Merozoite invasion of erythrocytes is a crucial step for the asexual cycle of Plasmodium falciparum. Multiple invasion pathways, which involve different ligand-receptor interactions, have been identified in P. falciparum by examining the entry of purified parasite into erythrocytes with different surface receptors, either mutant or under different enzyme treatments. The most critical step for a successful invasion assay is the isolation of erythrocytes infected with viable schizonts. Here, we applied a magnetic column to purify the schizonts for the erythrocyte invasion assay. Comparing to Percoll-sorbitol purification method, this modified approach showed great improvement on reproducibility and reliability of invasion assay, particularly for short-term, culture-adapted parasite isolates. The magnetic purification method is an excellent alternative for parasite isolates that do not tolerate or with unknown sensitivity to Percoll-sorbitol exposure.  相似文献

Aim Historical information about source populations of invasive species is often limited; therefore, genetic analyses are used. We compared inference about source populations from historical and genetic data for the oyster‐associated clam, Gemma gemma that invaded California from the USA Atlantic coast. Location Mid‐Atlantic (North Carolina, Maryland), Northeastern (New Jersey, New York, Massachusetts) and the California coasts (Elkhorn Slough, San Francisco Bay, Bolinas Lagoon, Tomales Bay, Bodega Harbor). Methods The documented history of transplantation of Eastern oysters to California was reviewed. Cytochrome c oxidase subunit I (COI) sequences from recent and archived clams were examined in a haplotype network. We used AMOVA to detect geographic genetic structure and a permutation test for significant reductions in diversity. Results Chesapeake Bay oysters were transplanted to New York prior to shipment to San Francisco Bay and from there to peripheral bays. Gemma in the Northeastern and Mid‐Atlantic regions were genetically differentiated. In California, populations in Bodega Harbor and Tomales Bay were genetically similar to those in the Mid‐Atlantic area while clams in San Francisco Bay, Elkhorn Slough and Bolinas Lagoon resembled populations in the Northeastern region. In California, genetic variation was not highest in San Francisco Bay despite greater magnitude of oyster plantings. Haplotypes varied over time in native and introduced populations. Main Conclusions Historical records and inferences from genetics agree that both Northeastern and Mid‐Atlantic regions were sources for Gemma in California. Only complex genetic hypotheses reconcile the strong segregation of haplotypes in California to the historical evidence of mixing in their proximate source (New York). These hypotheses include sorting of mixtures of haplotypes or selection in non‐native areas. Haplotype turnover in San Francisco and Massachusetts samples over time suggests that the sorting hypothesis is plausible. We suggest, however, that Gemma was introduced independently and recently to Tomales Bay and Bodega Harbor.  相似文献

In the mature chorion, one of the membranes that exist during pregnancy between the developing fetus and mother, human placental cells form highly specialized tissues composed of mesenchyme and floating or anchoring villi. Using fluorescence in situ hybridization, we found that human invasive cytotrophoblasts isolated from anchoring villi or the uterine wall had gained individual chromosomes; however, chromosome losses were detected infrequently. With chromosomes gained in what appeared to be a chromosome-specific manner, more than half of the invasive cytotrophoblasts in normal pregnancies were found to be hyperdiploid. Interestingly, the rates of hyperdiploid cells depended not only on gestational age, but were strongly associated with the extraembryonic compartment at the fetal-maternal interface from which they were isolated. Since hyperdiploid cells showed drastically reduced DNA replication as measured by bromodeoxyuridine incorporation, we conclude that aneuploidy is a part of the normal process of placentation potentially limiting the proliferative capabilities of invasive cytotrophoblasts. Thus, under the special circumstances of human reproduction, somatic genomic variations may exert a beneficial, anti-neoplastic effect on the organism.  相似文献

Facilitation, both by inter‐ and intra‐specific neighbours, is known to be an important process in structuring plant communities. However, only a small number of experiments have been reported on facilitation in plant invasions, especially between invasive con‐specific individuals. Here, we focus on how con‐specific neighbours of the invasive alien plant alligator weed affect the tolerance of alligator weed to herbivory by the introduced biological control agent, Agasicles hygrophila. We conducted greenhouse and garden experiments in which invasive plant density and herbivory intensity (artificial clipping and real herbivory) were manipulated. In the greenhouse experiment, artificial clipping significantly reduced plant biomass when plants were grown individually, but when con‐specific neighbours were present in the same pot, biomass was not significantly different from control plants. Similarly, when compared to control plants, plants that were subjected to herbivory by A. hygrophila produced more biomass when grown with two con‐specific neighbours than when grown alone. Real herbivory also resulted in an increased number of vegetative buds, and again when two con‐specific neighbours were present this effect was increased (a 55.3% increase in buds when there was no neighbour, but a 111.6% increase in buds when two con‐specific neighbours were present). In the garden experiment, in which plants were grown at high density (6 plants per pot), alligator weed fully recovered from defoliation caused by insects at levels from 20–30% to 100%. Our results indicate that the con‐specific association may increase the compensatory ability to cope with intense damage in this invasive plant.  相似文献

