In the conversion of quinolinic acid to 6-hydroxypicolinic acid by whole cells of Alcaligenes sp. strain UK21, the enzyme reactions involved in the hydroxylation and decarboxylation of quinolinic acid were examined. Quinolinate dehydrogenase, which catalyzes the first step, the hydroxylation of quinolinic acid, was solubilized from a membrane fraction, partially purified, and characterized. The enzyme catalyzed the incorporation of oxygen atoms of H2O into the hydroxyl group. The dehydrogenase hydroxylated quinolinic acid and pyrazine-2,3-dicarboxylic acid to form 6-hydroxyquinolinic acid and 5-hydroxypyrazine-2,3-dicarboxylic acid, respectively. Phenazine methosulfate was the preferred electron acceptor for quinolinate dehydrogenase. 6-Hydroxyquinolinate decarboxylase, catalyzing the nonoxidative decarboxylation of 6-hydroxyquinolinic acid, was purified to homogeneity and characterized. The purified enzyme had a molecular mass of approximately 221 kDa and consisted of six identical subunits. The decarboxylase specifically catalyzed the decarboxylation of 6-hydroxyquinolinic acid to 6-hydroxypicolinic acid, without any co-factors. The N-terminal amino acid sequence was homologous with those of bacterial 4,5-dihydroxyphthalate decarboxylases.  相似文献

Species differences in the response to dietary MoO42? as a metabolic antagonist of Cu are considered briefly. Suggestions that (i) the potency of MoO42? as a Cu antagonist is enhanced by normally innocuous dietary concentrations of S2? and (ii) that MoS42? may be a more effective antagonist than either MoO42? or S2? were investigated in a series of studies with rats. Diets including MoS42? but not of MoO42? or S2? alone promoted a decline in hepatic Cu and ceruloplasmin activity and induced clinical signs of Cu deficiency. Evidence of concurrent anomalies in the partition of Cu between tissues and in the distribution of Cu between proteins of plasma and kidney cytosol suggested that such effects were partly attributable to the development of systemic defects in Cu metabolism. The relationship of such findings to the suggested involvement of MoS42? or its derivatives in the etiology of Mo-induced Cu deficiency in ruminant animals is considered.  相似文献

Proton nmr spectroscopic evidence is presented for methylmercury(II) binding to the deprotonated amino groups in adenosine, 9-methyladenine, guanosine, 1-methylguanosine, and cytidine under basic conditions. Except for the guanosine case, 1H nmr spectra of the products from aqueous or ethanolic 1:1 mixtures of substrate and MeHgOH are consistent with methylmercuration of the deprotonated amino groups. Guanosine undergoes initial binding of MeHg to N1, and a second equivalent of MeHgOH is necessary to effect amino binding. The nmr spectra of the complexed adenine derivatives suggest that different geometrical isomers exist in (CD3)2SO solution, reflecting the partial double bond character of the C6N bond in these systems. Using a correlation relating the magnitude of the 199Hg-1H coupling constant (J) for MeHg-ligand complexes with the ligand pKa (J = ?3.88 pKa + 248.5, extending over 13 pK units, based on a variety of N and O donor ligands), estimates (± 0.3 pK unit) of the pKas of the amino groups of the above substrates have been made. In this way, pKa values of 15.5 (cytidine), 17.0 (adenosine and 9-methyladenine), 15.1 (guanosine), and 14.9 (1-methylguanosine) are obtained. In the cases where comparisons with literature pKa data can be made, good agreement is found.  相似文献

Aim To investigate environmental variation and associated assemblage changes of carabid beetles along an urban–rural gradient. Location ‘Quercus–Acer’ (oak–sycamore) woodlands in the city of Birmingham, UK. Methods We collected carabid data using pitfall traps on 12 sites in the city. The traps were run from April–September in 2000, and we collected environmental data on 24 individual variables associated with the individual sites and their landscape context. Changes in carabid assemblages were analysed using repeat measures anova and the environment–species relationships with a Redundancy Analyses (RDA) and Generalized Linear Modelling (GLM). Results We found that: (1) species richness and diversity were lower in the urban and suburban zone and higher in the rural zone; (2) Berger Parker dominance index was higher in the urban and suburban zones; (3) the number of woodland and woodland associated species was significantly higher at the rural end of the gradient; (4) the number of short‐winged (brachypterous) species was highest in the rural zone and decreased towards the urban woodlands, whereas the long‐winged species were more abundant in suburban woodlands; (5) the median weight length (WML) of the assemblage declined along the gradient from the rural to the urban zone, as did the number of large species; and (6) five of the 24 environmental variables showed a significant relationship with variation in the carabid assemblage. At site level the carabid assemblages were related to the level of site disturbance and soil penetrability, whereas site size and amount of woodland and urban land within 5 km of the site were important at a larger landscape scale. Main conclusions The results suggest that urbanization has a deleterious impact on carabid assemblages, causing a reduction in species richness from the rural fringe to the centre of the city. Changes in assemblage structure were related to woodland fragmentation, which led to variations in woodland size, woodland location and site disturbance due to trampling. Large, flightless and specialist woodland species are more susceptible to changes associated with urbanization, presumably due to their longer life spans, lower reproductive rates, more specialized niches and more limited dispersal potential.  相似文献

