Heat shock proteins (HSP) can protect organisms and cells from thermal damage. In this study, we cloned the full length cDNA encoding the HSP83 protein (the homologue of HSP90) of Tribolium castaneum (red flour beetle). The isolated cDNA contains the full coding sequence, a partial 5′ untranslated region of 55 bp and the complete 3′ untranslated region. We found the hsp83 gene is located on chromosome 5 of the T. castaneum genome. The predicted HSP83 protein sequence has a high similarity (on average 86.77%) with that of other insect species. The expression of the hsp83 gene in the whole body and in the ovary could be induced with heat stress (40°C for 1 h) in newly hatched (within 3 h post emergence) and mature (10 days post emergence) beetles. Under normal conditions, the hsp83 expression in the ovary is about 3-fold higher than in the whole body at both stages. No significant difference in hsp83 expression was observed between the two ovarian developmental stages regardless if the beetles were treated with heat shock or not. The expression of the HSP83 protein in the whole body could also be induced with heat stress in newly hatched and mature beetles. However, in the ovary, HSP83 was only expressed in the follicle cells of mature beetles and not in newly hatched beetles, regardless if the beetles were treated with heat shock or not. Furthermore, the females were not able to produce mature oocytes after knock-down of the hsp83 expression by injecting dsRNA. These results suggest that the HSP83 protein is involved in protection against heat stress and could be involved in oogenesis during ovarian maturation of T. castaneum. 相似文献

采用滤纸药膜选择法研究了蛇床子、茵陈、萹蓄等12种药用植物乙醇提取物对谷蠹Rhizopertha dominica(Fab.)和赤拟谷盗Tribolium castaneum Herbst成虫的驱避效应。12种药用植物以蛇床子(Cnidium monnieri)提取物对谷蠹成虫的驱避效果最佳,786μg/cm2处理24h对谷蠹的驱避率达93%;茵陈(Artemisia capillaries)提取物对赤拟谷盗成虫的驱避效果最佳,用393μg/cm2处理72h的驱避率为95%,茵陈提取物以262μg/cm2放置7d后对赤拟谷盗成虫的驱避率可达Ⅴ级水平。茵陈提取物对赤拟谷盗的驱避持效期长、效果好,具有一定的开发前景。 相似文献

Abstract. When females are inseminated by multiple males, male paternity success (sperm precedence) is determined by the underlying processes of sperm storage and sperm utilization. Although informative for many questions, two-male sperm competition experiments may offer limited insight into natural mating scenarios when females are likely to mate with several males. In this study, genetic markers in Tribolium castaneum are used to trace paternity for multiple sires, and to determine whether displacement of stored sperm that occurs after a third mating equally affects both previous mates, or if fertilizations are disproportionately lost by the female's most recent mate. For 20 days after triple-matings, first males retain significantly higher paternity success (relative to first male paternity in double-matings) compared with second males. These results demonstrate that when females remate before sperm mixing occurs, sperm stratification results in differential loss of sperm from the most recent mate. This study provides insight into the mechanisms underlying sperm precedence in a promiscuous mating system, and suggests that T. castaneum females could limit paternity success of particular mates by remating with more highly preferred males. 相似文献

Abstract We investigated the rates of egg and larval parasitism in transgenic and non-transgenic, conventional cottons. Sentinel eggs and larvae of the cotton bollworm, CBW, Helicoverpa armigera Hübner, were released and collected at regular intervals across the cotton growing season, and the relationship between parasitism and different cotton cultivars determined. Egg and larval parasitism were significantly lower in the transgenic cottons than in the non-transgenic conventional cottons. The egg parasitoid recovered was Trichogramma confusum Viggiani and the predominant larval parasitoids were Campoletis chlorideae Uchida and Meteorus pulchriconis (Wesmael). Our studies indicate a potential negative interaction between transgenic cottons and parasitoids of CBW but need to be interpreted with caution because no within-year replication was used and treatments were not spatially randomised across years. 相似文献

Thymus quinquecostatus Celak. of the Lamiaceae family has a long history of dual use of medicine and food with high economic value, and has been proved to have good antioxidative, antimicrobial, and antidiabetic activities. Essential oil (EO) extracted from the aerial part of T. quinquecostatus was obtained by hydrodistillation and analyzed by gas chromatography-mass spectrometry (GC/MS) and GC. Totally 45 compounds were identified accounting for 98.5 % of the EO. The major constituents of the EO were linalool (52.003 %), borneol (10.911 %) and anethole (5.325 %). Fumigant, contact and repellent activity of T. quinquecostatus essential oil (EO) against Tribolium castaneum (Coleoptera: Tenebrionidae), Lasioderma Serricorne (Coleoptera: Anobiidae), Liposcelis bostrychophila (Psocoptera: Liposcelididae) were evaluated in this work. T. quinquecostatus EO and three major constituents showed varying degrees of insecticidal and repellent activities to three stored-product insects. Anethole stated the best fumigant and contact activity than others did to all three insect species. EO and compounds showed general repellent activity against three target insects. This work suggests that the EO of T. quinquecostatus has promising potential to be developed into botanical pesticides and repellents controlling pest damage in warehouses and grain stores. 相似文献