Our previous studies revealed that leukocyte infiltration could trigger breast and prostate tumor invasion through physical disruption of tumor capsules. Our current study, involving multiple types of human tumors, further suggests that leukocyte infiltration also triggers metastasis through the following pathways : 1) the physical movement into the epithelium disrupts inter-cellular junctions and surface adhesion molecules, which cause the disassociation of tumor cells from tumor cores, 2) some of these tumor cells subsequently form tight junctions with the plasma membranes of leukocytes creating tumor cell-leukocyte chimeras (TLCs), and 3) the leukocytes of TLCs impart migratory capacity to associated tumor cell partners. Our findings suggest a novel pathway for tumor cell dissemination from primary sites and journey to new sites.  相似文献

  • 1 The host‐odour preferences of the rice leaf bug Trigonotylus caelestialium between the rice plant Oryza sativa L. and four species of graminaceous weeds, Poa annua, Alopecurus aequalis, Digitaria ciliaris and Eleusine indica, were investigated with an olfactometer aiming to clarify the influence of these odours on invasion of the bug to paddy fields at the flowering stage of rice.
  • 2 Both female and male adults significantly preferred the graminaceous weed A. aequalis in the flowering stage to rice in the fifth‐leaf stage. The bugs also significantly preferred flowering P. annua and A. aequalis to rice in the panicle‐formation stage. However, the bugs showed no preferences between rice in the flowering and grain‐filling stages and the flowering graminaceous weeds P. annua, D. ciliaris and E. indica.
  • 3 The preference of the rice leaf bug for the flowering graminaceous weeds before rice flowering coincides with the fact that these bugs mainly live on these weeds before rice flowering. It is considered that the bug's similar preference for flowering rice panicles as the flowering graminaceous weeds causes the intense invasion of the bug into paddy fields at this rice developmental stage.

Questions: Do spatial and temporal patterns of encroachment of Pinus contorta and Abies grandis in a montane meadow suggest strong biotic controls on the invasion process? Location: Forest–meadow mosaic, 1350 m a.s.l., Cascade Range, Oregon, US. Methods: We combined spatial point pattern analysis, population age structures, and a time‐series of stem maps to quantify spatial and temporal patterns of conifer invasion over a 200‐yr period in three plots totaling 4 ha. Results: Trees established during two broad, but distinct periods (late 1800s, then at much greater density in the mid‐1900s). Recent invasion was not correlated with climatic variation. Abies grandis dominated both periods; P. contorta established at lower density, peaking before A. grandis. Spatially, older (≥90 yr) P. contorta were randomly distributed, but older A. grandis were strongly clumped (0.2‐20 m). Younger (<90 yr) stems were positively associated at small distances (both within and between species), but were spatially displaced from older A. grandis, suggesting a temporal shift from facilitation to competition. Establishment during the 1800s resulted in widely scattered P. contorta and clumps of A. grandis that placed most areas of meadow close to seed sources permitting more rapid invasion during the mid‐1900s. Rapid conversion to forest occurred via colonization of larger meadow openings – first by shade‐intolerant P. contorta, then by shade‐tolerant A. grandis– and by direct infilling of smaller openings by A. grandis. Conclusions: In combination, spatial and temporal patterns of establishment suggest an invasion process shaped by biotic interactions, with facilitation promoting expansion of trees into meadows and competition influencing subsequent forest development. Once invasion is initiated, tree species with different life histories and functional traits can interact synergistically to promote rapid conversion of meadow to forest under a broad range of climatic conditions.  相似文献

克氏原螯虾(Procambarus clarkii)原产于美国南部和墨西哥北部,是一个著名入侵物种,作为一种水产经济资源物种在世界各地扩散。克氏原螯虾抗逆性强,所具有的广泛生境适应性、生长迅速、高生殖率等特点使它们迅速建立野生种群。近10余年的研究认为,克氏原螯虾通过捕食和资源竞争等机制严重威胁引入地的水生植物、无脊椎动物、两栖类等的生存,显著降低引入地的生物多样性。当前,由于克氏原螯虾的经济价值高,它会借助于人力的作用而继续扩散。为认清和减少克氏原螯虾对引入地的生态影响,应加强以下方面的研究:1)在中国开展克氏原螯虾的生态危害的调查和研究;2)克氏原螯虾种群调节和控制对策研究;3)被入侵地的生态恢复工作。  相似文献
