Twenty-seven populations of Tuomeya, including the types of Tuomeya fluviatilis Harvey and Baileya americana Kützing, were analyzed from the entire known range of the genus: northern Newfoundland to northern Florida and east of the Mississippi River. Key morphological features were examined in detail since it has been recently proposed that Tuomeya should be reduced to a section of the genus Batrachospermum. Our observations confirmed the presence of several characteristics unique to Tuomeya: gametophyte development from a basal mass of undifferentiated cells rather than a chantransia stage, pseudoparenchymatous growth, and carpogonia with obliquely to perpendicularly attached trichogynes. Based on these findings, we conclude that the genus Tuomeya should be retained. Using multivariate morphometrics, two groupings were found that differed significantly in plant length (X = 14.1 and 24.1 mm, respectively). However, since there was no other morphometric, environmental, or geographic basis for separation of the groupings, only one species is recognized, T. americana (Kützing) Papenfuss. A convenient synthesis of [1-14C]-mono-trans fatty acid using olefin inversion as a key-step is described. This methodology allows for a facile synthesis of [1-14C]-labelled mono-trans analogues of oleic, linoleic and linolenic acids. As an example, only eleven steps were necessary to obtain the [1-14C]-mono-E isomers of linolenic acid from its commercial all-Z form. In the first step, Barton's decarboxylation procedure yielded a bromo intermediate. Epoxidation of this compound resulted in the formation of three monoepoxides, which could be separated by HPLC. After identification by 1H NMR and MS, the pure monoepoxides were then subjected to inversion consisting of a stereospecific deoxygenation followed by a beta-elimination step. Finally, the labelling was introduced by substitution of the bromine by a [14C]-cyano group followed by hydrolysis. 相似文献

The study purpose is to optimize modeling parameters, specifically segmentation spacing and centerline extraction, to efficiently construct accurate 3D aortic models. Models are constructed by centerline extraction and orthogonal 2D segmentations. We examine the effect of segmentation interval spacing (2, 1, 0.5, 0.25 cm) and orthogonal segmentation and centerline extraction iteration (one, two, three iterations) for constructing models of Healthy, Tortuous, Aneurysmal, and Dissected human thoracic aortas. Aortic arclength, curvature, and cross-sectional axis ratio were computed to compare variations in modeling parameters. Centerline arclength is precisely characterized for all aortas with a single iteration of centerline extraction (≤1% deviation), however, complex anatomies required 1 cm segmentation intervals whereas the Healthy aorta only required 2 cm intervals. Centerline curvature is more sensitive to modeling methods, requiring 1 cm intervals for ≤5% deviation in peak curvature for the three diseased anatomies, and two iterations of segmentation and centerline extraction for the Aneurysmal and Dissected aortas. Accurate lumen cross-sectional characterization required 1 or 0.5 cm segmentation intervals, and two or three segmentation and centerline iterations, with greater refinement needed for more complex geometries. Depending on the geometric characteristic and complexity of anatomy and pathology, different levels of segmentation interval refinement and iterations of segmentation and centerline extraction are required. 相似文献

DNA-binding agents have been considered as an established opportunity for the development of anticancer drugs and DNA fluorescence probes. This work reported the synthesis of two novel carbazole derivatives (1 and 2) and investigated their DNA binding properties, living cell image, and cytotoxicity. The results demonstrated that both compounds presented the higher binding affinity to G-quadruplex than to duplex DNA by means of UV–Vis absorption titration and fluorescent intercalator displacement. Continuous variation analysis indicated that their binding stoichiometries of the compound/G-quadruplex were near 2 except the compound/bcl-2. Circular dichroism spectra showed that both compounds had no influence on the conformation of G-quadruplexes. Fluorescence titrations indicated that 2 had the potential to be a G-quadruplex selective fluorescent probe, while 1 could be used as a fluorescent probe for duplex DNA. Confocal fluorescence images indicated that both compounds could enter the living HepG2 cells, and 1 mainly located in nucleus whereas 2 mainly distributed in cytoplasm. DNase and RNase digest tests indicated that both compounds could enter into the nucleus and interact with duplex DNA, especially, 2 could interact with RNA and/or G-quadruplex DNA. They also exhibited an obvious antiproliferative activity to HepG2 by using MTT assays, with IC50 values of 2.7 and 9.5?μM for 1 and 2, respectively. 相似文献

X-ray absorption and phase contrast imaging to study the interplay between plant roots and soil structure 总被引:2,自引:0,他引:2
Plant performance is, at least partly, linked to the location of roots with respect to soil structure features and the micro-environment
surrounding roots. Measurements of root distributions from intact samples, using optical microscopy and field tracings have
been partially successful but are imprecise and labour-intensive. Theoretically, X-ray computed micro-tomography represents
an ideal solution for non-invasive imaging of plant roots and soil structure. However, before it becomes fast enough and affordable
or easily accessible, there is still a need for a diagnostic tool to investigate root/soil interplay. Here, a method for detection
of undisturbed plant roots and their immediate physical environment is presented. X-ray absorption and phase contrast imaging
are combined to produce projection images of soil sections from which root distributions and soil structure can be analyzed.
The clarity of roots on the X-ray film is sufficient to allow manual tracing on an acetate sheet fixed over the film. In its
current version, the method suffers limitations mainly related to (i) the degree of subjectivity associated with manual tracing
and (ii) the difficulty of separating live and dead roots. The method represents a simple and relatively inexpensive way to
detect and quantify roots from intact samples and has scope for further improvements. In this paper, the main steps of the
method, sampling, image acquisition and image processing are documented. The potential use of the method in an agronomic perspective
is illustrated using surface and sub-surface soil samples from a controlled wheat trial. Quantitative characterization of
root attributes, e.g. radius, length density, branching intensity and the complex interplay between roots and soil structure,
is presented and discussed.

