Objective: To identify the relative roles of climatic, edaphic and management factors in controlling the weighted mean traits of vegetation. Location: Eleven sites in Europe and one in Israel undergoing transitions in land use. Material and Methods: Standardised methods were used to collect information on species traits and attributes from plots covering a range of land uses at each site. This was combined with abundance data to create a plot × trait matrix. Variance partitioning was used to identify the relative roles of climate, soil and management on the weighted and unweighted mean traits of the vegetation in the full data set, and the data set divided into vegetative traits (including life‐form, clonality, defence and a range of leaf traits) and traits linked to regeneration via seeds (including seed mass, dispersal and pollination mechanism). Results: Variance partitioning of the full data set showed that climate (18.7%), explained more variance in the weighted mean traits of the vegetation than climate and soil together (9.2), soil (6.9) and management (6.1). There was a similar distribution of variance explained for both vegetative and regeneration via seed traits, although more variance was explained for the latter. This restricted set of climatic, edaphic and management variables could explain 45‐50% of the variance in the weighted mean traits of the vegetation between plots. There were only small differences between analyses of the weighted and unweighted data. Conclusions: Despite large variations in climate and soils between sites, there was still a separate and recognisable impact of management on the mean weighted traits of the vegetation. There was also a degree of shared variation between the three groups of factors, indicating that the response of plant traits to one group of factors may not be predictable because they may be modulated by their response to other groups.  相似文献

The current article examined the feasibility of inducing improved delivery and degradation of phenanthrene in a solid–liquid partitioning bioreactor system at bench scale by means of ultrasonic energy input. Initial degradation rates of phenanthrene by a microbial consortium, delivered from Desmopan, were improved 2.7‐fold in the presence of sonication relative to unsonicated controls. Results demonstrated that an operating window involving on/off sonication cycling improved substrate delivery and rational selection of ultrasound cycling profiles could lead to even further enhancements. Additionally, all results were obtained in a conventional bioreactor with commercial ultrasonic equipment and a commercially available polymer. Subsequent DGGE analysis demonstrated that the sonication cycles selected maintained consortium compositions, relative to control cases, and suggest that exposure would not reduce degradative capabilities under the periods of irradiation examined. Finally, consortium members were identified as belonging to the Pandoraea, Sphingobium, and Pseudoxanthomonas genera. Comparison of genetic sequences in the Ribosomal Database Project revealed that some of the bacterial members, identified at the strain level, had been previously observed in PAH degradations, while others have been reported only in the degradation of other aromatics, such as pesticides. Liquid-ordered (Lo) and liquid-disordered (Ld) phase coexistence has been suggested to partition the plasma membrane of biological cells into lateral compartments, allowing for enrichment or depletion of functionally relevant molecules. This dynamic partitioning might be involved in fine-tuning cellular signaling fidelity through coupling to the plasma membrane protein and lipid composition. In earlier work, giant plasma membrane vesicles, obtained by chemically induced blebbing from cultured cells, were observed to reversibly phase segregate at temperatures significantly below 37 °C. In this contribution, we compare the temperature dependence of fluid phase segregation in HeLa and rat basophilic leukemia (RBL) cells. We find an essentially monotonic temperature dependence of the number of phase-separated vesicles in both cell types. We also observe a strikingly broad distribution of phase transition temperatures in both cell types. The binding of peripheral proteins, such as cholera toxin subunit B (CTB), as well as Annexin V, is observed to modulate phase transition temperatures, indicating that peripheral protein binding may be a regulator for lateral heterogeneity in vivo. The partitioning of numerous signal protein anchors and full length proteins is investigated. We find Lo phase partitioning for several proteins assumed in the literature to be membrane raft associated, but observe deviations from this expectation for other proteins, including caveolin-1.  相似文献

