The mouse immunoglobulin (IgM) pre-mRNA contains a splicing inhibitor that bears multiple binding sites for the splicing repressor polypyrimidine tract binding protein (PTB). Here we show that the inhibitor directs assembly of an ATP-dependent complex that contains PTB and U1 and U2 small nuclear RNAs (snRNAs). Unexpectedly, although U2 snRNA is present in the inhibitor complex, it is not base-paired to the branch point. We present evidence that inhibitor-bound PTB contacts U2 snRNA to promote base-pairing to an adjacent branch point–like sequence within the inhibitor, thereby preventing the U2 snRNA–branch point interaction and resulting in splicing repression. Our studies reveal a novel mechanism by which PTB represses splicing.  相似文献

目的:明确LRIG2蛋白在人催乳素腺瘤细胞中的表达与定位。方法:采用免疫细胞化学方法检测LRIG2蛋白在人催乳素腺瘤原代细胞中表达情况,人胶质瘤细胞系U87细胞设为阳性对照。结果:LRIG2蛋白在原代培养的人催乳素腺瘤细胞中高表达(86.6±2.15)%,与其在U87细胞中表达率无明显统计学差异;同时免疫细胞化学结果提示LRIG2蛋白在人催乳素腺瘤细胞中定位于胞浆,也与其在U87细胞中表达一致。结论:LRIG2蛋白在人催乳素腺瘤细胞中高表达,定位于胞浆,提示其可能在垂体腺瘤发生、发展过程中发挥作用,为进一步研究垂体腺瘤发生机制奠定基础。  相似文献

We developed five microsatellites from Ulmus minor using an AFLP protocol followed by an enrichment step, with biotinylated primers containing tandem repeats. Ciguatera fish poisoning (ciguatera) is a common clinical syndrome in areas where there is dependence on tropical reef fish for food. A subset of patients develops recurrent and, in some instances, chronic symptoms, which may result in substantial disability. To identify possible biomarkers for recurrent/chronic disease, and to explore correlations with immune gene expression, peripheral blood leukocyte gene expression in 10 ciguatera patients (7 recurrent and 3 acute) from the U.S. Virgin Islands, and 5 unexposed Florida controls were evaluated. Significant differences in gene expression were noted when comparing ciguatera patients and controls; however, it was not possible to differentiate between patients with acute and recurrent disease, possibly due to the small sample sizes involved.  相似文献

Abstract The human histiocytic lymphoma cell line U937 can be induced to differentiate along the monocyte/macrophage pathway by either phorbol myristate acetate (PMA) or by the combination of retinoïc acid (RA) and 1,25-dihydroxyvitamin D3 (VD). U937 cells treated with either PMA or RA/VD were able to phagocytose Salmonella typhimurium in the presence of non-immune human serum. However, only cells differentiated by RA/VD were capable of developing an oxidative metabolic burst in response to infection. Since the oxidative burst is considered to be a potent antimicrobial mechanism, we investigated its effect on S. typhimurium . The oxidative burst failed to affect either the viability or the multiplication of S. typhimurium suggesting that if plays only a minor role in the host defence against S. typhimurium .  相似文献

Sperm whales ( Physeter macrocephalus ) and beaked whales ( Mesoplodon spp. and Ziphius cavirostris ) are deep-diving cetaceans that frequent shelf-edge and Gulf Stream waters off the northeast U. S. coast. Sighting data collected during seven summer (1990, 1991, 1993, and 1995–1998) shipboard surveys were analyzed using a geographic information system to determine habitat use based on bathymetric and oceanographic features. Although sighting rates were lower for beaked whales, both taxa occupied similar habitats. Beaked whales were concentrated at the colder shelf edge, whereas sperm whales were associated with warmer off-shelf water. Mean sighting rates for both taxa were higher in canyon features, but only beaked whale sighting rates were significantly different between canyon and non-canyon habitat (Wilcoxon signed rank test P = 0.007). Within the shared habitat, the two taxa were separated at fine-scale based on oceanographic features.  相似文献