This investigation presents a look back to ancient times of karyology with modern optical instruments. `Cryptopolyploidy',
i.e. an intrinsically polyploid but numerically non-polyploid structure of chromosome complements, today is an obsolete concept
of chromosome architecture and evolution, but was actively discussed up to the mid-seventies of the past century. We focus
here at a hypothesis of cryptooctoploidy in Vinca difformis (2n = 46), which was based on a measured four-fold chromosome volume compared with V. minor (2n = 46), the proposed diploid. We used DNA flow cytometry and Feulgen densitometry to see, if the postulate of cryptooctoploidy
in V. difformis in the retrospect could be justified. It was found not defendable, because V. difformis differed only about 1.55-fold in C-value from V. minor, which is far from a regular multiple and much less than the 4-fold. C-values are given also for V. major, V. herbacea and V. rosea. 相似文献

Three-dimensional pore space quantification of apple tissue using X-ray computed microtomography 总被引:1,自引:0,他引:1
The microstructure and the connectivity of the pore space are important variables for better understanding of the complex
gas transport phenomena that occur in plant tissues. In this study, we present an experimental procedure for image acquisition
and image processing to quantitatively characterize in 3D the pore space of apple tissues (Malus domestica Borkh.) for two cultivars (Jonagold and Braeburn) taken from the fleshy part of the cortex using X-ray computer microtomography.
Preliminary sensitivity analyses were performed to determine the effect of the resolution and the volume size (REV, representative
elementary volume analysis) on the computed porosity of apple samples. For comparison among cultivars, geometrical properties
such as porosity, specific surface area, number of disconnected pore volumes and their distribution parameters were extracted
and analyzed in triplicate based on the 3D skeletonization of the pore space (medial axis analysis). The results showed that
microtomography provides a resolution at the micrometer level to quantitatively analyze and characterize the 3D topology of
the pore space in apple tissue. The computed porosity was confirmed to be highly dependent of the resolution used, and the
minimum REV of the cortical flesh of apple fruit was estimated to be 1.3 mm3. Comparisons among the two cultivars using a resolution of 8.5 μm with a minimum REV cube showed that in spite of the complexity
and variability of the pore space network observed in Jonagold and Braeburn apples, the extracted parameters from the medial
axis were significantly different (P-value < 0.05). Medial axis parameters showed potential to differentiate the microstructure between the two evaluated apple

江苏省中国科学院植物研究所标本馆(NAS)是中国最早的植物标本馆之一,也是国内最早开展植物标本数字化的标本馆,其标本数字化发展经历了4个阶段: 20世纪80年代后期尝试的标本文字信息数字化的起步阶段; 20世纪90年代末的标本图像数字化和文字信息数字化规范阶段; 2004年以后的标本批量数字化与信息网络共享快速发展阶段; 2018年后的标本数字化信息维护与优化阶段。这一过程集中代表和反映了中国植物标本数字化的发展历程。此外,近年来开始了发掘和利用江苏植物标本的数字化信息工作,包括建设江苏省级数字植物标本馆、开发江苏省维管植物标本时空分布可视化系统、开展标本采集-入库过程数字化等。今后,将不断深化标本数字化的工作,以期形成有NAS特色的数字化植物标本馆。 相似文献

This work studied the effects of ultrasound-assisted liquid-state fermentation on the content of peptides and bioactivity of fermented soybean meal (FSBM) with Bacillus subtilis. Based on the optimization of basic ultrasonic parameters, effects of the main parameters including power density, incubation temperature and ultrasonic time on in vitro ACE inhibitory activity, peptides content and biomass of B. subtilis were investigated via Box-Behnken experimental design. Three mathematical models were determined to show the effects of each variable and their combinatorial interactions on dependent variables. A set of preferred condition was determined to be at power density of 0.08 W/mL for 1 h with an initial incubation temperature of 36.7 °C, according to the desirability function analyses. Under these conditions, the experimental values of in vitro ACE inhibitory activity, peptides content and biomass increased by 26.4%, 36.2% and 55.0% compared to those of the control, respectively, accompanied by the improvement of the peptides yield, purity and conversion rate in varying degrees. Additionally, FSBM resulted in a significant decrease of systolic blood pressure by 20.7 ± 1.8 mmHg in spontaneously hypertensive rats at 3 h. These results suggest that ultrasonication is an efficient solution for enhancing fermentation to produce bioactive components from agroindustrial by-products. 相似文献

The occurrence and development of ischemic stroke are closely related to cerebral blood flow. Real‐time monitoring of cerebral perfusion level is very useful for understanding the mechanisms of the disease. A wide field of view (FOV) is conducive to capturing lesions and observing the progression of the disease. In this paper, we attempt to monitor the whole‐brain microcirculation in middle cerebral artery occlusion (MCAO) rats over time using a wide FOV swept‐source OCT (SS‐OCT) system. A constrained image registration algorithm is used to remove motion artifacts that are prone to occur in a wide FOV angiography. During ischemia, cerebral perfusion levels in the left and right hemispheres, as well as in the whole brain were quantified and compared. Changes in the shape and location of blood vessels were also recorded. The results showed that the trend in cerebral perfusion levels of both hemispheres was highly consistent during MCAO, and the position of the blood vessels varied over time. This work will provide new insights of ischemic stroke and is helpful to assess the effectiveness of potential treatment strategies. 相似文献