The Atlantic Forest deserves special attention due to its high level of species endemism and degree of threat. As in other tropical biomes, there is little information about the ecology of the organisms that occur there. The objectives of this study were to verify how fruit-feeding butterflies are distributed through time, and the relation with meteorological conditions. Species richness and Shannon index were partitioned additively at the monthly level, and β diversity, used as a hierarchical measure of temporal species turnover, was calculated among months, trimesters, and semesters. Circular analysis was used to verify how butterflies are distributed along seasons and its relation with meteorological conditions. We sampled 6488 individuals of 73 species. Temporal diversity of butterflies was more grouped than expected by chance among the months of each trimester. Circular analyses revealed that diversity is concentrated in hot months (September–March), with the subfamily Brassolinae strongly concentrated in February–March. Average temperature was correlated with total abundance of butterflies, abundance of Biblidinae, Brassolinae and Morphinae, and richness of Satyrinae. The present results show that 3 mo of sampling between September and March is enough to produce a nonbiased sample of the local assemblage of butterflies, containing at least 70 percent of the richness and 25 percent of abundance. The influence of temperature on sampling is probably related to butterfly physiology. Moreover, temperature affects resource availability for larvae and adults, which is higher in hot months. The difference in seasonality patterns among subfamilies is probably a consequence of different evolutionary pressures through time.  相似文献

In biotechnology, endotoxin (LPS) removal from recombinant proteins is a critical and challenging step in the preparation of injectable therapeutics, as endotoxin is a natural component of bacterial expression systems widely used to manufacture therapeutic proteins. The viability of large‐scale industrial production of recombinant biomolecules of pharmaceutical interest significantly depends on the separation and purification techniques used. The aim of this work was to evaluate the use of aqueous two‐phase micellar system (ATPMS) for endotoxin removal from preparations containing recombinant proteins of pharmaceutical interest, such as green fluorescent protein (GFPuv). Partition assays were carried out initially using pure LPS, and afterwards in the presence of E. coli cell lysate. The ATPMS technology proved to be effective in GFPuv recovery, preferentially into the micelle‐poor phase (KGFPuv < 1.00), and LPS removal into the micelle‐rich phase (%REMLPS > 98.00%). Therefore, this system can be exploited as the first step for purification in biotechnology processes for removal of higher LPS concentrations. © 2010 American Institute of Chemical Engineers Biotechnol. A strategic approach for the design of two‐phase partitioning bioscrubbers (TPPBs) has been formulated using, as a basis, a re‐evaluation of extensive literature data available for the degradation of benzene by Achromobacter xylosoxidans Y234 in TPPBs with n‐hexadecane as the partitioning phase. Using a previously determined maintenance coefficient for benzene, we determined that an inlet benzene loading rate of 100 mg/h requires 5,928 mg cell mass at biological steady state and 243.0 mg O2/h. The total oxygen‐transfer rates (TOTRs) into the TPPB increased by 83.5% with 33.3% of organic phase compared with a single aqueous phase and were significantly influenced by gas flow rate, whereas agitation has a minor affect. The fraction of organic phase used was suggested to be the primary parameter with which the TOTR into the TPPB may be altered. Although the presence of an organic solvent in the TPPB remarkably increased the TOTR, the total benzene transfer rate into the TPPB remained largely insensitive due to the intrinsic low Henry's law constant (or relatively high solubility) of benzene in water. Finally, we have integrated the elements of this analysis into a set of heuristic criteria that can serve as a guideline for the design of TPPB systems for future volatile organic compound treatment applications. © 2010 American Institute of Chemical Engineers Biotechnol. Question: What is the relative importance of national‐, regional‐ and within‐beach‐scale influences on vegetation composition and floristic affinities of New Zealand gravel beaches? Location: Coastal New Zealand. Methods: We sampled vegetation composition at 61 gravel beaches, quantifying site factors and adjacent landscape characteristics. Site, climate and geographic relationships between gravel beaches and related ecosystems were inferred using GIS data layers. To simultaneously investigate influences at different spatial scales, we used ordination and variation partitioning to examine relationships between composition and environment, and hierarchical models to understand floristic affinities with related ecosystems. Results: At a national scale, compositional variation among beaches reflects mean annual temperature and spring vapour pressure deficit; within regions, proximity of native woody vegetation and coastal turfs are important; within‐beach variation is related to substrate stability and particle size distribution. The gravel beach flora is 50% exotic, reflecting the highly modified nearby landscapes; 30% of species are characteristic of coastal sands, 20% of braided riverbeds and 8% of coastal turfs. Affinities with coastal sand communities are unrelated to microsite sandiness or area of sand dunes within 50 km. Affinities with braided riverbeds are related to the bed area of those rivers draining within 200 km and proportion of gravel in the substrate. Affinities with coastal turfs are related to proximity to the nearest turf and the proportion of humus in the substrate. Conclusions: Examining multiple scales of influence in a landscape context is essential to understand composition of naturally discrete ecosystems that span wide geographic ranges and to underpin their conservation management.  相似文献

