Phosphate is an essential component of all cells that must be taken up from the environment. Prokaryotes commonly secrete alkaline phosphatases (APs) to recruit phosphate from organic compounds by hydrolysis. In this study, the AP from Halobacterium salinarum, an archaeon that lives in a saturated salt environment, has been functionally and structurally characterized. The core fold and the active-site architecture of the H. salinarum enzyme are similar to other AP structures. These generally form dimers composed of dominant β-sheet structures sandwiched by α-helices and have well-accessible active sites. The surface of the enzyme is predicted to be highly negatively charged, like other proteins of extreme halophiles. In addition to the conserved core, most APs contain a crown domain that strongly varies within species. In the H. salinarum AP, the crown domain is made of an acyl-carrier-protein-like fold. Different from other APs, it is not involved in dimer formation. We compare the archaeal AP with its bacterial and eukaryotic counterparts, and we focus on the role of crown domains in enhancing protein stability, regulating enzyme function, and guiding phosphoesters into the active-site funnel.  相似文献

In eukaryotic cells, phospholipids are synthesized exclusively in the defined organelles specific for each phospholipid species. To explain the reason for this compartmental specificity in the case of phosphatidylcholine (PC) synthesis, we constructed and characterized a Saccharomyces cerevisiae strain that lacked endogenous phosphatidylethanolamine (PE) methyltransferases but had a recombinant PE methyltransferase from Acetobacter aceti, which was fused with a mitochondrial targeting signal from yeast Pet100p and a 3 × HA epitope tag. This fusion protein, which we named as mitopmt, was determined to be localized to the mitochondria by fluorescence microscopy and subcellular fractionation. The expression of mitopmt suppressed the choline auxotrophy of a double deletion mutant of PEM1 and PEM2 (pem1Δpem2Δ) and enabled it to synthesize PC in the absence of choline. This growth suppression was observed even if the Kennedy pathway was inactivated by the repression of PCT1 encoding CTP:phosphocholine cytidylyltransferase, suggesting that PC synthesized in the mitochondria is distributed to other organelles without going through the salvage pathway. The pem1Δpem2Δ strain deleted for PSD1 encoding the mitochondrial phosphatidylserine decarboxylase was able to grow because of the expression of mitopmt in the presence of ethanolamine, implying that PE from other organelles, probably from the ER, was converted to PC by mitopmt. These results suggest that PC could move out of the mitochondria, and raise the possibility that its movement is not under strict directional limitations.  相似文献

Age, sex, and gonadal hormones have profound effects on ischemic stroke outcomes, although how these factors impact basic stroke pathophysiology remains unclear. There is a plethora of inconsistent data reported throughout the literature, primarily due to differences in the species examined, the timing and methods used to evaluate injury, the models used, and confusion regarding differences in stroke incidence as seen in clinical populations vs. effects on acute neuroprotection or neurorepair in experimental stroke models. Sex and gonadal hormone exposure have considerable independent impact on stroke outcome, but these factors also interact with each other, and the contribution of each differs throughout the lifespan. The contribution of sex and hormones to experimental stroke will be the focus of this review. Recent advances and our current understanding of age, sex, and hormone interactions in ischemic stroke with a focus on inflammation will be discussed.  相似文献

A high number of neurons express c-fos in response to unlimited food intake in fasted rats in the ventral subdivision of the hypothalamic dorsomedial nucleus (DMHv). We report here, that in same conditions, limited food consumption failed to induce Fos expression in DMHv neurons suggesting that satiation should be one of the important signals that activate these neurons. The possible origin of fibers conducting satiation signals to the DMHv could be in the lower brainstem, especially glucagon-like peptide-1 (GLP-1)-containing neurons in the nucleus of the solitary tract (NTS). We demonstrate that GLP-1-immunoreactive fibers and fiber terminals topographically overlap with activated Fos-positive neurons in the DMHv in refed rats. Using immunocytochemistry and in situ hybridization histochemistry, we demonstrated GLP-1 receptors in Fos-expressing neurons of the DMH. Unilateral transections of ascending GLP-1-containing fibers from the NTS inside the pons in refed rats (unlimited food consumption) resulted in a dramatic decrease in the density of GLP-1 fibers and in the number of Fos-immunoreactive neurons in the DMHv, but only on the side of the transection. Contralateral to the transection, neither the GLP-1 fiber density nor the number of Fos-positive cells changed significantly. Meanwhile, the density of GLP-1 immunoreactivity was markedly accumulated in transected nerve fibers caudal to the cuts, as a consequence of the interruption of the ascending GLP-1 transport route. These findings suggest that the solitary-hypothalamic projections may represent the neuronal route through GLP-1 neurons of the NTS activate DMHv neurons via GLP-1 receptors by conveying information on satiety.  相似文献

