Lipomatous myopathy is a degenerative muscle pathology characterized by the substitution of muscle cells with adipose tissue, sporadically reported in cattle, pigs, and rarely in sheep, horses and dogs. This study investigated the pathology of this myopathy in 40 muscle samples collected from regularly slaughtered Piedmontese cattle living in Piedmont region (Italy). None of the animals showed clinical signs of muscular disease. Muscle specimens were submitted to histological and enzymatic investigations. Gross pathology revealed a different grade of infiltration of adipose tissue, involving multiple or single muscles. The most affected regions were the ventral abdomen and the shoulders, especially the cutaneous muscles and the muscles of the thoracic group. Morphological staining revealed an infiltration of adipose tissue varying in distribution and severity, changes in muscle fibre size and increased number of fibres with centrally located nuclei, suggesting muscle degeneration–regeneration. Necrosis and non-suppurative inflammatory cells were also seen. Furthermore, proliferation of connective tissue and non-specific myopathic changes were present. Chemical and physical characteristics of the affected tissue were also evaluated. The authors discuss about the aetiopathogenesis and classification of this muscle disorder whose histological lesions were similar to those reported in human dystrophies.  相似文献

Phytochemical characterization of the major phenolic compounds and their ultrastructural localization were carried out on onion roots (Allium cepa L.) colonized by two vesicular-arbuscular mycorrhizal (VAM) fungi: Glomus intraradix Schenck & Smith and G. versiforme (Karst.) Berch. Free and wall-bound forms of phenolic components were quantified in relation to the duration of symbiosis. Both ferulic and p-coumaric acids, as well as N-feruloyltyramine were identified as the major phenolic metabolites bound to the cell walls of VAM onion roots. Results from mycorrhized and control plants suggest the presence of a mechanism leading to the oxidative condensation of phenols, the latter process depending on the presence or absence of symbiosis. Bioassays reveal that N-feruloyltyramine induces the branching of hyphae and reduces total fungal development. The overall results lead us to suggest that the progressive binding of phenolic compounds in VAM roots is directly involved in the control of VAM endophytic establishment and development, as it gradually reduces the plasticity and elasticity of the symbiotic matrix. Phenolic compounds bound to cell walls could also be indirectly responsible for the resistance of VAM roots to pathogenic fungi, since they result in increased resistance by the cell wall to the action of digestive enzymes.Presented in part at the 31st Annual Meeting and Symposium of the Phytochemical Society of North America, Fort Collins, Colo., June 1991  相似文献

ABSTRACT. Presporogonic (blood) stages of Sphaerospora epinepheli Supamattaya, Fischer-Scherl. Hoffmann, Boonyaratpalin, 1990 were observed in the circulating blood, sinus of kidney, glomerurar capillaries and liver arteries of grouper Epinephelus malabaricus. the earliest detectable stage was a primary cell with one secondary cell. After cell divisions, nine to 16 secondary cells were found in one primary cell. Ultrastructural examination revealed electron-dense bodies (118-145 nm) in the cytoplasm of primary cells. Sporogonic stages and spores were located in Bowman's space and in kidney tubule lumens. Electron micrographs revealed a similar pattern of spore development as described from other Sphaerospora spp. Kidneys infected with S. epinepheli showed highly vacuolated tubular epithelial cells and severely affected renal corpuscles.  相似文献

Research into fish and amphibian reproduction has increased exponentially in recent years owing to the expansion of the aquaculture industry, the need to recover fishery populations, the impact of endocrine disruptors on the aquatic environment and the global decline of amphibian populations. This review focuses on a group of parasites, the Myxozoa, that affect fish and amphibian reproduction. Lists of the myxosporeans that specifically infect gonads are provided. Most of these are parasitic of freshwater hosts, and most amphibian cases are reported from testes. Sex specificity and sex reversal are discussed in relation to gonadal parasitism. The immune response of the fish to the infection is described, and the contribution of the immunoprivilege of gonads to host invasion is emphasized. The pathological effect of these parasites can be significant, especially in aquacultured broodstocks, on some occasions, leading to parasitic castration. Although myxosporean parasites are currently not very frequent in gonads, their impact could increase in the future owing to the transactions in the global market. Their easy release into the aquatic environment with spawning could make their spreading even more feasible. In the absence of commercial drugs or vaccines to treat and prevent these infections, there is an urgent need to develop specific, rapid and reliable diagnostic tools to control and manage animal movements. In addition, much effort is still to be made on deciphering the life cycle of these organisms, their invasion strategies and their immune evasion mechanisms.  相似文献

Preweaning mortality in piglets is a welfare issue, as well as an ethical and economic concern in commercial pig farming. Studying the causes of preweaning mortality and their prevalence is necessary to reduce losses. Preweaning piglet mortality was investigated in a field study including 347 sows from 14 loose-housed Norwegian piglet-producing herds. A total of 5254 piglets were born in these herds during the study period, and 1200 piglets were necropsied. The cause of death was based on pathoanatomical diagnosis (PAD). Preweaning mortality of all piglets in the study was 23.4%, including 6.3% stillborn. The two main causes of preweaning mortality in live-born piglets (n=4924) were trauma (7.1%) and starvation (2.7%). Piglets dying of an infection accounted for 2.0%. Among the necropsied piglets (n=1200), 29.1% had died due to trauma, 26.8% were categorized as stillborn and 11% had died of starvation. Piglets that had died of trauma, had a mean time of death of 1 lactation day (LD 1), ranging from LD 0 to LD 21. The mean time of death of piglets that died due to bacterial infection was LD 9, ranging from LD 0 to LD 31, with Escherichia coli accounting for most infections found in necropsied piglets. Farmers were able to identify death by trauma in piglets, but were less able to identify death due to hunger. Most piglets that died in the preweaning period, died of trauma. Surprisingly, this included large and well-fed piglets. The second most prevalent cause of preweaning mortality was starvation. Improved monitoring may reveal piglets with low body mass index, and additional nutrition may contribute to increase the survival rate.  相似文献