1. Lotic ecosystems can be studied on several spatial scales, and usually show high heterogeneity at all of them in terms of biological and environmental characteristics. Understanding and predicting the taxonomic composition of biological communities is challenging and compounded by the problem of scale. Additive diversity partitioning is a tool that can show the diversity that occurs at different scales. 2. We evaluated the spatial distribution of benthic macroinvertebrates in a tropical headwater catchment (S.E. Brazil) during the dry season and compared alpha and beta diversities at the scales of stream segments, reaches, riffles and microhabitats (substratum types: gravels, stones and leaf litter). We used family richness as our estimate of diversity. Sampling was hierarchical, and included three stream segments, two stream reaches per segment, three riffles per reach, three microhabitats per riffle and three Surber sample units per microhabitat. 3. Classification analysis of the 53 families found revealed groups formed in terms of stream segment and microhabitat, but not in terms of stream reaches and riffles. Separate partition analyses for each microhabitat showed that litter supported lower alpha diversity (28%) than did stones (36%) or gravel (42%). In all cases, alpha diversity at the microhabitat scale was lower than expected under a null model that assumed no aggregation of the fauna. 4. Beta diversity among patches of the microhabitats in riffles depended on substratum type. It was lower than expected in litter, similar in stone and higher in gravel. Beta diversities among riffles and among reaches were as expected under the null model. On the other hand, beta diversity observed was higher than expected at the scale of stream segments for all microhabitat types. 5. We conclude that efficient diversity inventories should concentrate sampling in different microhabitats and stream sites. In the present study, sampling restricted to stream segments and substratum types (i.e. excluding riffles and stream reaches) would produce around 75% of all observed families using 17% of the sampling effort employed. This finding indicates that intensive sampling (many riffles and reaches) in few stream segments does not result in efficient assessment of diversity in a region.  相似文献

The cftr knockout mouse model of cystic fibrosis (CF) shows intestinal obstruction; malabsorption and inflammation; and a fatty acid imbalance in intestinal mucosa. We performed a lipid mapping of colon sections from CF and control (WT) mice by cluster time of flight secondary-ion mass spectrometry (TOF-SIMS) imaging to localize lipid alterations. Data were processed either manually or by multivariate statistical methods. TOF-SIMS analysis showed a particular localization for cholesteryl sulfate at the epithelial border, C16:1 fatty acid in Lieberkühn glands, and C18:0 fatty acid in lamina propria and submucosa. Significant increases in vitamin E (vE) and C16:0 fatty acid in the epithelial border of CF colon were detected. Principal component analysis (PCA) and partitioning clustering allowed us to characterize different structural regions of colonic mucosa according to variations in C14:0, C16:0, C16:1, C18:0, C18:1, C18:2, C20:3, C20:4, and C22:6 fatty acids; phosphatidylethanolamine, phosphatidylcholine, and phosphatidylinositol glycerolipids; cholesterol; vitamin E; and cholesteryl sulfate. PCA on spectra from Lieberkühn glands led to separation of CF and WT individuals. This study shows for the first time the spatial distribution of lipids in colonic mucosa and suggests TOF-SIMS plus multivariate analyses as a powerful tool to investigate disease-related tissue spatial lipid signatures.  相似文献

目的:发展一种简便快速的制备水稻基因组DNA PCR模板的方法。方法:用枪头捣碎水稻叶片代替液氮研磨法提取水稻基因组DNA作PCR模板,在去污剂SDS和表面活性剂TrionX-100的存在下在沸水中煮沸10min,然后取上清扩增微管蛋白(TubA1)基因内含子。结果:发现在利用煮沸法提取水稻基因组DNA的过程中,用枪头捣碎叶片可代替液氮碾磨,加入0.1%表面活性剂TrionX-100煮沸叶片对制备模板有促进效果,得到了预期的PCR片断。结论:该方法快速、简便、经济,具有良好的重复性与特异性,便于自动化。  相似文献