Morphology, molecular structure, and thermal properties of potato starch granules with low to high phosphate content were studied as an effect of mild acid hydrolysis (lintnerization) to 80% solubilization at two temperatures (25 and 45°C). Light microscopy showed that the lintners contained apparently intact granules, which disintegrated into fragments upon dehydration. Transmission electron microscopy of rehydrated lintners revealed lacy networks of smaller subunits. The molecular composition of the lintners suggested that they largely consisted of remnants of crystalline lamellae. When lintnerization was performed at 45°C, the lintners contained more of branched dextrins compared to 25°C in both low and intermediate phosphate‐containing samples. High‐phosphate‐containing starch was, however, unaffected by temperature and this was probably due to an altered amylopectin structure rather than the phosphate content. After lintnerization, the melting endotherms were broad with decreased onset and increased peak melting temperatures. The relative crystallinity was lower in lintners prepared at 45°C. A hypothesis that combines the kinetics of lintnerization with the molecular and thermal characteristics of the lintners is presented. © 2013 Wiley Periodicals, Inc. Concentrations of total and inorganic phosphate were measured in forty-seven sediments of the River Garonne (France) using an anion resin, NaOH 0.1 M, Ca-NTA and Na-EDTA. Algal available P was measured using Scenedesmus crassus. The sediments presented a wide range of Tot-P concentrations (226 to 923 μg g−1). Resin P varied between 1.1 to 55.5 μg g−1 and NaOH-P between 3.5 to 262 μg g−1. NTA-P and EDTA-P varied between 13.9 and 178 μg g−1 and between 14.4 to 261 μg g−1 respectively. Algal available P varied between 8.8 to 262 μg g−1/ All forms of P are highly correlated and specially with algal available P (r = 0.70, 0.77, 0.88 and 0.77 for resin-P, NaOH-P, NTA-P and EDTA-P respectively). Nevertheless the partial correlation between EDTA-P and algal available P dropped to 0.05 when the concentration of NTA-P was considered as constant, indicating a spurious correlation with apatitic phosphate (extracted by Na-EDTA). Because of the strongest R 2 and the lowest standard error of estimate, phosphate extracted by Ca-NTA can be considered as the best predictor of algal available phosphate.  相似文献

AimThe presence of Phosphorus as a macronutrient in soil is necessary for plant growth and its deficiency restricts crop yield. Therefore, the aim of current study is to isolate promising rhizospheric phosphate solubilizing bacteria presenting with plant growth promoting (PGP) traits and their utilization as biofertilizers to improve Triticum aestivum (Var. Galaxy 2013) growth and nutrition.MethodOut of 30 isolates obtained from rhizosphere of various plants of different regions, 10 best PSRB strains (WumS-3, WumS-4, WumS-5, WumS-11, WumS-12, WumS-21, WumS-24, WumS-25, WumS-26 and WumS-28) were selected based on their high P solubilization and good PGP (auxin, psiderphore, HCN, Nitrogen fixation) activities. Triticum aestivum (Var. Galaxy 2013) was used as an experimental crop under laboratory and field conditions.ResultsIn this study, P solubilization capacity of selected strains were found 4–7 solubilization index on agar plate and 30–246 µg/ml in liquid broth respectively. The optimum conditions for phosphate solubilization under in vitro condition were found 35 °C at pH 7, glucose as good carbon source and ammonium nitrate as a good nitrogen source. Furthermore, the selected strains had the ability to produces phytohormones (indole acetic acid), siderophore, ammonia and Hydrogen Cyanide. Finally, PSRB inoculum showed significant (p < 0.05) increase (50%–80%) in seed germination while 10–90% increase in root length and shoot length was found as compared to control in laboratory condition. Under natural conditions, 40–80% increase in seed germination while 5–34.8% increase in shoot length and 5–96% increase in seed weight was also observed.ConclusionIsolated strains are promising PSRB that enhance plant growth and this research is a base for recommending the use of these bacterial strains for biofertilizer, as an alternative of chemical fertilizer, for Triticum aestivum L. production.  相似文献
