Murine γδ T cells develop as the first T-cell lineage within the fetal thymus and disproportionately localize in mucosal tissues such as lung, skin, uterus, and intestine of adult mice. These unique developmental features and distribution patterns of γδ T cells enable rapid functioning against various insults from pathogens. γδ T cells are also able to respond to local inflammation and consequently regulate the pathogenesis of autoimmune disorders and development of tumors in mice and humans. Hence, it is clinically important to understand the mechanisms that regulate γδ T cell functions. Recent evidence has shown that generations of effector γδ T cell subsets producing IFN-γ, IL-4, and IL-17 are programmed in the murine thymus before their migration to peripheral tissues. This review outlines our current understanding of the development and function of γδ T cells as they influence both innate and acquired immunity.  相似文献

以实验室建立的$180小鼠肿瘤模型为研究对象,采用腹腔注射给药,观察葡萄球菌肠毒素A(SEA)在体内抑制肿瘤的效果。实验表明,SEA抑肿瘤率为40.18%,显示对肿瘤有一定的抑制作用;能显著刺激脾脏细胞增殖,使脾指数升高至11.3mg/g;使血清和脾组织中IL-2水平分别升至69.77pg/mL和682.43pg/mL;且能诱导肿瘤组织中产生大量的CD4^+T细胞和CD8^+T细胞。结果显示,SEA在机体内对免疫功能有正向调节作用,从而在一定程度上抑制了肿瘤细胞的生长。  相似文献

Cytotoxic lymphocytes (CLs) are responsible for the clearance of virally infected or neoplastic cells. CLs possess specialised lysosome-related organelles called granules which contain the granzyme family of serine proteases and perforin. Granzymes may induce apoptosis in the target cell when delivered by the pore forming protein, perforin. Here we follow the perforin-granzyme pathway from synthesis and storage in the granule, to exocytosis and finally delivery into the target cell. This review focuses on the controversial subject of perforin-mediated translocation of granzymes into the target cell cytoplasm. It remains unclear whether this occurs at the cell surface with granzymes moving through a perforin pore in the plasma membrane, or if it involves internalisation of perforin and granzymes and subsequent release from an endocytic compartment. The latter mechanism would represent an example of cross talk between the endo-lysosomal pathways of individual cells. The syntaxin 11 (STX11) gene is mutated in a proportion of patients with familial haemophagocytic lymphohistiocytosis (FHL) and exocytosis of cytotoxic granules is impaired in STX11-deficient NK cells. However, the subcellular localization, regulation of expression and molecular function of STX11 in NK cells and other cytotoxic lymphocytes remain unknown. Here we demonstrate that STX11 expression is strictly controlled by several mechanisms in a cell-type-specific manner and that the enzymatic activity of the proteasome is required for STX11 expression in NK cells. In resting NKL cells, STX11 was localized in the cation-dependent mannose-6-phosphate receptor (CD-M6PR)-containing compartment, which was clearly distinct from cytotoxic granules or Rab27a-expressing vesicles. These subcellular structures appeared to fuse at the contact area with NK-sensitive target cells as demonstrated by partial colocalization of STX11 with perforin and Rab27a. Although STX11-deficent allo-specific cytotoxic T-lymphocytes efficiently lysed target cells and released cytotoxic granules, they exhibited a significantly lower extent of spontaneous association of perforin with Rab27a as compared with STX11-expressing T cells. Thus, our results suggest that STX11 promotes the fusion of Rab27a-expressing vesicles with cytotoxic granules and reveal an additional level of complexity in the spatial/temporal segregation of subcellular structures participating in the process of granule-mediated cytotoxicity.  相似文献

目的:研究重组人粒细胞集落刺激因子(rhG-CSF)动员对供者CD4+T细胞表面分子淋巴细胞功能相关抗原-1(LFA-1)、细胞间黏附分子-1(ICAM-1)、L-选择素(LAM-1)和人整合素-4(VLA-4)的表达及其介导的CD4+T细胞功能的影响,探讨外周血干细胞移植过程中CD4+T细胞免疫耐受机制。方法:使用三色荧光标记检测动员前及动员后第5天供者外周血LFA-1、ICAM-1、LAM-1和VLA-4的表达率,ELISA方法检测动员前后CD4+T细胞分泌IFN-γ和IL-4能力,免疫磁性分选法分离纯化CD4+T细胞,检测动员前后CD4+T细胞对基质细胞衍生因子-1α(SDF-1α)的迁移能力和对ICAM-1的黏附能力。结果:动员前后CD4+T细胞LFA-1(CD11a)和VLA-4(CD49d)表达率差异无统计学意义(P〉0.01),动员前后CD4+T细胞LAM-1(CD62L)和ICAM-1(CD54)的表达率差异均有统计学意义,动员前显著高于动员后(P〈0.01);动员前后CD4+T淋巴细胞向SDF-1α的迁移率差异无统计学意义(P〉0.01);动员后CD4+T细胞对ICAM-1的黏附率降低(P〈0.01);动员后IL-4和IFN-γ两个细胞因子在外周血血清的浓度均降低(P〈0.01)。结论:rhG-CSF动员不影响CD4+T细胞LFA-1和VLA-4表达及CD4+T细胞迁移,但影响CD4+T细胞ICAM-1和LAM-1表达以及CD4+T细胞通过LFA-1对ICAM-1的黏附能力影响,并可能影响CD4+T细胞分泌细胞因子IL-4及IFN-γ的功能。  相似文献

