Physiological properties of bacteriophage T5 gene A1 mutants, whose growth is inhibited in λ lysogens, and designated T5 lr, have been studied. In the presence of λ gene rex, which is responsible for lr growth inhibition, gene A1 product is synthesized and functional. However, several physiological defects were observed: phage DNA synthesis is inhibited; late phage-induced proteins are synthesized in markedly decreased amounts after a delay of about 15 minutes; phage DNA transfer into the host goes beyond the first-step transfer fragment but, in most bacteria, is interrupted after penetration of about 55% of the genome. Relationships between these different defects are discussed.  相似文献

The successful eradication of introduced rodents from islets off the coast of Mauritius has led to local conservation bodies investigating the possibility of translocation as a measure of safeguarding endemic reptile populations. The present study was the first to determine the habitat and microhabitat requirements of Telfair's skinks (Leiolopisma telfairii) on Round Island, Mauritius, with a view to aiding future translocation projects to islands within their historic range. Contrasting preferences found for Telfair's skink at macro- and micro- habitat levels underline the importance of sampling at multiple ecological scales in such investigations. Significantly fewer sightings of L. telfairii were recorded in bare rock habitats compared to more vegetated habitats. Conversely, at a microhabitat scale principal component analysis indicated structural characteristics were the primary determinant of microhabitat choice. The first dietary analysis of Telfair's skinks confirmed their status as omnivores. Cockroaches (Blattodea spp.) appeared to be a primary food source. Four exotic plant species were also present in faecal samples and the potential for L. telfairii to aid their dispersal is discussed. Implications for the long-term management and proposed translocation of Telfair's skinks are discussed.  相似文献

In this study, we conducted genomic prediction for two Yorkshire purebred populations (Yichun and Chifeng) from two different provinces of China that both had a limited population size. Two growth traits (age adjusted to 100 kg weight, AGE; back‐fat thickness adjusted to 100 kg weight, BF) and one reproduction trait (total number of piglets born, TNB) were analyzed with four prediction strategies: one‐population BLUP, joint two‐population BLUP, one‐population single‐step BLUP (SSBLUP) and joint two‐population SSBLUP. Our results illustrate that accuracies of genomic estimated breeding values were improved for BF and TNB for the Yichun population and for BF for the Chifeng population by genomic prediction (one‐population SSBLUP and joint two‐population SSBLUP). The accuracy of TNB for the Yichun population was increased two fold when comparing the one‐population SSBLUP to the one‐population BLUP prediction. Meanwhile, prediction biases were dramatically reduced for AGE for the Yichun population and for TNB for the Chifeng population. The conclusions of this study are as follows: first, genomic prediction is useful for improving prediction accuracy for purebred pig breeding farms with a limited population size; second, joint genomic prediction for different populations of the same breed with certain genetic links has the trend to further improve prediction accuracy.  相似文献

Habitat selection models are used in ecology to link the spatial distribution of animals to environmental covariates and identify preferred habitats. The most widely used models of this type, resource selection functions, aim to capture the steady-state distribution of space use of the animal, but they assume independence between the observed locations of an animal. This is unrealistic when location data display temporal autocorrelation. The alternative approach of step selection functions embed habitat selection in a model of animal movement, to account for the autocorrelation. However, inferences from step selection functions depend on the underlying movement model, and they do not readily predict steady-state space use. We suggest an analogy between parameter updates and target distributions in Markov chain Monte Carlo (MCMC) algorithms, and step selection and steady-state distributions in movement ecology, leading to a step selection model with an explicit steady-state distribution. In this framework, we explain how maximum likelihood estimation can be used for simultaneous inference about movement and habitat selection. We describe the local Gibbs sampler, a novel rejection-free MCMC scheme, use it as the basis of a flexible class of animal movement models, and derive its likelihood function for several important special cases. In a simulation study, we verify that maximum likelihood estimation can recover all model parameters. We illustrate the application of the method with data from a zebra.  相似文献

Genetic founder effects of a historical translocation (1965; app. 12 generations ago) of endangered softmouth trout from the only remaining indigenous population to an adjacent uninhabited river were investigated. A comparison of 15 microsatellites from contemporary samples in both the source and re-established populations indicated a very low level of genetic diversity within the two populations. Furthermore, considerable differences in estimates of variability and effective population sizes were detected between populations, but no recent bottlenecks were evident. Our data suggest that the re-established population has lost variation due to genetic drift associated with founder effects following the translocation. Accordingly, in relation to management, we suggest that the re-established population could serve as a secondary source of individuals to buffer possible extinction due to demographic events. Finally, conservation initiatives to enhance the census population size and increase levels of variability in the re-established population are proposed. The concentrations of metallothionein (MT) in bivalves, a potential biomarker of metal pollution, are variable according to specific organs, the highest concentrations being encountered in the digestive glands of oysters. Thus, the present study has been focussed on this organ with a view to validate the use of MT as a biomarker in the field, the temporal changes of metal and metallothionein concentrations have been examined from March to October 1997 in the digestive gland of resident oysters from a clean site (Bay of Bourgneuf, France) and a metal-rich site, the Gironde estuary which has been shown as the most Cd-contaminated marine area in France but is also enriched with Cu and Zn. Moreover, oysters from the clean site have been translocated to the Gironde estuary over the same period. Taking into account all the samples collected over the 7 months of the study, MT concentrations in the digestive gland were positively correlated with weight whereas metal levels were negatively correlated with weight. However, considering monthly samples including specimens from both sites (resident or translocated oysters), a positive correlation was shown between MT and metal concentrations in autumn (September and October) but not in spring and summer. These findings limit the interest of using the digestive gland of oysters as the preferred tissue for the determination of MT concentration as a biomarker.The alternative use of gills should be considered.  相似文献

Translocation, or the purposeful movement of organisms from one location to another for conservation, is currently being used to bolster populations of the endangered greater prairie-chicken (Tympanuchus cupido). We used radiotelemetry to compare survival between 58 resident birds and 54 newly translocated greater prairie-chickens that were sourced from a location more than 325 km away. Model averaged survival estimates were lower in translocated birds (0.42; 95% CI: 0.17–0.66) than in resident prairie-chickens (0.65; 95% CI: 0.46–0.79) through the breeding season. Habitat, sex and year were each included in at least 1 of the top 4 models, but the model averaged confidence intervals for each parameter encompassed zero. Survival of both resident and translocated prairie-chickens increased throughout the breeding season. Both translocated and resident prairie-chickens selected for core prairie habitat over agriculture, and birds tended to avoid surrounding private grasslands and wooded areas. We suggest that future translocation projects account for reduced survival of translocated birds when determining the appropriate release cohort sizes and sex ratios. We also recommend that future management for greater prairie-chicken habitat focus on the expansion of core protected patches of prairie to promote elevated survival and better chances of conservation success.  相似文献

Summary The costs and benefits of resource integration in Hydrocotyle bonariensis were examined by comparing severed and intact clones grown across multiple resource gradients. Basipetal movement of water, nitrogen and photosynthates was demonstrated to occur between two rhizome branch systems interconnecting hundreds of ramets within a clone. Hydrocotyle clones of this size and larger have been shown to span highly patchy and unpredictable resource conditions in coastal dune environments. The extensive movement of water and nitrogen to portions of a clone deficient in these resources, resulted in a significant net benefit to the clone in terms of fitness-related traits: total biomass, ramet proliferation and seed production. The translocation of photosynthates across light gradients allowed for sexual reproduction and clonal expansion in the shade although this incurred a net cost to the clone in terms of ramet and seed production.  相似文献