ABSTRACT. Ultrastructural studies on Eimeria (syn. Epieimeria ) anguillae (Apicomplexa), parasite of the digestive tract of the eel, have shown that the development of this parasite takes place completely within the host cell. Merogony and gamogony are intracellular but in the epicytoplasmic position. Sporogony is also located within the epithelial cells, which agrees with assignment of this coccidian in the family Eimeriidae. However, depending on the intensity of infection and the physiopathological reaction of the host, the gamont may behave in two ways. 1) In massive infections, gamogony stages cause a genuine destruction of intestinal epithelium. Large numbers of gamonts form nodules and parts of the seriously destroyed epithelium peel off and are released into the lumen of the gut and quickly discharged into the outer environment. This discharged epithelium envelops cells containing immature oocysts that then sporulate outside the host. 2) In light infections, the host cells, which are necrotic due to the presence of a zygote, are pushed between the surrounding intact cells towards the base of the epithelium. Closely above its basal lamella, the oocyst then undergoes sporulation. These results show no taxonomically important biological features (e.g. special mode of implantation to the host cell or active movement of the zygote). Because the morphological characteristics of Epieimeria do not differ significantly from Eimeria , we propose to suppress the genus Epieimeria Dyková and Lom, 1981, and relegate its species into the genus Eimeria .  相似文献

Cryo-electron tomography provides detailed views of macromolecules in situ. However, imaging a large field of view to provide more cellular context requires reducing magnification during data collection, which in turn restricts the resolution. To circumvent this trade-off between field of view and resolution, we have developed a montage data collection scheme that uniformly distributes the dose throughout the specimen. In this approach, sets of slightly overlapping circular tiles are collected at high magnification and stitched to form a composite projection image at each tilt angle. These montage tilt-series are then reconstructed into massive tomograms with a small pixel size but a large field of view. For proof-of-principle, we applied this method to the thin edge of HeLa cells. Thon rings to better than 10 Å were detected in the montaged tilt-series, and diverse cellular features were observed in the resulting tomograms. These results indicate that the additional dose required by this technique is not prohibitive to performing structural analysis to intermediate resolution across a large field of view. We anticipate that montage tomography will prove particularly useful for lamellae, increase the likelihood of imaging rare cellular events, and facilitate visual proteomics.  相似文献

Age is the main risk factor for the development of neurodegenerative diseases. In the aged brain, axonal degeneration is an early pathological event, preceding neuronal dysfunction, and cognitive disabilities in humans, primates, rodents, and invertebrates. Necroptosis mediates degeneration of injured axons, but whether necroptosis triggers neurodegeneration and cognitive impairment along aging is unknown. Here, we show that the loss of the necroptotic effector Mlkl was sufficient to delay age-associated axonal degeneration and neuroinflammation, protecting against decreased synaptic transmission and memory decline in aged mice. Moreover, short-term pharmacologic inhibition of necroptosis targeting RIPK3 in aged mice, reverted structural and functional hippocampal impairment, both at the electrophysiological and behavioral level. Finally, a quantitative proteomic analysis revealed that necroptosis inhibition leads to an overall improvement of the aged hippocampal proteome, including a subclass of molecular biofunctions associated with brain rejuvenation, such as long-term potentiation and synaptic plasticity. Our results demonstrate that necroptosis contributes to age-dependent brain degeneration, disturbing hippocampal neuronal connectivity, and cognitive function. Therefore, necroptosis inhibition constitutes a potential geroprotective strategy to treat age-related disabilities associated with memory impairment and cognitive decline.  相似文献

Eight Miocene odontocete partial rostra (six specimens from the Chesapeake Group of Maryland, one from the Chesapeake Group of Virginia, and another from the Hawthorn Group of Florida) exhibit periostitis, of unknown etiology, characterized by proliferative bone growth. Periostitis is an inflammation of the periosteum secondary to a predisposing event such as a fracture or infection. Computed tomography reveals that the lesions are limited to the premaxillae and that they became progressively swollen and gnarled as evidenced by the onion-like layering within the deformity. The level of maturity and degree of organization of the periostitis indicates that it likely developed over a period of months or years in these individuals. Given this length of time, these pathologies seem not to have been life-threatening despite the gross size and shape of most of these periosteal reactions. The fossils range in age from about 11 to 15 million yr and all eight rostra appear to be derived from the same, but as yet unnamed or unrecognized species of odontocete. The family from which these odontocetes derive remains unknown. Un-deformed rostra attributed to this species have not been identified, which opens the possibility that “abnormal” was the new normal for this species of odontocete.  相似文献