研究玉竹生物活性成分C对小鼠细胞免疫功能的影响,并探讨其作用机制.采用MTT比色法和CFSE检测小鼠淋巴细胞转化情况,流式细胞术检测对αβT和γδT细胞增殖及CD4+ αβT细胞/CD8+αβT细胞比值的影响,ELISA法检测脾细胞培养上清中Th1细胞因子IFN-γ和Th2细胞因子IL-4的分泌水平.结果显示,玉竹生物活性成分C在一定浓度范围内可促进小鼠脾αβT细胞增殖,其中以1 000μg/mL的浓度作用最强.1 000 μg/mL浓度的玉竹生物活性成分C具有降低CD4+ αβT细胞与CD8+ αβT细胞比值,提高CD8+αβT细胞教量的作用.1 000 μg/mL浓度的玉竹生物活性成分C能有效刺激小鼠脾淋巴细胞释放细胞因子IFN-γ,而对IL-4的产生没有明显的影响.本研究为开发玉竹生物活性成分C提供实验依据.  相似文献

目的:检测急性甲、乙型病毒性肝炎患者外周血T淋巴细胞亚群变化,探讨其对疗效和预后意义。方法:采用APAAP桥联酶标法检测115例急性甲、乙型病毒型肝炎患者外周血CD3+、CD4+、CD8+T细胞亚群比例,计算CD4+/CD8+值。并检测了34例患者治疗前后T淋巴细胞亚群变化。结果:115例急性甲、乙型病毒性肝炎患者外周血CD3+、CD4+T细胞比例及CD4+/CD8+值均低于正常对照组(P<0.05),而CD8+T细胞比例均高于正常对照组(P<0.01)。6例无明显疗效者,各亚群比例在治疗前后无显著差异(P>0.05)。28例有明显疗效者,治疗后各亚群比例恢复正常水平,与治疗前相比差别具有统计学意义(P<0.05)。结论:急性甲、乙型病毒性肝炎患者外周血CD4+/CD8+T细胞比值可在一定程度上反映疗效及预后。  相似文献

Survival and recurrence rates in breast cancer are variable for common diagnoses, and therefore the biological underpinnings of the disease that determine those outcomes are yet to be fully understood. As a result, translational medicine is one of the fastest growing arenas of study in tumor biology. With advancements in genetic and imaging techniques, archived biopsies can be examined for purposes other than diagnosis. There is a great deal of evidence that points to the stroma as the major regulator of tumor progression following the initial stages of tumor formation, and the stroma may also contribute to risk factors determining tumor formation. Therefore, aspects of stromal biology are well-suited to be a focus for studies of patient outcome, where statistical differences in survival among patients provide evidence as to whether that stromal component is a signpost for tumor progression. In this review we summarize the latest research done where breast cancer patient survival was correlated with aspects of stromal biology, which have been put into four categories: reorganization of the extracellular matrix (ECM) to promote invasion, changes in the expression of stromal cell types, changes in stromal gene expression, and changes in cell biology signaling cascades to and from the stroma.  相似文献

During the immune response, the generation of memory B lymphocytes in germinal centers involves affinity maturation of the cells’ antigen receptors, based on somatic hypermutation of receptor genes and antigen-driven selection of the resulting mutants. Affinity maturation is vital for immune protection, and is the basis of humoral immune learning and memory. Lineage trees of somatically hypermutated immunoglobulin genes often serve to qualitatively illustrate claims concerning the dynamics of affinity maturation in germinal centers. Here, we derive the quantitative relationships between parameters characterizing affinity maturation dynamics (proliferation, differentiation and mutation rates, initial affinity of the Ig to the antigen, and selection thresholds) and the mathematical properties of lineage trees, using a computer simulation which combines mathematical models for all mature B cell populations, stochastic models of hypermutation and selection, lineage tree generation and measurement of graphical tree characteristics. We identified seven key lineage tree properties, and found correlations of these with initial clone affinity and with the selection threshold. These two parameters were found to be the main factors affecting lineage tree shapes in both primary and secondary response trees. The results also confirm that recycling from centrocytes back to centroblasts is highly likely.  相似文献

B 细胞成熟抗原 (BCMA)是 B淋巴细胞刺激因子(BLyS)的受体之一.它的胞外区与人IgG1 Fc的融合蛋白eBCMA-Fc,又称为诱饵受体,具有拮抗BLyS的活性.为了设计新的拮抗肽,基于BCMA和Fc的晶体结构,通过计算机图形学技术、分子模拟方法,建立了eBCMA-Fc融合蛋白的三维理论结构.利用均方根位移(root mean square distance, RMSD)对eBCMA-Fc融合蛋白与单体eBCMA、Fc构象差异进行分析.融合蛋白eBCMA-Fc中的eBCMA段与单体eBCMA的主链碳原子间RMSD值为0.036 nm,Fc段与单体Fc的主链碳原子间RMSD值为0.064 nm.结果表明,对比单体,融合蛋白eBCMA-Fc并未因eBCMA与Fc直接连接而发生构象的变化.分子对接方法显示,融合蛋白eBCMA-Fc中的BCMA与BLyS作用,而Fc扮演着稳定BCMA构象的支架作用.为进一步验证上述理论分析,构建eBCMA-Fc融合基因,并将载有eBCMA-Fc融合基因的原核表达质粒转化BL21 (DE3)菌、在细菌中表达.目的蛋白经蛋白A亲和柱纯化大约为36 kD,与理论预测值34 kD相近.免疫印迹表明抗人IgG抗体能够识别eBCMA-Fc融合蛋白.ELISA证实,eBCMA-Fc融合蛋白能够结合BLyS.随着eBCMA-Fc融合蛋白增加,结合BLyS的融合蛋白也相应增加.而对照人IgG,即使在高浓度条件下,也不结合BLyS.此外,eBCMA-Fc 融合蛋白能够抑制BLyS对B细胞肿瘤Daudi细胞的作用.这些研究为下一步设计和筛选BLyS拮抗肽提供了实验基础.  相似文献
