Transgenic white poplar (Populus alba L.) plants expressing a novel Arabidopsis thaliana cysteine proteinase inhibitor (Atcys) gene have been produced using Agrobacterium tumefaciens-mediated gene transfer. Internodal stem segments of cv. Villafranca were co-cultivated with the EHA105 pBI-Atcys A. tumefaciens strain. Sixteen putative transgenic plant lines were regenerated from different calli with a transformation efficiency of 11%. The integration and expression of the cysteine proteinase inhibitor (Atcys) gene into the plant genome was confirmed by Southern and northern blot analyses. Papain inhibitory activity was detected in poplar transgenic tissues by means of a specific in vitro assay. Such activity was sufficient to inhibit most of the digestive proteinase activity of chrysomelid beetle (Chrysomela populi L.) and confer resistance to C. populi larvae on selected transgenic plants. A close correspondence between the inhibition of papain and resistance to poplar leaf beetle was observed in all tested transgenic lines. Our results indicate that Atcys could be succesfully employed in breeding programmes aimed at the selection of new poplar genotypes resistant to major insect pests.  相似文献

未来全球变化 ,尤其降水变化的进一步加剧 ,将对毛乌素沙地陆地生态系统分布格局和生产力带来巨大影响。为了预测未来变化 ,提出合理的对应策略 ,实验选择毛乌素沙地 3种优势灌木、半灌木为研究对象 ,人工控制 4种降水量水平来探讨水分平衡对全球降水量变化的响应。结果表明 ,沙地蓄水量和不同沙层深度含水量季节变化明显受到不同施水量和 3种植物种类的影响。 3种植物沙地蓄水量和不同深度沙层含水量随着施水量的增加逐渐增大 ,并且不同施水量和植物沙地含水量随深度增加逐渐增大。在同一施水量处理下的蓄水量和同一深度含水量均大体为杨柴 (HedysarummongolicumTurcz.) >沙柳 (SalixpsammophilaC .WangetCh .Y .Yang) >油蒿 (Artemisiaordosi caKrasch .) ;3种植物的蒸发量和蒸腾量季节变化明显受到施水量的影响。随着施水量的增加 ,即 15 7 5、315 0、472 5和 6 30 0mm ,蒸发总量依次为 12 3.6 6、2 5 8.6 8、376 .30、45 8.5 7mm ;蒸腾总量 ,油蒿分别为 5 0 .80、6 8.93、10 8.39、16 3.36mm ;沙柳分别为 47.37、6 8.17、93.6 5、135 .97mm ;杨柴分别为 46 .73、6 7.37,86 .0 7、10 9.6 4mm。蒸发量显著大于蒸腾量  相似文献

Phosphite is a cost-effective fungicide used to control the pathogen Phytophthora cinnamomi which is damaging the diverse flora of the southwest of Western Australia. Three annual species of the southwest jarrah (Eucalyptus marginata) forest of Western Australia (Pterocheata paniculata, Podotheca gnaphalioides and Hyalosperma cotula), were studied to determine the effect of the fungicide phosphite on the species’ reproduction. Phosphite at concentrations of 2.5, 5 and 10 g L–1 reduced pollen fertility of Pt. paniculata when plants were sprayed at the vegetative stage. Pollen fertility of all three species was reduced when plants were sprayed at anthesis with 10 g L–1 phosphite. Seed germination was reduced by phosphite in Pt. paniculata and H. cotula when plants were sprayed in the vegetative stage. Phosphite when sprayed at anthesis at a concentration of 5 g L–1 reduced seed germination of H. cotula. Phosphite at concentrations of 5 and 10 g L–1 killed a proportion of plants from all three species and up to 90% of Po. gnaphalioides plants. The frequent application of phosphite, therefore, may reduce the abundance of annual plants in this ecosystem. Received: 14 December 2000 / Revision accepted: 10 March 2001  相似文献

Mortality and turnover of some of the most characteristic woody plants of Galápagos have been studied during 25 to 32 years on the islands of Santa Cruz, Santa Fe and Pinta. Scalesia pedunculata, S. helleri subsp. helleri and S. baurii subsp. hopkinsii display comparable patterns of high mortality, rapid turnover and relatively short life expectancy, but occur in different habitats and islands in the archipelago. In contrast, Opuntia echios var. gigantea, O. echios var. barringtonensis, O. galapageia var. galapageia, and Bursera graveolens display low mortality (especially of adults), slow turnover and long life expectancies, and occur in similar habitats on different islands. Conditions that favour one taxon may at the same time negatively affect another taxon, and adults, juveniles and seedlings within the taxa may react differently to the same environmental factors. In the humid zone species S. pedunculata, diebacks may be triggered by strong El Niño events, and mass seedling events are associated with El Niños and a subsequent year of drought. In the other taxa investigated, which are arid zone species, the response to strong El Niño events is less pronounced and, in some respects, also different. The differences in mortality, turnover and life expectancy of the species of Scalesia on the one hand and the Opuntia taxa and Bursera on the other, suggest that both humid and arid vegetation types containing species of Scalesia should get higher conservation priority than arid vegetation types without species of Scalesia.  相似文献