An emerging consensus among scholars of Muslim political and social identity suggests that Western Muslims live out an anti-essentialist critique of identity construction. Considering this view, this paper examines a cross-national comparison of British Bangladeshis in London and Spanish Moroccans in Madrid that solicits the perceptions of working-class Muslim men. While the results indeed reaffirm respondents' concomitant relationships to a variety of identity paradigms, interview content demonstrates that subjects' multiplicity is complicated by their desire to meet – not reject – the essentialist standards of belonging to the identity paradigms discursively available to them. Rather than defiantly cherry-picking preferred characteristics of religion, ethnicity and nationality, individuals' responses suggest that they are trying to fulfil perceived standards of authenticity. Such a contention helps explain the prevalence of Western Muslims' expressed and well-documented ‘identity crisis’, suggests the enduring relevance of identity essentialisms, and more broadly, complicates post-modern conceptions of identity formation.  相似文献

We have collected live and dead foraminiferal times-series data at 2-weekly intervals for a 12-month period from the intertidal zone of Cowpen Marsh, Tees Estuary, UK. The data from the 689 samples show profound differences between live and dead assemblages, although assemblages are dominated by just three species, Haynesina germanica, Jadammina macrescens and Trochammina inflata, which represent over 70% of the assemblage. The cumulative increase in species of most environments approximates to a lognormal or log series. None of the datasets show a broken stick pattern. The cumulative maximum number of species, which represents the species carrying capacity of the environment, is recorded earlier in the life assemblages than the dead counterparts. The dead assemblage of Cowpen Marsh is found to have a higher abundance (435 compared to 163 individuals/10 cm3) and number of species (52 compared to 28) than its live counterpart because the dead assemblage represents many generations added over a long period of time. In contrast, some species are recorded in the live dataset that were not found in the dead assemblage, indicating the dead record is either incomplete (e.g. taphonomic change) or inadequately sampled.We investigated the influence of patterns in cumulative increase on dead assemblages for sea-level reconstructions through the development of foraminiferal-based transfer functions. The cumulative transfer functions suggest that the performance improves during the first six sample intervals of the time-series dataset with reconstruction differing by 1.2 m and remains constant thereafter.  相似文献

Uridine monophosphate (UMP) kinase is a conserved enzyme that catalyzes the ATP-driven conversion of uridylate monophosphate into uridylate diphosphate, an essential metabolic step. In prokaryotes, the enzyme exists as a homohexamer that is regulated by various metabolites. Whereas the enzymatic mechanism of UMP kinase (UK) is well-characterized, the molecular basis of its regulation remains poorly understood. Here we report the crystal structure of UK from Bacillus anthracis (BA1797) in complex with ATP at 2.82 Å resolution. It reveals that the cofactor, in addition to binding in the active sites, also interacts with separate binding pockets located near the center of the hexameric structure. The existence of such an allosteric binding site had been predicted by biochemical studies, but it was not identified in previous crystal structures of prokaryotic UKs. We show that this putative allosteric pocket is conserved across different bacterial species, suggesting that it is a feature common to bacterial UKs, and we present a structural model for the allosteric regulation of this enzyme.  相似文献

Recent surveys have established that the belted beauty moth Lycia zonaria britannica, which is an endemic subspecies found only in the British Isles, now occurs at just three sites in England and Wales. Populations at all three sites are small, with annual peak counts of usually less than 50 females. At Morfa Conwy in Wales and Meols Common in England, habitat quality has been declining for more than a decade as open, herb-rich, early successional dune grassland is replaced by fixed, rank grassland. Uniquely at Sunderland Point in England, the moth is primarily associated with areas of saltmarsh, but distribution is constrained by surrounding farmland and man-made structures. Attempts are now being made to restore areas of open grassland and to return the moth to its former distribution. Since the winter of 2000-2001, small plots of rank grassland on Meols Common have been excavated annually, with all vegetation removed, together with any humic layer present. The exposed bare sand has then been allowed to re-vegetate naturally. Within 18 months, the plots have developed into open, herb-rich habitats suitable for the moth. Adults were recorded on the plots for the first time in spring 2003 and may have been the product of the small numbers of eggs and larvae introduced to the plots in spring 2002. Similar excavations have been initiated at Morfa Conwy, which is in the process of being notified as a Site of Special Scientific Interest (SSSI). A 3-year programme to establish a second Welsh population is underway and, to date, more than 2000 captive-reared larvae have been released on the receptor site which lies 21 km to the east of Morfa Conwy.  相似文献
