Cidea蛋白调节脂肪代谢,在机体能量平衡过程中起重要作用,在转录和翻译后水平受到严格调控,但在翻译水平的调节还不清楚.通过对CIDEA基因敲除小鼠模型研究,鉴定了小鼠棕色脂肪组织内源性表达Cidea蛋白N端缺失异构体mCidea-22.定点突变等研究表明其产生机制为选择性起始翻译.并且,在异位表达时,N端缺失异构体和全长异构体的比例呈现细胞系特异性.此外,蛋白质稳定性实验表明mCidea-22半衰期很短.亚细胞定位研究显示mCidea-22是内质网和脂滴定位蛋白.为深入理解Cidea蛋白的功能和精细调节提供了新的思路和方向.  相似文献

The N-carbamoyl- -amino acid amidohydrolase ( -carbamoylase) gene (dcb) from Agrobacterium tumefaciens AM 10 was cloned by polymerase chain reaction in plasmid pET28a and was overexpressed in Escherichia coli JM109 (DE3). However, almost 80% of the enzyme remained trapped in inclusion bodies. To facilitate the expression of the properly folded active enzyme, the chaperones GroEL/ES were coexpressed in plasmid pKY206. This resulted in a 43-fold increase in active enzyme production compared to the wild-type strain. The histidyl-tagged -carbamoylase was purified by a single step nickel-affinity chromatography to a specific activity of 9.5 U/mg protein.  相似文献

We used a novel mouse model to study the effects of selectivebreeding for high locomotor activity (14 generations) on relativeorgan sizes, hematocrit (Hct), and blood hemoglobin (Hb) concentration.We also examined effects of exercise training and genotype-by-environmentinteractions by housing animals for 8 weeks with wheels thatwere either free to rotate or locked. Mice from the four replicateHigh-Runner (HR) lines were smaller in total body mass but hadlarger body mass-adjusted kidneys relative to the four Controllines (P < 0.05). Control and HR lines did not differ significantlyfor mass-adjusted tail length or masses of the "triceps surae"hindlimb muscle group, heart (ventricle), spleen, liver, adrenalglands or gonads. Wheel access caused a reduction in body massand an increase in relative heart mass. In females only, wheelaccess caused a reduction in relative spleen mass. Wheel accessdid not affect relative tail length or relative mass of thetriceps surae, liver, adrenal gland or gonads. Significant interactionsbetween selection history and wheel access were observed infemales for spleen, liver, and gonad mass as well as Hct andHb. Wheel access caused increases in both Hct and Hb, mainlyin the HR lines. The mini-muscle phenotype, caused by a Mendelianrecessive allele that halves hindlimb muscle mass, was significantlyassociated with several other body composition traits, includingreduced body mass, increased tail length, increased heart mass,increased liver mass (females only), increased mean adrenalgland mass (females only), increased mean kidney mass (malesonly), and reduced Hct (wheel-access females only). Resultsare discussed in context of the beneficial acclimation hypothesis,genotype-by-environment interactions, and the potential for"nurture" to be self-reinforcing of "nature" in some complexbehavioral-physiological phenotypes.  相似文献

An auxin analog, 2,4-D, stimulates the activity of endo-1,4-beta-glucanase (EGase) in rice (Oryza sativa L.). The auxin-induced activity from three protein fractions was purified to homogeneity from primary root tissues (based on SDS-PAGE and isoelectric focusing after Coomassie brilliant blue staining). Amino acid sequencing indicated that the 20 N-terminal amino acid sequence of the three proteins was identical, suggesting that these proteins may be cognates of one EGase gene. An internal amino acid sequence of the the rice EGase (LVGGYYDAGDNVK) revealed that this enzyme belongs to glycosyl hydrolase family 9 (GHF9). The major isoform of this rice GHF9 [molecular weight based on matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS): 51,216, isoelectric point (pI): 5.5] specifically hydrolyzed 1,4-beta-glycosyl linkages of carboxymethyl (CM)-cellulose, phosphoric acid-swollen cellulose, 1,3-1,4-beta-glucan, arabinoxylan, xylan, glucomannan, cellooligosaccharides [with a degree of polymerization (DP) >3] and 1,4-beta-xylohexaose, indicating a broader substrate range compared with those of other characterized GHF9 enzymes or EGases from higher plants. Hydrolytic products of two major hemicellulosic polysaccharides in type II cell walls treated with the purified enzyme were profiled using high-performance anion exchange chromatography (HPAEC). The results suggested that endolytic attack by rice EGase is not restricted to either the cellulose-like domain of 1,3-1,4-beta-glucan or the unsubstituted 1,4-beta-xylosyl backbone of arabinoxylan, but results in the release of smaller oligosaccharides (DP <6) from graminaceous hemicelluloses. The comparatively broader substrate range of this EGase with respect to beta-1,4-glycan backbones (glucose and xylose) may partly reflect different roles of gramineous and non-gramineous GHF9 enzymes.  相似文献

The unc-52 gene of Claenorhabditis elegans encodes a homologue of the basement membrane heparan sulfate proteoglycan perlecan. Viable alleles reduce the abundance of UNC-52 in late larval stages and increase the frequency of distal tip cell (DTC) migration defects caused by mutations disrupting the UNC-6/netrin guidance system. These unc-52 alleles do not cause circumferential DTC migration defects in an otherwise wild-type genetic background. The effects of unc-52 mutations on DTC migrations are distinct from effects on myofilament organization and can be partially suppressed by mutations in several genes encoding growth factor-like molecules, including EGL-17/FGF, UNC-129/TGF-beta, DBL-1/TGF-beta, and EGL-20/WNT. We propose that UNC-52 serves dual roles in C. elegans larval development in the maintenance of muscle structure and the regulation of growth factor-like signaling pathways.  相似文献

We have developed a cell culture procedure that can produce large quantities of confluent monolayers of primary human fetal retinal pigment epithelium (hfRPE) cultures with morphological, physiological and genetic characteristics of native human RPE. These hfRPE cell cultures exhibit heavy pigmentation, and electron microscopy show extensive apical membrane microvilli. The junctional complexes were identified with immunofluorescence labeling of various tight junction proteins. Epithelial polarity and function of these easily reproducible primary cultures closely resemble previously studied mammalian models of native RPE, including human. These results were extended by the development of therapeutic interventions in several animal models of human eye disease. We have focused on strategies for the removal of abnormal fluid accumulation in the retina or subretinal space. The extracellular subretinal space separates the photoreceptor outer segments and the apical membrane of the RPE and is critical for maintenance of retinal attachments and a whole host of RPE/retina interactions.  相似文献