Protoporphyrin IX is the last common intermediate of tetrapyrrole biosynthesis. The chelation of a Mg2+ ion by magnesium chelatase and of a ferrous ion by ferrochelatase directs protoporphyrin IX towards the formation of chlorophyll and heme, respectively. A full length cDNA clone encoding a ferrochelatase was identified from a Nicotiana tabacum cDNA library. The encoded protein consists of 497 amino acid residues with a molecular weight of 55.4 kDa. In vitro import of the protein into chloroplasts and its location in stroma and thylakoids confirm its close relationship to the previously described Arabidopsis thaliana plastid-located ferrochelatase (FeChII). A 1700-bp tobacco FeCh cDNA sequence was expressed in Nicotiana tabacum cv. Samsun NN under the control of the CaMV 35S promoter in antisense orientation allowing investigation into the consequences of selective reduction of the plastidic ferrochelatase activity for protoporphyrin IX channeling in chloroplasts and for interactions between plastidic and mitochondrial heme synthesis. Leaves of several transformants showed a reduced chlorophyll content and, during development, a light intensity-dependent formation of necrotic leaf lesions. In comparison with wild-type plants the total ferrochelatase activity was decreased in transgenic lines leading to an accumulation of photosensitizing protoporphyrin IX. Ferrochelatase activity was reduced only in plastids but not in mitochondria of transgenic plants. By means of the specifically diminished ferrochelatase activity consequences of the selective inhibition of protoheme formation for the intracellular supply of heme can be investigated in the future.  相似文献

根据15年的调查结果,对浙江省水生维管束植物的区系与地理分布特点进行了研究。已知浙江有水生维管束植物150种,隶属于78属,42科。将78属归纳为11个分布区类型,以世界分布类型所占比例最高,热带分布属总数多于温带分布属,泛热带分布和北温带分布比例非常高。将150种植物归纳为13个分布区类型,温带分布远多于热带分布,以东亚分布类型最为突出,体现了中国-日本植物区系的明显特征,探讨了浙江水生植物的水平地带性和垂直地带性分布规律,其分布不仅与气候带有关,更与水生的生境的分布格局有关。最后,报浙江水生植物濒危种的受威胁状况。  相似文献

田峪河流域是陕西秦岭植物园的核心区,有种子植物128科,605属,1231种(不含种下等级)。区系分析表明,该流域仍以温带成分为主,占总属的70.96%,热带成分占21.13%,地中海与中亚成分占4.41%,中国特有属占3.5%,稀有濒危植物、单种属和少种属、中国特有属及木本植物均占较大比重,说明了历史起源的古老性,地理成分复杂,联系广泛,分布交错,具有明显的温带特征。  相似文献

Spartina alterniflora Lois. is a dominant species growing in intermediate and saline marshes of the US Gulf coast and Atlantic coastal marshes. S. alterniflora plants were subjected to a range of soil redox potential (Eh) conditions representing a well aerated to reduced conditions in a rhizotron system under controlled environmental conditions. The low soil Eh resulted in inhibition of root elongation shortly after treatment initiation. Root elongation was reduced as soil Eh approached values below ca. +350 mV. Substantial decrease in root elongation was noted when soil Eh fell below +200 mV. Generally, net photosynthetic rate (PN) decreased as soil Eh was reduced, with substantial reductions in PN found when Eh approached negative values. Average PN was reduced to 87, 64, and 44% of control under +340, +245, and -180 mV treatments, respectively. The reductions in root elongation and PN in response to low soil Eh indicated the adverse effects of low soil Eh on plant functioning and the need for periods of soil aeration that allow plants to resume normal functioning. Thus periods of drainage allowing soil aeration during the growing season appear to be critical to S. alterniflora by providing favorable conditions for root growth and gas exchange with important implications for plant carbon fixation. The present study optimized the extraction characterization and antioxidant activities of water-soluble compound polysaccharides (CPs) from hawthorn, lotus leaf, Fagopyrum tataricum, semen cassiae, Lycium barbarum, and Poria cocos Chinese herbal medicines that have mass ratios of 4 : 2 : 2 : 1.5 : 1 : 1. The CPs yield equation was predicted using quantitative theory, to which a maximum CPs yield of 7.18±0.24 % under the following optimal extraction conditions: a water-to-raw material ratio of 30 mL/g, an extraction temperature of 65 °C, an extraction time of 45 min, and extraction mode ultrasonic-assistant extraction. CPs were consisted of Ara, Gal, Glc, Xyl, Man, GalA and GlcA in a molar ratio of 3.1 : 2.6 : 50.6 : 1.7 : 20.4 : 17.2 : 4.2. The HPGPC profiles and FT-IR spectra implied that CPs were heterogeneous acidic polysaccharides and possessed the β-d -pyranose configuration. Congo red test, CD spectrum and SEM revealed that CPs with three helix conformation showed a flocculent, granulous or sheet-like appearance. Furthermore, the relationships between antioxidant activity and concentration of CPs displayed significant positive correlation, and the scavenging abilities for DPPH, hydroxyl radical, ABTS, superoxide-anion radical and reducing power of CPs were 93.56±2.51 %, 84.03±1.69 %, 83.29±1.93 %, 37.49±1.93 % and 0.467±0.006 at a concentration of 4.0 mg/mL. Therefore, CPs could be applied as a potential natural antioxidant in pharmaceutical or functional food fields.  相似文献