ABSTRACT.   Dispersal is a critical link between organismal and population biology, yet, because of their mobility, our understanding of the causes and consequences of long-distance dispersal by birds remains poorly known. Methods used to study dispersal include (1) marking and recapturing individuals in a limited study area to estimate survival and dispersal rates, and (2) relying on volunteers to mark and recapture individuals over larger areas. We compared these two methods for measuring dispersal distances of Tree Swallows ( Tachycineta bicolor ) using recapture data from a limited-area study in New York State (the Swallow Dispersal Study, SDS) and the recapture dataset from the U.S. Bird Banding Laboratory (BBL). Analysis of BBL records revealed a difference in the dispersal distance distributions (DDD) for data reported before and after 1967. In the earlier data, 84% of the 238 records were for birds within 13.6 km of their first banding location, whereas only 22% of the 799 records in the more recent data were reported in this closest distance belt. These differences are almost certainly due to changes in reporting protocols instituted by the BBL in the mid-1960s. We corrected for recapture effort in the SDS, and, using this corrected SDS data for the proportion of birds returning in the closest distance belt and the recent BBL for the proportions of more distant movements, we created what we think is the best composite DDD for Tree Swallows. Even though dispersal distances up to 2367 km have been reported, the composite DDD indicates that fewer than 3% of birds disperse more than 100 km and that 85% disperse less than 15 km between years. Thus, our results suggest that the dispersal behavior of most individuals can be examined effectively at more local spatial scales. Studies of dispersal and mortality would be facilitated if all recaptures of banded birds were reported with accurate spatial coordinates to the Bird Banding Laboratory.  相似文献

研究人巨细胞病毒(HCMV)感染对神经胶质瘤U87细胞自噬的影响。通过观察微管相关蛋白1轻链3(LC3)、自噬相关基因Beclin1及其蛋白表达的变化,从而探讨HCMV与神经胶质瘤发生、发展的关系及意义。用HCMV AD169(MOI=5)感染神经胶质瘤U87细胞,同时将未感染HCMV的U87细胞作为对照组。分别在6、12、24、48 h用RT-PCR检测Beclin1的表达,Western-blot和免疫荧光检测Beclin1和LC3编码蛋白的表达,最后用CCK-8检测细胞的增殖活性。结果显示,HCMV感染的U87细胞LC3-II蛋白表达水平逐渐下降(P<0.05);同时,HCMV感染的U87细胞Beclin1基因及蛋白的表达水平也逐渐下降(P<0.01),且HCMV感染U87细胞增殖显著(P<0.01)。以上结果表明,HCMV感染抑制胶质瘤U87细胞自噬,并会引起Beclin1表达水平下调,进而导致胶质瘤细胞增殖。  相似文献

Membrane lipid peroxidation processes yield products that may react with proteins to cause oxidative modification. Recently, we demonstrated that the control of cytosolic and mitochondrial redox balance and oxidative damage is one of the primary functions of NADP+-dependent isocitrate dehydrogenase (ICDH) through to supply NADPH for antioxidant systems. When exposed to lipid peroxidation products, such as malondialdehyde (MDA), 4-hydroxynonenal (HNE) and lipid hydroperoxide, ICDH was susceptible to oxidative damage, which was indicated by the loss of activity and the formation of carbonyl groups. The structural alterations of modified enzymes were indicated by the change in thermal stability, intrinsic tryptophan fluorescence and binding of the hydrophobic probe 8-anilino 1-napthalene sulfonic acid. Upon exposure to 2,2′-azobis(2-amidinopropane) hydrochloride (AAPH), which induces lipid peroxidation in membrane, a significant decrease in both cytosolic and mitochondrial ICDH activities were observed in U937 cells. Using immunoprecipitation and immunoblotting, we were able to isolate and positively identify HNE adduct in mitochondrial ICDH from AAPH-treated U937 cells. The lipid peroxidation-mediated damage to ICDH may result in the perturbation of the cellular antioxidant defense mechanisms and subsequently lead to a pro-oxidant condition.  相似文献
