Wheat seedlings were grown hydroponically in absence and presence of 100 mM NaCl for 7 d. Cell membrane permeability to nonelectrolytes and water was determined by the plasmometric method for individual intact cells. NaCl increased membrane permeability to urea, methylurea and ethylurea and decreased permeability to water. Membrane lipid partiality was decreased by NaCl. The effects of NaCl on cell permeability parallel changes in the lipid composition of the plasma membranes induced by NaCl stress suggesting that nonelectrolyte permeability is a useful tool to probe alterations in the lipid matrix of the membrane.  相似文献

从5个异源八倍体小冰麦(Triticum -Agropyron)的叶片、幼穗及成熟胚诱导愈伤组织,建立体细胞无性系,获得大量再生植株。附加一个冰草染色体组的异源八倍体小冰麦杂种无性系中37.5% 表现变异,其中非整倍体植株变异较多,很多变异的再生植株形态与小麦近似,同时出现一定数量染色体重排、交换、易位、断裂、融合等变异。结果表明,通过杂种无性系变异进行染色体基因转化及遗传修饰是一条可行的途径。实验还观察了小冰麦愈伤组织分化过程中绿点的形成过程,首次提出两种类型绿点,即芽绿点和根绿点,并描述了两者的差异  相似文献

An in vivo model system to study the initiation of embryo development is presented. From the so-called Salmon system of wheat (alloplasmic lines with a 1BL-1RS chromosome translocation), three completely isogenic and homozygous lines were produced by selection for uniformity in about 20 selfing/backcross generations as well as between sublines of doubled haploids. The line (aestivum)-Salmon is male fertile and sexual. The lines (caudata)-Salmon and (kotschyi)-Salmon are male sterile and have a parthenogenetic capacity of about 90%. The expression of nuclear-cytoplasmic male sterility is different for the two parthenogenetic lines. The initiation of autonomous embryo development at defined developmental stages of the ovaries and the maximum degree of parthenogenesis are identical in both parthenogenetic lines as proved by the auxin test and progeny analyses. The protein patterns from ovary extracts of the three isogenic lines were identical for more than 200 spots of 2-D polyacrylamide gels, confirming their homogeneity. However, one protein (P 115.1) was found 3 days before and during anthesis only in ovaries of the parthenogenetic lines. It seems to be involved in the initiation of parthenogenesis.  相似文献

对小麦中期染色体中银染蛋白的大小、形状和分布频率进行图像分析,看到:染色体顺的银染蛋白以颗粒状的形式存在,其大小不同,分布不均匀,数量差异也较大;从形状来看,大的银粒为点状,小的银粒有的为点状,有的实际为短纤维状,结果表明:染色体骨架在小麦中是真实存在的,骨架蛋白以颗粒和纤维状的形式分布于整个染色体中。  相似文献

Abstract Polysomes and ribosomes recovered from a number of plant species were tested for stability when incubated at 25°C in salt solutions in the absence of ATP and initiation factors. Stability was assessed by sucrose density gradient analysis. The stability was inversely proportional to salt concentrations above 125 mol m−3 KCl. Polysomes were less stable in the presence of Na+ than K+ salts, and were much less stable in Cl than in acetate salts. Polysomes from Triticum aestivum. Hordeum vulgare, Capsicum annuum, Helianthus annuus. Pisum sativum, Atriplex nummularia, Beta vulgaris, Cladophora sp., Enteromorpha sp. and Corallina cuvieri were similarly sensitive to KCl. Polysomes from Ulva lactuca were more sensitive than the other species. Cytoplasmic and plastid polysomes from T. aestivum were similarly unstable in 500 mol m−3 KCl. Unprogrammed ribosomal subunit couples from T. aestivum, B. vulgaris and U. lactuca showed Mg2+-dependent conformational instability and dissociation in KCl. Slight differences in ribosomal stability were observed between species, but these were unrelated to the salt tolerances of the plants. The ‘compatible’ organic solutes, glycinebetaine and proline, failed to reduce ion-induced instability. Ribosome yield and polysome profiles were similar in leaves of B. vulgaris containing significantly different levels of both Na+ and Cl after growth in media containing 50 or 200 mol m−3 NaCl. The results are consistent with the hypothesis that plants maintain a cytoplasmic solute environment that is compatible with ribosomal stability.  相似文献

In vitro DNA:DNA hybridizations and hydroxyapatite thermal-elution chromatography were employed to identify the diploid wheat species ancestral to the B genome of Triticum turgidum. 3H-T. turgidum DNA was hybridized to the unlabeled DNAs of T. urartu, T. speltoides, T. sharonensis, T. bicorne, T. longissimum, and T. searsii. 3H-Labeled DNAs of T. monococcum and a synthetic tetraploid AADD were hybridized with unlabeled DNAs of T. urartu and T. searsii to determine the relationship of the A genome of polyploid wheat and T. urartu. The heteroduplex thermal stabilities indicated that T. searsii was most closely related to the B genome of T. turgidum (AB) and that the genome of T. urartu and the A genome have a great deal of base-sequence homology. Thus, it appears that T. searsii is the B-genome donor to polyploid wheat or a major chromosome donor if the B genome is polyphyletic in origin.Published with the approval of the Director of The West Virginia Agricultural Experiment Station as Scientific Paper No. 1837.  相似文献

The dynamics of stomatal resistance and osmotic adjustment in response to plant water deficits and stage of physiological development was studied in the leaves of spring wheat ( Triticum aestivum L., GWO 1809). Plants were germinated and grown in pots in a growth chamber at the Duke University Phytotron to four physiological stages of development (4th leaf, 7th leaf, anthesis, and soft dough), during which time stomatal resistance, total water potential and osmotic potential were measured on the last fully developed leaf of water stressed and non-stressed plants. Pressure potential was obtained by difference. Stomatal closure of the abaxial and adaxial surfaces were independent of each other, each having a different critical total water potential. The total water potential required to close the stomata on the last fully developed leaf were different at different stages of physiological development, decreasing as the plants grew older. The development of osmoregulation in wheat allows the closure of stomata during the vegetative stage at a high total water potential, but insures that stomata remain open from anthesis through the ear filling period to a lower total water potential.  相似文献

Summary Phytohormones like IAA and kinetin inhibit chlorophyll loss during aging of wheat chloroplasts duringin vivo andin vitro. GA, on the other hand, stimulates the pigment degradation during aging of attached leaves in contrast to its senescence inhibiting action in detached leaves and isolated chloroplasts. A shift in optimum concentration of hormone in inhibiting chlorophyll degradation suggests a differential pool size of endogenous hormone regulating aging of chloroplastsin vivo andin vitro. The retardation of chlorophyll loss by kinetin, IAA and GA during aging of chloroplastsin vitro would indicate that the effect of hormones in preventing yellowing of senescing leaves may be mediated through their direct action on chloroplasts.  相似文献

Summary The claim that osmium-containing deposits which lack lead are frequently and incorrectly interpreted as enzymatic reaction products in lead precipitation techniques for ATPase localization in plants is without foundation. Proper controls clearly demonstrate the enzymatic origin of membrane-located deposits and the presence of lead is confirmed by analytical electron microscopy.  相似文献
