We report here on the use of RNA interference (RNAi) to create pupal and adult loss-of-function phenotypes in the red flour beetle, Tribolium castaneum, by injection of double-stranded RNA (dsRNA) into late instar larvae (we refer to this method as larval RNAi). RNAi is well-established as a useful method to mimic loss-of-function phenotypes in many organisms including insects. However, with a few exceptions (such as in the fruit fly Drosophila melanogaster), RNAi analysis has usually been limited to studies of embryogenesis. Here we demonstrate that injection of green fluorescent protein (GFP) dsRNA into the larval body cavity can inhibit GFP expression beginning shortly after injection and continuing through pupal and adult stages. RNAi analysis of the Tc-achaete-scute-homolog (Tc-ASH) revealed that larval RNAi can induce morphological defects in adult beetles, and also that larval RNAi affects the entire body rather than being localized near the site of injection. The larval RNAi technique will be useful to analyze gene functions in post-embryonic development, giving us the opportunity to study the molecular basis of adult morphological diversity in various organisms.Edited by D. We used small perturbations in adult numbers to control large fluctuations in the chaotic demographic dynamics of laboratory populations of the flour beetle Tribolium castaneum . A nonlinear mathematical model was used to identify a sensitive region of phase space where the addition of a few adult insects would result in a dampening of the life stage fluctuations. Three experimental treatments were applied: one in which perturbations were made whenever the populations were inside the sensitive region ("in-box treatment"), another where perturbations were made whenever the populations were outside the sensitive region ("out-box treatment"), and an unperturbed control. The in-box treatment caused a stabilization of insect densities at numbers well below the peak values exhibited by the out-box and control populations. This study demonstrates how small perturbations can be used to influence the chaotic dynamics of an ecological system.  相似文献

The Drosophila columnar genes are key regulators of neural precursor formation and patterning along the dorsal-ventral axis of the developing CNS and include ventral nerve cord defective (vnd), intermediate nerve cord defective (ind), muscle segment homeodomain (msh), and Epidermal growth factor receptor (Egfr). To investigate the evolution of neural pattern formation, we identified and determined the expression patterns of Tribolium vnd, ind, and msh, and found that they are expressed in the medial, intermediate, and lateral columns of the developing CNS, respectively, in patterns similar, but not identical, to their Drosophila orthologs. The pattern of Egfr activity suggests that the genetic regulatory mechanisms that initiate Tc-vnd expression are similar in Drosophila and Tribolium, whereas those that initiate Tc-ind have diverged. RNAi analyses of gene function show that Tc-vnd and Tc-ind promote the formation of medial and intermediate column neural precursors and that vnd-mediated repression of ind establishes the boundary between the medial and intermediate columns. These data suggest that columnar gene expression and function underlie neural pattern formation in Drosophila, Tribolium, and potentially all insects, but that subtle spatiotemporal differences in expression of these genes may produce species-specific morphological differences.  相似文献

The relative efficiency of a reduced index (IR) relative to a full index (IF) was compared in an experiment with Tribolium. The selection objective included pupal length, adult weight, and egg mass. The reduced index was based on pupal length and adult weight, whereas the full index was based on these traits and also on egg mass. There were five generations of selection with four replicates, and a selected proportion of 20%. For each replicate, an unselected control was produced. Responses were significant in the IF and IR lines. Responses for the selection objective differed significantly between lines (p < 0.01). The efficiency of the IR line relative to the IF line was 0.52, similar to the expected efficiency of 0.51. The IF and IR lines did not differ significantly for pupal length nor adult weight, whereas the response for egg mass in the IF line was significantly different from the response in the IR line. Realized heritability was greater in the IF line (0.63 ± 0.05) than in the IR line (0.37 ± 0.16). The deleted trait (egg mass) has high heritability and genetic and phenotypic correlations nearly equal to zero with the other two traits included in the selection criterion. The results show the importance of including index traits with high genetic value that are independent of other traits, and they could be useful in breeding programs simultaneously considering production and reproduction traits with nearly zero correlations between them.  相似文献

The effect of larval age range on survival of two Tribolium castaneum strains in mixed cultures was studied in an experimental situation in which other selective forces were reduced to a minimum. Increasing the difference in larval age setween the strains by one day resulted in a reduction of survival of the faster-developing mutant strain, because more of its earlier pupae were cannibalized by the larvae. General aspects of the effects of pupal cannibalism by larvae on the outcome of intra-specific competition (sensu lato) in Tribolium are discussed.  相似文献

Competition for food, space, or other depletable resources has strong impacts on the fitness of organisms and can lead to a pattern known as negative density dependence, where fitness decreases as population density increases. Yet, many resources that have strong impacts on fitness are nondepletable (e.g., moisture or temperature). How do these nondepletable resources interact with depletable resources to modify negative density dependence? We tested the hypothesis that negative density dependence is modulated by temperature in red flour beetles and tested the prediction that the strength of negative density dependence should decrease as temperature decreases. We measured the number of eggs laid, offspring development time, and the number of offspring that reached maturity at three temperatures and two food treatment combinations as we simultaneously manipulated adult population density. We demonstrated that low temperatures weaken negative density dependence in the number of eggs laid; this pattern was most evident when food was abundant. Density had no effect on development time, but low temperatures increased development time. The percent of eggs that emerged as adults decreased with both density and temperature and increased with food. Temperature, an abiotic driver, can thus modulate density-dependent processes in ectotherms. Therefore, models of population growth for ectotherms should incorporate the effects of temperature.  相似文献
