldehydes were formed enzymatically from unsaturated fatty acids and released into the culture fluid. A Percoll/mannitol discontinuous density gradient separation of the heterogeneous protoplasts led to a selection of cell lines with high production of bioflavor. The cells that regenerated from protoplasts were immobilized by polymer matrices such as alginate, -carrageenan, agarose, and agar. Living cells entrapped in alginate beads in aerated cultures survived best. However, the beads started to breakdown after two months. The immobilized cells demonstrated a higher bioflavor production than did the cultured cells.  相似文献

The use of Ulva lactuca L. as an indicator of metal contamination was assessed by analysing the levels of Mn, Fe, Ni, Cu, Zn, Cd and Pb in the alga collected from 24 intertidal sites around the Island of Hong Kong. Twelve of the sites are in the rural southern parts of the Island where the coastal waters are relatively clean. The remaining 12 sites are located in the north and within Victoria Harbour which receives, apart from industrial effluents, untreated domestic sewage from a population of some 3.5 million. The mean levels of Mn, Fe, Ni, Cu, Zn and Pb in Ulva from the urban sites were respectively 4.0, 4.6, 1.8, 2.3, 2.4 and 4.6 folds those from the rural sites. However, similar levels of Cd were found in the alga amongst all the sites. Locations of high levels of metal contamination, particularly to the eastern end of the Harbour, have been identified. Preliminary results indicate that Ulva is a good indicator of Mn, Fe, Cu, Zn and Pb contaminations.  相似文献

Three species of the cosmopolitan genus Ulva (U. paschima , U. chaugulii and U. ohnoi ) from the Persian Gulf were identified using morphological and molecular markers, and were analyzed for total lipids, fatty acids, proteins and amino acid profiles. Our results show that the studied Ulva species have high protein content (9?25% dry weight), contain essential fatty acids and nearly all essential amino acids. This suggests Ulva could have potential as a form of human nutrition and animal feed with a low carbon footprint.  相似文献

Discs of thallus cut from the macroalga Ulva lactuca were incubated in filtered seawater containing cadmium, zinc, copper or cobalt (30 m). The metal uptake rates differed for each metal in the order Cu > Zn > Cd > Co. Exposure of the macroalga to metals resulted in a disruption of intracellular monovalent cation composition. Intracellular potassium was irreversibly lost and sodium was accumulated by cadmium- or copper-treated U. lactuca, which was assumed to indicate irreversible disruption of the plasmalemma. Exposure to zinc caused an increase in sodium concentrations, whereas potassium concentrations were not significantly different from the controls, suggesting that the integrity of the plasmalemma had been maintained at the zinc concentration used. Intracellular magnesium was also lost from copper-treated algae, which again indicated a loss of integrity of the cell membrane.  相似文献
